研究用于癌症诊断与治疗的光敏剂血卟啉(hematoporphyrin derivative,HPD)的超快光动力学过程,采用超短脉冲激光光谱技术和皮秒时间相关单光子计数系统,测量经血卟啉培养的活体癌细胞与正常细胞的荧光光谱、荧光寿命特性及荧光峰值强度随时间变化曲线,并测量单一细胞内部不同位置的荧光寿命特性,观测到:癌细胞样品在645 nm处具有特有的光谱谱峰;癌细胞样品荧光寿命的快成分约150 ps慢成分约1200 ps,而正常细胞样品快成分约300 ps慢成分约2500 ps;癌细胞样品的荧光峰值强度经12小时衰减约10%,而正常细胞样品衰减约55%;在细胞内部荧光寿命300 ps的快成分十分显著,且中心部位血卟啉浓度最高.癌细胞与正常细胞的荧光光谱、荧光寿命特性及荧光峰值强度随时间变化曲线相差十分明显,反映了癌细胞与正常细胞对血卟啉亲和特性有显著的差异,测量结果确认了荧光光谱技术诊断与治疗癌症的可行性,并对发展超短脉冲激光光谱技术早期诊断与治疗癌症具有重要的指导意义和临床应用价值. 相似文献

Ethanol at 50% (v/v) and sodium bicarbonate at 1% (wt/v), either alone or in combination, were applied to organically grown strawberries 1 h before harvest to control the natural incidence of postharvest diseases. Botrytis cinerea was the major cause of decay in all of the experiments. In three experiments, ethanol significantly reduced the decay incidence after storage for 3 days at 1°C followed by 2 days at 24°C, while the efficacy of sodium bicarbonate was inconsistent. The combination of ethanol and sodium bicarbonate did not increase their efficacy. Postharvest hot water dips at 55 and 60°C for 30 s significantly reduced the decay incidence to 3.4 and 2.7%, respectively, while decay incidence in the control was 28.5% (the first experiment). The efficacy of the hot water treatments at 55 and 60°C for 30 s was consistent in three experiments. In the third experiment, the efficacy of hot water treatment at 60°C was significantly higher than that of hot water treatment at 55°C. All pre‐ and postharvest treatments significantly reduced natural fungal populations on the surfaces of fruits. None of the pre‐ and postharvest treatments caused surface injuries to the fruit or adversely affected weight loss and taste parameters. 相似文献

Nonsense-mediated mRNA decay (NMD) is the best-characterized mRNA surveillance mechanism; this process removes faulty mRNAs harboring premature termination codons (PTCs). NMD targets newly synthesized mRNAs bound by nuclear cap-binding proteins 80/20 (CBP80/20) and exon junction complex (EJC), the former of which is thought to recruit the ribosome to initiate the pioneer round of translation. After completion of the pioneer round of translation, CBP80/20 is replaced by the cytoplasmic cap-binding protein eIF4E, which mediates steady-state translation in the cytoplasm. Here, we show that overexpression of eIF4E-T preferentially inhibits cap-dependent steady-state translation, but not the pioneer round of translation. We also demonstrate that overexpression of eIF4E-T or Dcp1a triggers the movement of eIF4E into the processing bodies. These results suggest that the pioneer round of translation differs from steady-state translation in terms of ribosome recruitment. 相似文献

We identified human decapping enzyme 2 (hDCP2) as a binding protein with Ro52, being colocalized in processing bodies (p-bodies). We also showed that the N-terminus and C-terminus of Ro52 bound to hDCP2. Moreover, Ro52 enhanced decapping activity of hDCP2 in a dose-dependent manner. Our data support the novel notion of the association between Ro52 with hDCP2 protein in cytoplasmic p-bodies, playing a role in mRNA metabolism in response to cellular stimulation. 相似文献

Recent data challenged the assumption that light has little effect on retina development. Here, we report evidence that dark rearing permanently changes the synaptic input to GCs. A reduced spontaneous postsynaptic currents (SPSCs) frequency was found in retinal GCs from rats born and raised in the dark for three months. Glutamate antagonists (CNQX and AP-5) reversibly reduced SPSCs frequency in control and dark-reared (DR) retinae. The GABA antagonist picrotoxin (PTX) reduced SPSCs frequency in control retinas, but increased SPSCs frequency in DR, mainly by presynaptic action on excitatory currents. In DR animals exposed to normal cyclic light for 3 months, SPSCs frequency remained lower then in control rats and increased following PTX, suggesting that long-term dark rearing induces permanent modifications of the retinal circuitry. Our results strongly support the idea that light stimulation plays a role in establishing normal synaptic input to GCs. 相似文献

The solubility of plutonium was estimated for waste buried at the Greater Confinement Disposal site in Nevada. The EQ3/6 thermochemical database was modified to include recent data on Pu complex formation, and the solubilities of two critical phases (probertite (CaNaB5O9·5H2O), added as a backfill material; and Ca sac-charate) were determined by experiment. Reaction path runs were used to model effects of cellulose degradation, including complexation of actinides by organic acids and carbonate, decay of the complexing agents, and the buildup and diffusive loss of CO2 through the permeable alluvium. For most waste interaction scenarios, long-term (≈103 years) concentrations of Pu in pore waters are ≤10?7 molal and are dominated by carbonate complexes, although organic complexes could dominate in the first ≈103 years. In unusual circumstances, carbonation of buried lithium could produce very high Pu solubilities; however, even in such a system, a slight lowering of the effective redox potential dramatically lowers Pu solubility. A sensitivity analysis suggests that the “base case” calculations are conservative, tending to overestimate long-term solubility, with the system redox and the identity of the organic acids the major sources of uncertainty. 相似文献
