The fluorescence decays of pyrene in small and large unilamellar L,-dipalmitoylphosphatidylcholine vesicles have been investigated as a function of probe concentration and temperature. When the molar ratio of pyrene to phospholipid equals 1:3000, no excimer emission is observed and the fluorescence decays are mono-exponential. When this ratio is equal to or higher than 1:120, excimer formation is observed.Above the phase transition temperature the observed fluorescence decays of monomer and excimer can be adequately described by a bi-exponential function. The monomer decays can be equally well fitted to a decay law which takes into account a time-dependence in the probe diffusion rate constant. The fluorescence decay kinetics are compatible with the excimer formation scheme which is valid in an isotropic medium. The excimer lifetime and the (apparent) rate constant of excimer formation have been determined as a function of probe concentration at different temperatures above the phase transition temperature. The activation energy of excimer formation is found to be 29.4±1.3 kJ/mol. In small unilamellar vesicles the diffusion constant associated with the pyrene excimer formation process varies from 8.0x10-7 cm2/s at 40°C to 2.2x10-6 cm2/s at 70°C.Below the phase transition temperature the monomer decays can be described by a decay law which takes into account a time dependence of the rate constant of excimer formation. The lateral diffusion coefficient of pyrene calculated from the decay fitting parameters of the monomer region varies from 4.0x10-9 cm2/s at 20°C to 7.9x10-8 cm2/s at 35°C. Minirhizotron observations of roots of leeks and Brussels sprouts grown in the Wageningen Rhizolab were used to study the dynamics of root length. Day of appearance and the time of decay were assessed for individual root segments visible on the minirhizotron surface.A Brussels sprouts crop produced much more root length than leeks, but the average longevity of these roots was about half that of leek roots.To investigate whether root colour or UV fluorescence could be used as a quantitative index of root functionality or root age, changes in root colour (on a scale of greys) over time were measured with interactive image analysis. In both crops a gradual change towards black was found with ageing. Measurements of the intensity of the UV fluorescence showed that leek roots fluoresced more than Brussels sprouts roots. Over time, UV fluorescence decreased in Brussels sprouts roots but increased in leek roots. It is concluded that UV fluorescence cannot be used as a universal indicator of root age or root functionality, but in some plant species it may be used to separate (transparent) roots from the background with image analysis techniques.  相似文献

Sanosil-25, a disinfectant containing 48% hydrogen peroxide and silver salts as stabilising agents, inhibited the mycelial growth of the two main decays causing fungi of melons, Alternaria alternata and Fusarium solani in vitro at concentrations between 5000 and 10 000 μl litre-1. However, in in vivo experiments, Sanosil-25 markedly decreased decay at a concentration of 5000 μl litre-1 when incorporated into a wax treatment and produced no phytotoxic effect. This treatment may provide an alternative to imazalil which, although more effective, gives problems with residue levels. A concentration of 10 000 μl litre-1 proved phytotoxic.  相似文献

The bacterial secretion of glutamate was studied through plasma membrane fluidity, measured by anisotropy using the fluorophore TMA-DPH incorporated in the lipid part of the cell membrane. Cells of Brevibacterium sp. ATCC 13869 (wild type) were switched from the biotin-limited, producing state to the biotin-supplemented, non-producing state, and back. The following conclusions could be drawn: 1. It was not possible to detect any change in anisotropy by switching the cells from biotin-limited biotin-supplemented, as well as from biotin-supplemented, to biotin-limited, media. 2. The anisotropy value in the glutamic acid fermentation remains constant during the lag, exponential, growth, production and stationary phases. 3. The treatment of cells with a neutral synthetic polyester of ethylene-and propyleneoxide with soya oil-fatty acids increased the anisotropy values, indicating incorporation of the surfactant. 4. Glutamate secretion is not coupled with membrane fluidity, so a leak providing a general fluidization of the membrane could not be detected.  相似文献

Aim

One of the oldest and most powerful ways for ecologists to explain distinct biological communities is to invoke underlying environmental differences. But in hyper-diverse systems, which often display high species richness and low species abundance, these sorts of community comparisons are especially challenging. The classic view for Amazonian birds posits that riverine barriers and habitat specialization determine local and regional community composition. We test the tacit, complementary assumption that similar bird communities should therefore permeate uniform habitat between major rivers, regardless of distance.

Location

Upland (terra firme) rainforests of central Amazonia.

Methods

We conducted intensive whole-community surveys of birds in three pairs of 100-ha plots, separated by 40–60 km. We then used dissimilarity indices, cluster analysis, and ordination to characterize differences among the six avian communities.

Results

In all, we detected 244 forest-dependent birds, with an average of 190 species (78%) per plot. Species turnover was negligible, no unique indicator species were found among plot pairs, and all documented species were already known from a complete inventory at one of the three sites.

Main Conclusions

Our study corroborates the classic biogeographical pattern and suggests that turnover contributes little to regional avian diversity within upland forests. Using a grain size of 100 ha, this implies that upland birds perceive the environment as uniform, at least over distances of ~60 km. Therefore, to maximize both local species richness and population persistence, our findings support the conservation of very large tracts of upland rainforest. Our analyses also revealed that the avifauna at Reserva Ducke, encroached by urban sprawl from the city of Manaus, shows the hallmarks of a disturbed community, with fewer vulnerable insectivores. This defaunation signals that even an enormous preserve (10 × 10 km) in lowland Amazonia is not insulated from anthropogenic degradation within the surrounding landscape.  相似文献

Mnemiopsin 1 (Mn1) and Mnemiopsin 2 (Mn2) are photoproteins found in Mnemiopsis leidyi. We have tried to answer the question of whether the structural features of photoproteins can explain the observed activity data. According to the activity measurements data, they have the same characteristic wavelength. However, the initial intensity of Mn2 is significantly higher than that of Mn1, and decay time of Mn1 (0.92 s−1) is lower than that of Mn2 (1.46 s−1). The phylogenetic analysis demonstrates that, compared with Obelin and Aequorin from Obelia longissima and Aequorea victoria, respectively, a gene modification event may have caused the expansion of the N-terminal side of all photoproteins from M. leidyi. An in silico study has shown that the stability of the photoprotein–substrate complex of Mn2 is higher than that of Mn1, indicating a higher affinity of the substrate for Mn2 compared with Mn1. It was revealed that the active EF-hand loops 1 and III in Mn2 is locally more rigid compared with those in Mn1. We concluded that different stability of the photoprotein complexes leads to different initial intensity. While different patterns of the local dynamics of loops I and III may influence the decay rate.  相似文献
