Most peptide and protein drugs are short-lived species in vivo with a circulatory half-life of several minutes. This is particularly valid for non-glycosylated proteins with a molecular
mass of less than 50 kDa. Since peptide/protein drugs are not absorbed orally, prolonged maintenance of therapeutically active
drugs in the circulatory system is of primary clinical importance. Another major obstacle of injected polypeptide drugs is
the elevated concentration of 100–1000 times above the therapeutical level that may be present in the circulatory system shortly
after administration. Such overdosing may lead to undesirable side effects such as over-stimulation or down-regulation of
receptor sites.
In this review we describe two new strategies that overcome these two problems of systemically injected peptide/protein drugs.
The first strategy includes Fmoc and FMS derivatization of peptides, proteins and low molecular-weight drugs, converting them
to inactive prodrugs that undergo reactivation with desirable pharmacokinetic patterns in body fluids. Based on this Fmoc/FMS-technology,
we have developed a second strategy, reversible pegylation. Inactive pegylated peptide/protein drugs release the native active
parental molecule at slow rates, and in homogeneous fashion under physiological conditions, thus facilitating prolonged therapeutic

骨关节炎(osteoarthritis,OA)是运动系统常见的退行性疾病,具有高发病率和高致残率。骨关节炎的发病机制目前尚不明确,既往研究认为骨关节炎发病主要与创伤因素相关,而近期研究表明,以胆固醇代谢异常为主的代谢性因素同样与骨关节炎密切相关,骨关节炎的治疗以早期对症治疗和晚期手术治疗为主,尚无针对病因的特效药物。既往研究中发现,有一种具有软骨保护作用的小分子药物BNTA,其在创伤引起的骨关节炎中具有较好的疗效,但其对高胆固醇引起的骨关节炎的作用尚不明确。本研究为探究BNTA对高胆固醇引起的骨关节炎的治疗作用及其机制,采用高胆固醇饮食构建了大鼠骨关节炎模型,取膝关节石蜡切片进行组织学评估,使用油红O染色评估大鼠软骨细胞内的脂质积聚情况,使用RT-qPCR、免疫荧光和免疫组化评估软骨细胞合成代谢、分解代谢及胆固醇代谢相关基因和蛋白质的表达。结果显示,BNTA可缓解高胆固醇大鼠骨关节炎模型中的病理表现,改善OARSI评分。在大鼠软骨细胞中,BNTA可促进合成代谢相关基因col2、sox9、acan的表达,抑制分解代谢相关基因mmp13、adamts5的表达,可改善高胆固醇引起的大鼠软骨细胞脂质积聚。在大鼠软骨细胞和高胆固醇大鼠骨关节炎模型中BNTA均可上调Insig1表达。本研究证实,高胆固醇可在体内和体外实验中加重骨关节炎,可引起大鼠软骨细胞脂质积聚增加。在体内和体外实验中BNTA均能缓解高胆固醇引起的骨关节炎表型,改善软骨细胞内的异常脂质积聚,其作用可能为通过上调Insig1抑制细胞内的胆固醇生物合成,从而缓解脂质异常积聚。 相似文献

Antiepileptic drugs (AED) have been associated to in vivo deleterious consequences in bone tissue. The present work aimed to characterize the cellular and molecular effects of five different AED on human osteoclastogenesis and osteblastogenesis. It was observed that the different drugs had the ability to differentially modulate both processes, in a way dependent on the identity and dose of the AED. Shortly, valproic acid stimulated either osteoclastogenesis and osteoblastogenesis, whereas carbamazepine, gabapentin, and lamotrigine revealed an opposite behavior; topiramate elicited a decrease of osteoclast development and an increase in osteoblast differentiation. This is the first report describing the direct effects of different AED on human primary bone cells, which is a very important issue, because these drugs are usually consumed in long-term therapeutics, with acknowledged in vivo effects in bone tissue. 相似文献

The prostaglandins (PG) a group of physiologically active lipid compounds having diverse hormone like effects are important mediators of the body’s response to pain and inflammation, and are formed from essential fatty acids found in cell membranes. This reaction is catalyzed by cyclooxygenase, a membrane associated enzyme occurring in two isoforms, COX-1 and COX-2. Nonsteroidal anti-inflammatory drugs (NSAIDs) act by inhibiting the activity of COX. In view of this, a series of novel benzophenones conjugated with oxadiazole sulphur bridge pyrazole moiety 8a-l were designed, synthesized, characterized and subsequently evaluated for anti-inflammatory and analgesic property. The investigation of novel analogues 8a-l for potential anti-inflammatory activity showed high levels of COX-1 and COX-2 inhibitory activity. Among the series, compound 8i with electron withdrawing fluoro group at the para position of the benzoyl ring of benzophenone was characterized by highest IC50 values for both COX-1 and COX-2 inhibition, which is comparable to the standard drug. Protein docking methods are powerful computational tools to study protein-protein interactions (PPI). While a significant number of docking algorithms have been developed, they are usually based on rigid protein models or with limited considerations of protein flexibility and the desolvation effect is rarely considered in docking energy functions, which may lower the accuracy of the predictions. To address these issues, we introduce a PPI energy function based on the site-identification by ligand competitive saturation (SILCS) framework and utilize the fast Fourier transform (FFT) correlation approach. The free energy content of the SILCS FragMaps represent an alternative to traditional energy grids and they can be efficiently utilized to guide FFT-based protein docking. Application of the approach to eight diverse test cases, including seven from Protein Docking Benchmark 5.0, showed the PPI prediction using SILCS approach (SILCS-PPI) to be competitive with several commonly used protein docking methods indicating that the method has the ability to both qualitatively and quantitatively inform the prediction of PPI. Results show the utility of the SILCS-PPI docking approach for determination of probability distributions of PPI interactions over the surface of both partner proteins, allowing for identification of alternate binding poses. Such binding poses are confirmed by experimental crystal contacts in our test cases. While more computationally demanding than available PPI docking technologies, we anticipate that the SILCS-PPI docking approach will offer an alternative methodology for improved evaluation of PPIs that could be used in a variety of fields from systems biology to excipient design for biologics-based drugs. 相似文献

Fenfluramine is an anorectic drug widely used for the regulation of food intake that presents some adverse effects at the central and peripheral levels. d-Fenfluramine, an isomer of dl-fenfluramine, is postulated to be more effective and to induce less side effects than the racemic compound. These drugs act preferentially on the serotonergic system. Some authors have suggested that fenfluramine causes a degeneration of serotonergic neurons. Alterations of the serotonergic system are also observed during the aging process, and in this case, a relationship with reactive oxygen species has been already established. In view of these data, the present study was conducted to investigate the relationship between fenfluramine and brain antioxidant defense system in mature and aged animals. Rats aged 4 and 17 months were chronically treated with dl-fenfluramine, d-fenfluramine, or saline. Brain activity of superoxide dismutase and glutathione peroxidase was significantly affected by aging. Catalase activity was altered by the treatment. Total glutathione content and chemiluminescence in the brains were also altered by aging. Glutathione levels were altered as a function of the interaction between age and treatment. These findings suggest that treatment with d- or dl-fenfluramine results in alteration of the antioxidant system that could be exacerbated when associated with the aging process. © 1997 John Wiley & Sons, Inc. 相似文献

NSAIDs are potent chemopreventive agents for colon cancer. Although their mechanism of action is unknown, it probably relates to their modulation of colon epithelial cell kinetics, i.e. apoptosis and/or cell proliferation. NSAIDs are pleiotropic in their biochemical activities; their best known effect is inhibition of prostaglandin H synthase (PHS), the enzyme catalyzing the biosynthesis of prostaglandins. Current data appear to lead to two conflicting conclusions. One body of data indicates that PHS is important in induction of apoptosis and colon carcinogenesis and that its inhibition by NSAIDs is required for induction of apoptosis and their overall chemopreventive effect. Another set of data indicates that NSAIDs may induce apoptosis and prevent colon cancer without inhibiting the activity of PHS. Both sides of this argument are presented and discussed. This apparent contradiction may be resolved if one accepts that both mechanisms are correct but that they act on different steps of this multistep process. 相似文献

Summary. Endogenous histamine has been reported to be involved in regulation of seizure susceptibility. Enhancement of histamine neurotransmission engendered by L-histidine treatment produces anticonvulsant effects in experimental animals. The present study investigated the influence of L-histidine on the protective effects of carbamazepine and phenytoin against maximal electroshock-induced seizures in mice.L-Histidine, administered at the doses that did not influence the threshold for electroconvulsions (250–500mg/kg), enhanced by nearly 30% the protective effects of carbamazepine against maximal electroshock-induced seizures. D-Histidine (1000mg/kg), an inactive isomer of histidine, was without any effect in this regard. L-Histidine (500mg/kg) also augmented the protective effects of phenytoin. Importantly, the enhancement of the anticonvulsant effects of these antiepileptic drugs produced by L-histidine co-administration was not associated with augmentation of their unwanted effects on memory and motor performance. A pharmacokinetic interaction was also excluded since the free plasma levels of these antiepileptics remained unchanged in the presence of L-histidine. Numerous studies have shown that endogenous and/or environmental neurotoxins and oxidative stress may participate in the pathogenesis of Parkinson's disease (PD), but the detailed mechanisms are still unclear. While dopamine (DA) replacement therapy with L-DOPA (levodopa) improves PD symptoms, it does not inhibit the degeneration of DA neurons in the substantia nigra. Recently, bromocriptine, pramipexole and several other agonists of the dopamine D2-receptor subfamily (including D2, D3 and D4-subtypes) have been shown to have neuroprotective effects in parkinsonian models in vitro and in vivo. Their neuroprotective effects may be mediated directly and/or indirectly by antioxidant effects, mitochondrial stabilization or induction of the antiapoptotic Bcl-2 family. 相似文献
