Summary To develop a successful protocol forAgrobacterium-mediated transformation in plants it is essential to determine the most efficient bacterial strain/plant genotype interaction. In the present work, we evaluated the susceptibility ofEucalyptus grandis ×E. urophylla to fiveAgrobacterium rhizogenes and twelveA. tumefaciens wildtype strains. The results showed different degrees of virulence, according to the strain tested, indicating that transformation of this eucalypt hybrid by Agrobacterium-derived vectors is possible. All developed tumours showed an autonomous growth when transferred to a hormone-free medium. Some of these tumours formed shoots spontaneously, with a normal phenotype. Polymerase Chain Reaction (PCR) and Southern blot analyses were performed to confirm the absence of the oncogenic T-DNA in plants derived from these shooty tumours.  相似文献

Abstract: The semi-longitudinal collection of growth measurements in male and female hamadryas baboons has enabled documentation of the timing of puberty and the development of sexually dimorphic growth patterns in body weight, crown-rump length (CRL), limb lengths, and muscle mass. In addition, another sexually dimorphic characteristic appears to be the presence of a pubertal growth spurt in body weight, and possibly CRL, in male but not female baboons. Serum testosterone levels rose during male development; however, there was a progressive decrease in dehydroepiandrosterone sulfate levels indicating the absence of adrenarche. Insulin-like growth factor-I (IGF-I) and its major binding protein, IGFBP-3, both rose during pubertal development; however, a simultaneous rise in the IGF-I:IGFBP-3 molar ratio suggests other factors may enhance the bioactivity of IGF-I during puberty. A distinct rise in serum osteocalcin levels was also associated with puberty in male baboons. These growth and hormonal changes during puberty in the hamadryas baboon indicate that this species provides a close primate model for human puberty.  相似文献

A study was carried out on the linkage relationship between the Frl locus carrying resistance to Fusarium oxysporum f.sp. radicis-lycopersici and the Tm-2 locus carrying resistance to several races of tobacco mosaic virus in the tomato inbred line IRB-301-31. The inbred line Motelle (Frl+/Frl+, Tm-2+/Tm-2+) was crossed with the inbred line IRB-301-31 (Frl/Frl, Tm-2/Tm-l). The resulting 222 F2 plants were selfed, and from each F3 family groups of 15–60 seedlings were tested for resistance to either F. oxysporum f.sp. radicis-lycopersici or tobacco mosaic virus race 0. Segregation data indicated a very tight linkage between Frl and Tm-2, equal to 5.1 ± 1.07 map units.  相似文献

Individual leaves of perennial ryegrass cv. Aberystwyth S23 of two leaf ages and at two levels of nitrogen fertilisation were point inoculated with Puccinia coronata f.sp. coronata in a growth chamber. In general, there was no significant difference in the lifespan of inoculated versus control leaves. However, the higher rate of nitrogen extended leaf lifespan more markedly in rusted than in control leaves. Uredospore production varied according to leaf age: colonies on juvenile leaves produced three times as many spores as those on mature leaves.  相似文献

The enzyme tryptophan decarboxylase (TDC) (EC 4.1.1.28) catalyses a key step in the biosynthesis of terpenoid indole alkaloids inC. roseus by converting tryptophan into tryptamine. Hardly anytdc mRNA could be detected in hormone-independent callus and cell suspension cultures transformed by the oncogenic T-DNA ofAgrobacterium tumefaciens. Supply of tryptamine may therefore represent a limiting factor in the biosynthesis of alkaloids by such cultures. To investigate this possibility, chimaeric gene constructs, in which atdc cDNA is linked in the sense or antisense orientation to the cauliflower mosaic virus 35S promoter and terminator, were introduced inC. roseus cells by infecting seedlings with an oncogenicA. tumefaciens strain. In the resulting crown gall tumour calluses harbouring thetdc sense construct, an increased TDC protein level, TDC activity and tryptamine content but no significant increase in terpenoid indole alkaloid production were observed compared to empty-vector-transformed tumour calluses. In tumour calluses containing thetdc antisense construct, decreased levels of TDC activity were measured. Factors which might be responsible for the lack in increased terpenoid indole alkaloid production in thetdc cDNA overexpressing crown gall calluses are discussed.  相似文献

Novel enantiopure pyridino‐18‐crown‐6 ether‐based sensor molecules containing an anthracene fluorophore unit were synthesized. Their enantiomeric recognition abilities toward the enantiomers of 1‐phenylethylamine hydrogen perchlorate (PhEt), 1‐(1‐naphthyl)ethylamine hydrogen perchlorate (NapEt), phenylglycine methyl ester hydrogen perchlorate (PhgOMe), and phenylalanine methyl ester hydrogen perchlorate (PheOMe) were examined in acetonitrile using fluorescence spectroscopy. The sensor molecules showed appreciable enantiomeric recognition toward the enantiomers of NapEt, PhEt, and PhgOMe. The highest enantioselectivity was found in the case of crown ether containing isobutyl groups in the macroring and the enantiomers of NapEt. 目的:研究正畸带环与树脂临时冠对牙隐裂暂时性保护的临床疗效,为临床治疗提供依据。方法:选取2013年5月到2015年4月我院收治的牙隐裂患者196例,按照随机数字表法将患者分为A组和B组,每组98例,A组给予正畸带环治疗,B组给予树脂临时冠治疗,比较两组固定效果、咬合情况、牙周情况以及满意度。结果:两组固定成功率比较无统计学意义(P0.05);A组牙龈炎发生率为12.2%显著高于B组的3.1%,差异具有统计学意义(P0.05);B组满意度为95.9%显著显著高于A组的80.6%,差异具有统计学意义(P0.05)。结论:树脂临时冠较正畸带环对牙隐裂暂时性保护作用较好,患者满意率较高,值得临床推广应用。  相似文献

目的:探讨金属桩与玻璃纤维桩对老年上颌前牙残根残冠修复的临床疗效。方法:收集我院口腔科收治的上颌前牙残根残冠修复患者40例,随机分为玻璃纤维桩组和金属桩组,每组各20例,玻璃纤维桩组患者给予玻璃纤维桩进行残根残冠修复,金属桩组给予金属桩进行残根残冠修复,治疗结束后1年随访,对所有患者的牙龈指数(GI)、修复齿出血指数(SBI)、不良反应发生率、患者修复效果以及患者疗效进行检测并比较。结果:与治疗前相比,两组患者的牙龈指数(GI)以及修复齿出血指数(SBI)均下降(P0.05);与金属桩组相比,玻璃纤维桩组患者的牙龈指数(GI)以及修复齿出血指数(SBI)较低(P0.05),不良反应发生率较低(P0.05),颜色匹配率以及边缘适合率较高(P0.05),两组患者修复体完整例数百分率比较无明显差异(P0.05),玻璃纤维桩组患者的治疗成功率较高(P0.05)。结论:玻璃纤维桩在老年上颌前牙残根残冠修复的应用与金属桩组相比临床效果较好。  相似文献

In a cloud forest dominated by Quercus costaricensis, the gap size between the crowns was consistent and crowns were asymmetrical, being elongated perpendicular to the prevailing wind direction. A wind storm of 20 m/s removed 1.5% of the leaves, consistent with the idea that wind abrasion shapes tree crowns.  相似文献

The distribution and density of Pratylenchus minyus and possible relationships of several environmental components, including ammonium nitrate, were investigated in a wheat field in South Australia. Seasonal variation as measured every 2-4 weeks was eliminated from the observations by periodic regression. Correlation and regression analyses were then used to investigate the association of host plant, rainfall, temperature, and the fungus Gaeumannomyces graminis with P. minyus. Other than seasonal effects, soil moisture and G. graminis were the only components associated with P. minyus. Ammonium nitrate usually was correlated with fewer P. minyus in wheat roots. Much higher numbers of P. minyus were observed in seminal than in crown roots of wheat.  相似文献
