Exposure of blue-green or red algal cells to temperatures exceeding 60–65°C for several minutes resulted in bleaching of all phycobilin absorption in the visible range, with virtually no alteration in chlorophyll or carotenoid absorption. Difference spectra of non-bleached vs bleached cells appeared identical to absorption spectra of purified phycobilisomes isolated from the same cell culture in high phosphate medium. All phycobilin chromophores were bleached at approximately the same rate during heating. There were no changes in apparent molecular weights or relative amounts of the phycobilisome apoproteins during chromophore bleaching. Phycobilisomes in cell extracts from Anacystis nidulans resisted bleaching when suspended in medium of high phosphate concentration, but were bleached at 60–65°C within a few minutes when placed in diluted medium. The results indicate that phycobilisomes in vivo are stabilized by a mechanism other than high osmotic and ionic strength. Summary An overview of recent molecular analyses regarding origins of plastids in algal lineages is presented. Since different phylogenetic analyses can yield contradictory views of algal plastid origins, we have examined the effect of two distance measurement methods and two distance matrix tree-building methods upon topologies for the ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit nucleotide sequence data set. These results are contrasted to those from bootstrap parsimony analysis of nucleotide sequence data subsets. It is shown that the phylogenetic information contained within nucleotide sequences for the chloroplast-encoded gene for the large subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase, integral to photosynthesis, indicates an independent origin for this plastid gene in different plant taxa. This finding is contrasted to contrary results derived from 16S rRNA sequences. Possible explanations for discrepancies observed for these two different molecules are put forth. Other molecular sequence data which address questions of early plant evolution and the eubacterial origins of algal organelles are discussed. Offprint requests to: W. Martin  相似文献

A two year's limnological study of the Afram arm of the Volta Lake is reported. This arm of the lake is shallow; it receives frequent winds, which together with the floods of June-September create near isothermal conditions. The upper io m is well oxygenated with some oxygen, about 2 mg/l, reaching the bottom for most part of the year. It is poor in nutrients, values of total dissolved solids lying between 40–60 mg/l. But frequent algal blooms of Oscillatoria curviceps, O. princeps and Mycrocystis aeruginosa occur and the net primary production rate in some areas range between 8.4 and 142.5 mg C/m3/hr. It is suggested that the high tropical temperatures encourage quick recycling leading to relatively high organic production, thus making this arm non-oligotrophic.  相似文献

Biofuels made from algal biomass are being considered as the most suitable alternative energy in current global and economical scenario. Microalgae are known to produce and accumulate lipids within their cell mass which is similar to those found in many vegetable oils. The efficient lipid producer algae cell mass has been reported to contain more than 30% of their cell weight as lipids. According to US DOE microalgae have the potential to produce 100 times more oil per acre land than any terrestrial plants. This article reviews up to date literature on the composition of algae, mechanism of oil droplets, triacylglycerol (TAG) production in algal biomass, research and development made in the cultivation of algal biomass, harvesting strategies, and recovery of lipids from algal mass. The economical challenges in the production of biofuels from algal biomass have been discussed in view of the future prospects in the commercialisation of algal fuels.  相似文献

目的:近年来,赤潮在我国的发生呈增加的趋势,并造成了极大的经济损失。过去研究赤潮发生的机理主要集中在理化因素的影响,而越来越多的证据表明仅凭借营养盐等环境因素并不能解释大部分赤潮现象,藻际微生物可能发挥着重要作用。本文跟踪观测了2010年7月深圳大鹏湾海域爆发的锥状斯氏藻赤潮生消过程中细菌群落丰度种类的变化,从微生物与赤潮藻相互作用的角度探讨了赤潮的生消过程,讨论了不同时期不同关键菌群的特殊作用,为解释赤潮爆发和消亡提供了新的视角,为赤潮的监控和防治新方法的建立奠定了理论基础。方法:本文按时间顺序共采集该赤潮9次样本,利用末端限制性酶切片段长度多态性分析(T-RFLP)等分子生物学方法,通过主成分分析、克隆文库的构建,研究了微生物群落的变化过程,并探讨了特定种属的微生物在赤潮发生、发展和消亡过程中的作用。结果:从浮游细菌丰度来看,随着锥状斯氏藻细胞数量的波动,浮游细菌总数也随之呈现相应的变化。从浮游细菌的种类来看,它们主要属于变形杆菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)和厚壁菌门(Firmicutes)。从浮游细菌的动态变化过程来看,赤潮生消过程中,浮游细菌群落呈现出一定的演替现象,特别是在赤潮后期,群落主成分变化尤为显著。从关键菌群的作用来看,属于γ变形杆菌门的Alteromonas sp.一直占有较高的丰度,赤潮中后期受到菌群相互作用导致比例下降,而赤潮后期其他关键菌群的丰度的增高可能是导致赤潮消亡的重要原因。结论:本文利用T-RFLP这一DNA指纹技术分析微生物群落结构和多样性特征,通过研究赤潮生消过程中藻际浮游细菌群落的动态变化,发现随着赤潮的发展,浮游细菌群落发生着相应的变化。结果说明赤潮藻体丰度数量的改变影响着浮游细菌群落的组成。相对地,细菌群落的适应调整迅速造成赤潮藻体局部生长环境的改变,从而影响赤潮的发展过程。  相似文献

Morels, fungi from the genus Morchella, are popular edible mushrooms. However, little knowledge of their asexual reproduction and inaccessible pure mitospores hamper illumination of their life cycle. Herein, we successfully induced conidiation, conidial germination and chlamydospore formation in pure culture of Morchella sextelata. Conidiation proceeded via four morphologically distinct stages: development of the conidiophore stalk, stalk branching, phialide differentiation, and conidium production. Terminal and intercalary chlamydospores were formed on conidial hyphae. The development of conidiophores occurred earlier, with more conidia produced, in cross-mating cultures than in single-spore cultures. Mature conidia were spherical and 2.5–8 μm in diameter, with a vast majority (nearly 99%) 2.5–5 μm in diameter. Each conidium contained one to three nuclei (80.2% conidia contained one nucleus, 19.1% contained two nuclei, and 0.7% contained three nuclei). The conidial nucleus diameter was 1–2 μm. The nuclear mitosis in detached conidia that was observed may benefit their colony initiation. Additionally, morel conidia formed conidial anastomosis tubes. Conidia (mitospores) likely not only function as spermatia, but also as reproductive propagules in Morchella. Further research is imperative to elucidate the relationship between the conidia and chlamydospores, and their unique function in the morel life cycle.  相似文献

Abstract

The aim of this study was to quantify algal colonisation on anthropogenic surfaces (viz. building facades and roof tiles) using chlorophyll a (chl a) as a specific biomarker. Chl a was estimated as the initial fluorescence F0 of ‘dark adapted’ algae using a pulse-modulated fluorometer (PAM-2000). Four isolates of aeroterrestrial green algae and one aquatic isolate were included in this study. The chl a concentration and F0 showed an exponential relationship in the tested range between 0 and 400 mg chl a m?2. The relationship was linear at chl a concentrations <20 mg m?2. Exponential and linear models are presented for the single isolates with large coefficients of determination (exponential: r2 > 0.94, linear: r2 > 0.92). The specific power of this fluorometric method is the detection of initial algal colonisation on surfaces in thin or young biofilms down to 3.5 mg chl a m?2, which corresponds to an abundances of the investigated isolates between 0.2 and 1.5 million cells cm?2.  相似文献

This study investigated the effect of single and mixed algal diets on growth, fatty acid composition and ingestion rates for Dendraster excentricus larvae. Larvae were assigned to three single algal diet treatments Isochrysis galbana, Dunaliella tertiolecta or Rhodomonas sp. and four mixed algal diet treatments D. tertiolecta and Rhodomonas, I. galbana and D. tertiolecta, I. galbana and Rhodomonas, D. tertiolecta, Isochrysis galbana and Rhodomonas sp. Small amounts (0.36-0.6%) of stearic acid (18:0) were seen in the three algae used but a relatively large percentage (7-25%) of this SAFA was found in Dendraster larvae. The alga D. tertiolecta had the highest percentage (51.7%) of the short chain polyunsaturated fatty acid (PUFA) linolenic acid 18:3(n-3) and trace amounts (0.02-0.14%) of the long chain PUFAs eicosapentanoic (EPA, 20:5(n-3) and docosahexanoic acids (DHA, 22:6(n-3)). However, sand dollar larvae demonstrated the ability to elongate and desaturate shorter chain (18 carbon) polyunsaturated fatty acids (PUFA) to longer chain (20 carbon) n-3 PUFA. Thus high levels of 18:3(n-3) in D. tertiolecta led to high levels of EPA and low levels of 18:3(n-3) in Dendraster larvae fed this diet. Rhodomonas sp. had the highest percentage of stearidonic acid (18:4(n-3), 38.14%) and EPA (10.6%). Despite high levels of 18:4(n-3) in Rhodomonas sp. this acid was absent or found at very low levels in larvae fed this alga, or any combination of this alga. I. galbana had the highest percentage of DHA(14.3%) but was almost devoid of EPA (0.43%). Although Rhodomonas sp. and I. galbana had high levels of EPA and DHA sand dollar larvae did not incorporate higher levels of these long chain PUFAs into their lipids compared to those fed the alga D. tertiolecta. Dendraster larvae synthesized a number of 20 and 22 carbon non-methylene interrupted dienes (NMID), with levels increasing with larval stage. Higher ingestion rates were observed for Dendraster larvae fed single algal diets (Rhodomonas sp. or D. tertiolecta) and lower ingestion rates for those fed mixed algal diets. The highest ingestion rates were for 8-arm Dendraster larvae fed the large alga Rhodomonas sp. presented as a single algal diet. When fed a combination of three algae, selection of particles varied slightly depending on stage with 8-arm larvae ingesting slightly more of the larger algal cell in the mixture than 6-arm larvae. The present study suggests that regardless of the ratios given larvae might have an optimum ratio of different sized particles at which they can feed. The mixed algal diet of I. galbana and D. tertiolecta was the best algal diet leading to significantly larger larvae with high survival and development to metamorphosis. The single algal diets of Rhodomonas sp. or Dunaliella tertiolecta were the second and third best algal diets based on growth and survival to metamorphosis.  相似文献

The crude extract of the red seaweed, Asparagopsis sp. was evaluated for in vivo antibacterial activity against the shrimp vibrio pathogens. The algal extract was rationalized with commercial shrimp feed and orally administered for different duration of time followed by the artificial bacterial challenge experiment. In dose titration experiments, the oral administration of Asparagopsis sp. at a dosage of 850 mg kg–1 of biomass was highly efficacious in the treatment of natural infestations of Vibriosis in Penaeus monodon. The results of the confirmatory dose experiment revealed that the prophylactic treatment with moderate dose of 850 mg kg–1 of biomass day–1 for four weeks followed by 14 days of post infection therapy was highly effective in controlling Vibrio infection in shrimps. Moreover, results of the percent survival index and microbiological analysis clearly show that Asparagopsis extract incorporated medicated feed had broad therapeutic potential for managing shrimp Vibriosis. In addition, in vivo trials and results obtained in this work are based on the crude organic extract sourced from an unidentified Asparagopsis cryptic lineage, therefore further molecular analysis to identify the species will be required.  相似文献
