In the past as well as today there have been two conflicting opinions as to whether changes in the algal species in water bodies indicate polymorphism or the development of separate species. Similar changes were also found in High Rade Algae Pond (HRAP) used for wastewater treatment, effluent reclamation and protein production. To critically examine both opinions, samples of HRAP effluent were taken and the algal species identified and measured continuously, using conventional methods.Two main algal species were identified. These remained stable during all four monitoring sessions over a three-year period. The external changes observed in the algae were a reflection of controlled periods of organic loading and the conditions under which the pond was operated, such as retention time (a dependant of radiation), ambient temperature, effluent depth and aeration methods.Current address: Institute for Desert Research, Ben-Gurion University of the Negev, Sde-Boker, Israel 84990  相似文献

Biomass and lipid productivities of Chlorella vulgaris under different growth conditions were investigated. While autotrophic growth did provide higher cellular lipid content (38%), the lipid productivity was much lower compared with those from heterotrophic growth with acetate, glucose, or glycerol. Optimal cell growth (2 g l−1) and lipid productivity (54 mg l−1 day−1) were attained using glucose at 1% (w/v) whereas higher concentrations were inhibitory. Growth of C. vulgaris on glycerol had a similar dose effects as those from glucose. Overall, C. vulgaris is mixotrophic.  相似文献

Past studies have shown that the initiation of symbiosis between the Red-Sea soft coral Heteroxenia fuscescens and its symbiotic dinoflagellates occurs due to the chemical attraction of the motile algal cells to substances emanating from the coral polyps. However, the resulting swimming patterns of zooxanthellae have not been previously studied. This work examined algal swimming behaviour, host location and navigation capabilities under four conditions: (1) still water, (2) in still water with waterborne host attractants, (3) in flowing water, and (4) in flow with host attractants. Algae were capable of actively and effectively locating their host in still water as well as in flow. When in water containing host attractants, swimming became slower, motion patterns straighter and the direction of motion was mainly towards the host—even if this meant advancing upstream against flow velocities of up to 0.5 mm s−1. Coral-algae encounter probability decreased the further downstream of the host algae were located, probably due to diffusion of the chemical signal. The results show how the chemoreceptive zooxanthellae modify their swimming pattern, direction, velocity, circuity and turning rate to accommodate efficient navigation in changing environmental conditions.  相似文献

An in vitro oxidative stress test has been developed to assess pollutant tolerance in freshwater algae using Euglena gracilis as the test organism and FeSO4 and NaCl as the pollutants. The test evaluates free radical-mediated oxidative stress through the concomitant application of three biochemical assays: (1) the non-invasive, gas chromatographic-volatile headspace analysis of hydroxyl radicals (OH) using dimethyl-sulphoxide as a radical trap; (2) the spectroscopic determination of total antioxidant activity; (3) a fluorescent microscopy viability test. In vitro pollutant testing was devised to simulate contaminant loadings that impact urban retention ponds. E. gracilis was found to be tolerant to FeSO4 (2–10% (w/v)) and NaCl (10–5000 ppm) as indicated by high positive viabilities (ca. 100%) and low, or no OH production, as compared to controls. Total antioxidant activity increased with increasing pollutant loading suggesting that the organism has the capacity to enhance antioxidant defence in response to pollutant stress. This in vitro test provides a new approach to monitor the effects of water quality on the biological components of urban and/or polluted aquatic ecosystems. It also has a potential application in the identification of putative algal phytoremediators.  相似文献

Arachnitis uniflora, a myco-heterotrophic plant species, has fleshy tuberous roots colonized by the arbuscular mycorrhizal fungal genus Glomus (Phylum Glomeromycota). These roots produce apical and lateral propagules, both reported here for the first time. The objective of the study was to characterize the ontogeny and structure of the propagules, and to determine their function. Scanning electron microscopy, laser scanning confocal microscopy and light microscopy were used to study the ontogeny and structure of the propagules. Propagules developed either from cortical parenchyma cells or from cells immediately beneath the root cap; they developed a shoot meristem and cells in the basal region which were colonized by various fungal structures including hyphae and vesicles. These propagules may detach from the roots, establishing new plants.  相似文献

Monostroma latissimum (Kützing) Wittrock is an intertidal rock-dwelling green algal species of commercial importance in Japan. This article reports on the seasonality of its growth and occurrence from three distinct habitats of marine and estuarine regions in Tosa Bay, Japan, for two consecutive growth seasons. Thallus lengths of individuals and environmental parameters were monitored monthly between November 2005 and July 2007. Culture studies were carried out to establish the species-level identity of the specimens. Nuclear encoded ITS 1 (Internal Transcribed Spacer 1) sequences reveal that the naturally occurring strain of M. latissimum has identical nucleotide sequences to those of the commercially cultivated strain from the Shimanto river estuary. We have also found that the two strains are cross-fertilizing. Seasonal fluctuations in thallus length were found to be distinctive to the habitat where the strain grows and re-occur annually. Algal cover was highest during winter months and lowest during the summer. However, we found only a very weak positive correlation between chosen environmental parameters and thallus length. Appearance and decay of thalli occurred earlier in high saline habitats. Therefore, it is likely that salinity influences the maturation of microscopic sporophytes or the growth and survival of germlings in M. latissimum. Handling editor: P. Viaroli  相似文献

Pennate diatom Nitzschia palea can be cultured in outdoor vertical-bed photobioreactors to produce biodiesel. To assess the production of biomass and lipids, non-axenic cultures of Nitzschia palea were grown outdoors, and the growth of these cultures was measured biweekly. During the annual cycle of algal culture, the culture temperature ranged from 17.3 °C to 33.5 °C, the dry weight biomass ranged from 0.11 g l−1 to 0.25 g l−1, light energy] ranged from 1.94 Wm−2 to 3.9 Wm−2 and intracellular lipid content ranged from 7.1% to 11.4% of biomass weight after drying at 60 °C. Gas chromatography/mass spectroscopy (GC/MS) analysis of n-hexane extracts showed that the intracellular lipids were primarily C14:0 myristic acid (9.01%), C15:0 pentadecyclic acid (8.26%) and two types of C16:0, palmitic acid (41.13%) and palmitoleic acid (29.25%). Gel permeation analysis showed that carboxylic acids comprised 28.9% of lipids, 16.3% of monoglycerides, 27.3% of diglycerides and 24.3% of triglycerides. Alcoholysis of lipids resulted in the conversion of about 93.9% of fatty acids to equivalent fatty acid methyl esters (FAME) or biodiesel, which, on basis of wt%, consisted primarily of C15:0 methyl myristate (8.3%), C16:0 methyl pentadecanoate] (7.2%), C17:1methyl palmitoleate (28.7%) and methyl palimtate](39.8%).  相似文献

The Havana aeromycobiota diversity was studied from November 2010 to October 2011 using two complementary volumetric methods. A total of 35 fungal genera were characterised, 26 of them were recognised only by non-viable methods, six with viable methodology and the other three with both sampling methods. Furthermore, 47 species were identified by cultivation and the spores collected with the non-viable methodology. These could not be included in a specific genus, and thus, were categorised into five fungal types. In general, the main, spread worldwide, mitosporic fungi also predominated the Havana atmosphere. The predominant species were Cladosporium cladosporioides, Aspergillus flavus and Penicillium citrinum. Moreover, several Zygomycetes (Syncephalastrum racemosum, Rhizopus stolonifer and Rhizopus oryzae), Ascomycetes (Chaetomium globosum) and Basidiomycetes such as Coprinus or Ganoderma were isolated. In the present paper, the review of the airborne fungi conducted in previous studies in Cuba was completed by the detection of two new genera and the first isolation of ten new records in the Cuban atmosphere. Currently, very few studies address the relationship between diversity and biomass/lipid production in primary producer communities for biofuel production. Basic studies on the growth of microalgal communities, however, provide evidence of a positive relationship between diversity and biomass production. Recent studies have also shown that positive diversity–productivity relationships are related to an increase in the efficiency of light use by diverse microalgal communities. Here, we hypothesize that there is a relationship between diversity, light use, and microalgal lipid production in phytoplankton communities. Microalgae from all major freshwater algal groups were cultivated in treatments that differed in species richness and functional group richness. Polycultures with high functional group richness showed more efficient light use and higher algal lipid content with increasing species richness. There was a clear correlation between light use and lipid production in functionally diverse communities. Hence, a powerful and cost‐effective way to improve biofuel production might be accomplished by incorporating diversity related, resource‐use‐dynamics into algal biomass production.  相似文献

ABSTRACT

Raphidiopsis (Cylindrospermopsis) raciborskii is one of the most studied potentially harmful cyanobacteria. Single environmental factors such as increased temperature or light are reported to be promoters of R. raciborskii growth, but the interaction of two or more promoting factors is less understood. The performance of two strains of R. raciborskii (MVCC19, Uruguayan and LP1, Brazilian) were evaluated under acclimation and temperature shifts (25–32°C) in combination with two transitions from phosphorus (P) sufficiency to limiting growth conditions. When subjected to transition from high P sufficiency to a P-limiting state, strains were able to grow only at the warmer temperature if previously acclimated. The MVCC19 strain showed higher specific growth rates and a shorter growth phase than LP1. Morphological differences were also found: the MVCC19 strain produced shorter filaments, while the LP1 strain increased in length and the number of cells per filament. The results show the positive effect of high temperature on the ability of R. raciborskii to withstand P-limiting conditions, which may confer resilience of populations to periods of severe nutrient limitation in warm lakes. This finding contributes to the understanding of the success of this species in diverse environmental conditions. The effect of temperature on the tolerance of nutrient deficiency and the performance of strains under lake conditions suggests the need for a drastic reduction in nutrient loads to avoid R. raciborskii dominance in warmer lakes.  相似文献
