漆酶是一种含铜多酚氧化酶,在白腐菌中普遍存在,少数低等真菌和植物中也产生,多为分泌型糖蛋白。至少20种漆酶得到了分离和纯化。该酶是一种氨基酸残基在500个左右的单体酶,一般都为酸性蛋白,含有4个铜离子,形成3个活性区域;表面一些氨基酸被不同程度地糖基化。晶体结构和其它一些波谱学研究解释了其空间结构和可能的电子传递机制。运用PCR技术和cDNA文库技术,越来越多的漆酶基因被克隆,许多来源的基因都是以家族形式存在于染色体上的。已研究的漆酶基因中都含有10个左右的内含子,这些内含子在活性域位置上有比较高的保守性。一些特殊序列的存在与否决定了该酶的表达形式-诱导型或组成型,诱导型菌株的调控序列中含有一段受酚类化合物作用的序列,而不含有该序列的酶基因则都是组成型表达的。漆酶在S.cerevisiae、Trichodermareesei、A.oryzaeTATAamylase和Pichiapasti等异源表达系统中有成功表达的报道。漆酶的应用集中在以下几方面:漆酶参与的有机合成;生物检测;有毒化合物的消除;工业废水处理;纸浆的生物漂白;等等。  相似文献

Xylanase production by the Antarctic psychrophilic yeast Cryptococcus adeliae was increased 4.3 fold by optimizing the culture medium composition using statistical designs. The optimized medium containing 24.2 g l−1 xylan and 10.2 g l−1 yeast extract and having an initial pH of 7.5 yielded xylanase activity at 400 nkat (nanokatal) ml−1 after 168-h shake culture at 4°C. In addition, very little endoglucanase, β-mannanase, β-xylosidase, β-glucosidase, α-l-arabinofuranosidase, and no filter paper cellulase activities were detected. Among 12 carbon sources tested, maximum xylanase activity was induced by xylan, followed by lignocelluloses such as steamed wheat straw and alkali-treated bagasse. The level of enzyme activity produced on other carbon sources appeared to be constitutive. Among the complex organic nitrogen sources tested, the xylanase activity was most enhanced by yeast extract, followed by soymeal, Pharmamedia (cotton seed protein), and Alburex (potato protein). A batch culture at 10°C in a 5-l fermenter (3.5-1 working volume) using the optimized medium gave 385 nkat at 111 h of cultivation. The crude xylanase showed optimal activity at pH 5.0–5.5 and good stability at pH 4–9 (21 h at 4°C). Although the enzyme was maximally active at 45°–50°C, it appeared very thermolabile, showing a half-life of 78 min at 35°C. At 40°–50°C, it lost 71%–95% activity within 5 min. 1.以双对硝基苯磷酸钠为底物,测得蝮蛇毒磷酸二酯酶的最适pH在10左右,最适温度在60℃左右;酶在37℃和60℃的Km分别为5.65×10~(-4)M和7.97×10~(-4)M。 2.酶在pH5—10.5范围和50℃以下稳定。 3.钙离子和镁离子对酶活力有激活作用,但钡离子、镁离子、锌离子、二价铅离子、钴离子、二价和三价铁离子、锰离子、镍离子等都有不同程度地抑制作用。 4.除乙酸根离子外,碳酸根,硫酸根,乙二胺四乙酸根、磷酸根,钼酸根,酒石酸根,柠檬酸根,亚硝酸根,巴比妥酸根和硫代硫酸根等阴离子都有不同程度地抑制作用。 5.蝮蛇毒磷酸二酯酶能充分水解RNA和DNA但后者的水解速度比前者快许多。  相似文献

柑叶片蔗糖酶的分离纯化及其部分性质的研究   总被引:8,自引:0,他引:8  
柑(Citrusreticulata Blanco)幼叶中存在高活性的酸性蔗糖酶。经硫酸铵盐析、DEAE-琼脂糖离子交换层析、SephacrylS-200 凝胶层析纯化,活性回收率6.4% ,纯化倍数179.2 倍。纯化的酶经聚丙烯酰胺凝胶电泳显示单一蛋白带,SDS-PAGE显示1 条蛋白带,其亚基分子量40 kD。用SephacrylS-200 凝胶层析法测得分子量为80 kD。推测该酶由两个相同亚基构成。以蔗糖为底物测定该酶的表观Km 为1.6×10- 2 m ol·L- 1,Vm ax为100 m g 还原糖·m g- 1蛋白质·h- 1。最适pH 5.0,酸碱稳定区在pH 4.5—5.5 之间。最适温度55℃  相似文献

Methods of potentiometry, turbidimetry, colorimetry, IR spectroscopy, and element analysis were used to investigate the conditions of formation and the properties of non-stoichiometric polyelectrolyte complexes of chitosan hydrochloride (CHC) and sodium dextransulfate (the molecules of both polysaccharides appear as semirigid chains, but their charges are opposite). It was determined that the complexes' formation of polyelectrolytes studied is predominantly electrostatic in the presence of urea. As was also found turbidity and stability of the polycomplexes solutions depended markedly on pH value of CHC and a nature of the low-molecular-weight salts added. The complexes obtained were soluble in water, aqueous urea, and water-organic mixtures. The extent of solubility depended on the composition of the complexes and could be influenced by addition of appropriate concentrations of certain low-molecular-weight salts.  相似文献

Morningness scales have been translated into several languages, but a lack of normative data and methodological differences make cross‐cultural comparisons difficult. This study examines the psychometric properties and factor structure of the Composite Scale of Morningness (CSM) in samples from five countries: France (n=627), Italy (n=702), Spain (n=391), Thailand (n=503), and Australia (n=654). Strong national differences are identified. A quadratic relationship between age and CSM total score was apparent in the Australian data with a downward trend after age 35 yrs. There was no age effect in any sample in the range from 18 to 29 yrs. Factor analysis identified a three‐factor solution in all groups for both men and women. Tucker's congruence coefficients indicate that: (1) this solution is highly congruent between sexes in each culture, and (2) a morning affect factor is highly congruent between cultures. These results indicate there are national differences in factorial structure and that cut‐off scores used to categorize participants as morning‐ and evening‐types should be established for different cultural and age groups.  相似文献

1. Aggregative feeding of larvae is widespread in the Lepidoptera, and many hypotheses have been proposed to explain the adaptive significance of this trait. Group feeding occurs disproportionately more in species with aposematic, chemically defended larvae compared with species with cryptic, non‐chemically defended larvae, consistent with the hypothesis that group feeding provides an enhanced aposematic signal to natural enemies. Most species characterised as having chemically defended larvae are cryptic during the first instar, when they are most highly aggregated and most vulnerable to predation. 2. The benefits of group feeding in terms of decreased predation were explored for first‐instar larvae of the pipevine swallowtail, Battus philenor, a species that sequesters aristolochic acids from its Aristolochia host plant and exhibits aposematism in later instars and as adults. We found that groups of larvae with experimentally enhanced aristolochic acid content had significantly lower survivorship due to predation both in the field and in laboratory experiments compared with groups of larvae without enhanced chemical defence. 3. A laboratory experiment found that aristolochic acid does not deter the generalist predator Hippodamia convergens. 4. No evidence was found that was consistent with the hypothesis that group feeding and increased sequestered chemical defence interact to decrease rates of larval mortality in non‐aposematic, first‐instar larvae. Future research on chemical defence, aposematism, and aggregative feeding should continue to appreciate that particular chemical defences and feeding behaviours are not universally effective against all natural enemies.  相似文献

The antagonist actions of three sub-series of tetrahydro- β -carbolines at the serotonin 2B (5HT 2B) contractile receptor in the rat stomach fundus are analyzed in relation to the physicochemical properties of the molecules. Significant correlations are obtained between the 5HT 2B receptor antagonist affinity and the hydrophobic, steric, electronic, hydrogen bond acceptor and some indicator variables of substituents. Based on these findings, the mode of actions of these congeneric series and future strategy to synthesize more potential compounds are discussed.  相似文献
