Aegilops markgrafii contains resistance genes to powdery mildew, leaf rust and stripe rust, and also has high crude protein and lysine contents, which can be useful for wheat improvement. These important traits are localized on different chromosomes. Disomic Triticum aestivum-Ae. markgrafii addition lines and euploid introgression lines showing leaf-rust and powdery mildew resistance were screened with RAPDs to detect chromosome-specific markers which can accelerate the breeding process. RAPD markers for all six available disomic addition lines were obtained. The additional chromosomes B, C, D, E, F and G were identified by three, three, three, two, one and seven primers, respectively. All three chromosome-B-specific RAPD markers demonstrated the presence of alien chromatin in the leaf-rust-resistant 42-chromosome introgression lines as well as in the segregating progeny. The three chromosome-C-identifying primers also demonstrated the presence of that chromosome in powdery mildew-resistant euploid introgression lines. The substitution lines (5A)5C and (5D)5C with different genetic backgrounds for both parents, in comparison to the lines mentioned above, showed the chromosome C-specific band with only two of the three primers. The chromosome F-specific primer and a primer evident on all the Ae. markgrafii chromosomes analysed did not generate the expected fragments on the chromosome Fdel addition line, indicating that the markers are located on the deleted part of chromosome F.  A new Hessian fly (Mayetiola destructor) resistance gene from Aegilops ventricosa and its transfer to hexaploid wheat is described. The 4D(4Mv) substitution line H-93-33 derived from the cross [(Triticum turgidum H-1-1×Aegilops ventricosa no. 11)×Triticum aestivum H-10-15] was highly resistant to the Spanish population tested. Resistance seemed to be inherited as a single dominant factor in the F2 generation resulting from a cross of H-93-33 with its susceptible parent (H-10-15). Resistance in Ae. venticosa no. 10 was located on chromosome 4Mv using Mv wheat/Ae. ventricosa addition lines. The resistance gene transferred from Ae. ventricosa no. 11 to H-93-33 (H27) is allelic with respect to that of Ae. ventricosa no. 10 and is non-allelic with respect to the genes H3 and H6 from Monon and Caldwell respectively. The assignment of H27 gene to chromosome 4Mv is further supported by its linkage to a gene encoding isozyme Acph-Mv1, previously located on chromosome 4Mv in the line H-93-33. A new marker from homoeologous chromosome group 4 (Amp-Mv2) present in H-93-33 and the 4Mv addition line is described. Received: 12 October 1996 / Accepted: 22 November 1996  相似文献

Summary Thirty lines from a cross between VPM/ Moisson 421 and Selection 101 were used in the study to determine whether strawbreaker foot rot resistance derived from Aegilops ventricosa was associated with an allele for endopeptidase. The progeny examined for foot rot lesions represented F2 derived F5 lines and enzyme assays were done on F6 seedlings. The results indicate that the wheat and VPM/Moisson 421 endopeptidase alleles are distinctly different. The endopeptidase allele frequencies of 30 lines were compared with strawbreaker foot rot resistance as measured by the lesion severity index. The results demonstrate a close association between the gene for strawbreaker foot rot resistance and the endopeptidase allele derived from Ae. ventricosa.  相似文献

Summary The gene encoding a variant of alcohol dehydrogenase, Adh-, has been found to be associated with the chromosome of the Mv genome which is present in type 9 wheat/Aegilops ventricosa addition line, to which the genes for protein CM-4 and for a phosphatase variant, Aph-v, had been previously assigned. Transfer line H-93-33, which has 42 chromosomes and has been derived from the cross (Triticum turgidum x Ae. ventricosa) x T. aestivum, carries genes encoding all three biochemical markers. Linkage between these genes has been demonstrated by analysis of individual kernels of the F2 (H-93-33 x T. aestivum cv. Almatense H-10-15). A study of the hybrids of line H-93-33 with T. aestivum H-10-15 and with the 4DS ditelosomic line has confirmed that, as suspected, the linkage group corresponds to chromosome 4Mv from Ae. ventricosa. Additionally, it has been found that the previously reported resistance of line H-93-33 to powdery mildew (Erysiphe graminis) is also linked to the biochemical markers; this indicates that either the gene responsible for it is different from that in lines H-93-8 and H-93-35, or that a translocation between two different Mv chromosomes has occurred in line H-93-33.  相似文献

Summary In the progeny of a hybrid between monotelosomic line 3B of Chinese Spring wheat and Chinese Spring — Aegilops longissima ditelosomic addition line G a cytologically stable strain was selected consisting of 20 wheat chromosome pairs, one pair of telosomic chromosome 3BL and one pair of telosomic longissima chromosome G. Inoculating Chinese Spring — Aegilops longissima addition and substitution lines with ten different powdery mildew isolates, partial resistance was observed. The infection grade as well as the number of spores/cm2 leaf area were significantly reduced.  相似文献

Prolamin and resistance gene families are important in wheat food use and in defense against pathogen attacks, respectively. To better understand the evolution of these multi‐gene families, the DNA sequence of a 2.8‐Mb genomic region, representing an 8.8 cM genetic interval and harboring multiple prolamin and resistance‐like gene families, was analyzed in the diploid grass Aegilops tauschii, the D‐genome donor of bread wheat. Comparison with orthologous regions from rice, Brachypodium, and sorghum showed that the Ae. tauschii region has undergone dramatic changes; it has acquired more than 80 non‐syntenic genes and only 13 ancestral genes are shared among these grass species. These non‐syntenic genes, including prolamin and resistance‐like genes, originated from various genomic regions and likely moved to their present locations via sequence evolution processes involving gene duplication and translocation. Local duplication of non‐syntenic genes contributed significantly to the expansion of gene families. Our analysis indicates that the insertion of prolamin‐related genes occurred prior to the separation of the Brachypodieae and Triticeae lineages. Unlike in Brachypodium, inserted prolamin genes have rapidly evolved and expanded to encode different classes of major seed storage proteins in Triticeae species. Phylogenetic analyses also showed that the multiple insertions of resistance‐like genes and subsequent differential expansion of each R gene family. The high frequency of non‐syntenic genes and rapid local gene evolution correlate with the high recombination rate in the 2.8‐Mb region with nine‐fold higher than the genome‐wide average. Our results demonstrate complex evolutionary dynamics in this agronomically important region of Triticeae species.  相似文献

Relationships between the chromosomes of Aegilops umbellulata and wheat   总被引:3,自引:0,他引:3  
 A comparative genetic map of Aegilops umbellulata with wheat was constructed using RFLP probes that detect homoeoloci previously mapped in hexaploid bread wheat. All seven Ae. umbellulata chromosomes display one or more rearrangements relative to wheat. These structural changes are consistent with the sub-terminal morphology of chromosomes 2 U, 3 U, 6 U and 7 U. Comparison of the chromosomal locations assigned by mapping and those obtained by hybridization to wheat/Ae. umbellulata single chromosome addition lines verified the composition of the added Ae. umbellulata chromosomes and indicated that no further cytological rearrangements had taken place during the production of the alien-wheat aneuploid lines. Relationships between Ae. umbellulata and wheat chromosomes were confirmed, based on homoeology of the centromeric regions, for 1 U, 2 U, 3 U, 5 U and 7 U. However, homoeology of the centromeric regions of 4 U with wheat group-6 chromosomes and of 6 U with wheat group-4 chromosomes was also confirmed, suggesting that a re-naming of these chromosomes may be pertinent. The consequences of the rearrangements of the Ae. umbellulata genome relative to wheat for gene introgression are discussed. Received: 10 July 1997 / Accepted: 19 September 1997  相似文献

Two contrasting genome-specific DNA sequences were isolated from Aegilops speltoides (wild goat grass) and Hordeum chilense (wild barley), each representing more than 1 % of the genomes. These repetitive DNA fragments were identified as being genome-specific before cloning by genomic Southern hybridization (using total genomic DNA as a probe), and hence extensive screening of clones was not required. For each fragment, up to six recombinant plasmid clones were screened and about half were genome-specific. Clone pAesKB52 from Ae. speltoides was a 763 by EcoRI fragment, physically organized in simple tandem repeats and shown to localize to sub-telomerec chromosome regions of species with the Triticeae S-genome by in situ hybridization to chromosomes. The sequence data showed an internal duplication of some 280 bp, which presumably occurred before sequence amplification and dispersion, perhaps by unequal crossing-over or reciprocal translocation. In situ hybridization showed that the sequence distribution varied between closely related (S-genome) species. Clone pHcKB6 was a 339 by DraI fragment from H. chilense, also tandemly repeated but more variable; loss of the DraI site resulting in a ladder pattern in Southern blots which had little background smear. In situ hybridization showed that the tandem repeats were present as small clusters dispersed along all chromosome arms except at a few discrete regions including the centromeres and telomeres. The clone hybridized essentially specifically to the H-genome of H. chilense and hence was able to identify the origin of chromosomes in a H. chilense x Secale africanum hybrid by in situ hybridization. It has a high A + T content (66%), small internal duplications, and a 50 by degenerate inverted repeat. We speculate that it has dispersed by retrotransposition in association with other sequences carrying coding domains. The organization and evolution of such sequences are important in understanding long-range genome organization and the types of change that can occur on evolutionary and plant breeding timescales. Genome-specific sequences are also useful as markers for alien chromatin in plant breeding.  相似文献

Summary Each of the three genomes in hexaploid wheat controls the expression of a specific lectin in the embryo. The chromosomes which control their synthesis were determined using nullisomic-tetrasomic and inter-varietal chromosome substitution lines of Chinese Spring. All three wheat lectins were shown to be controlled by the homoeologous group 1 chromosomes. Using ditelosomic lines of Chinese Spring the lectin genes could be localized on the long arms of chromosomes 1A and 1D. Inter-specific addition and substitution lines of Aegilops umbellulata chromosomes to Chinese Spring indicated that chromosome 1U, which is homoeologous to the group 1 chromosomes of wheat, controls lectin synthesis.  相似文献
