Hydrophobic protein (H protein) was isolated from membrane fractions of Bacillus subtilis and constituted into artificial membrane vesicles with lipid of B. substilis. Glutamate was accumulated into the vesicle when a Na+ gradient across the membrane was imposed. The maximum effect of Na+ on the transport was achieved at a concentration of about 40 mM, while the apparent Km for Na+ was approximately 8 mM. On the other hand, Km for glutamate in the presence of 50 mM Na+ was about 8 μM. Increasing the concentration of Na+ resulted in a decrease in Km for glutamate, maximum velocity was not affected. The transport was sensitive to monensin (Na+ ionophore).Glutamate was also accumulated when pH gradient (interior alkaline) across the membrane was imposed or a membrane potential was induced with K+-diffusion potential. The pH gradient-driven glutamate transport was sensitive to carbonylcyanide m-chlorophenylhydrazone and the apparent Km for glutamate was approximately 25 μM.These results indicate that two kinds of glutamate transport system were present in H protein: one is Na+ dependent and the other is H+ dependent.  相似文献

Mg2 + dependent —adenosine triphosphatase activity has been studied in the muscle, brain, kidney and liver tissues of frog,Rana hexadactyla (Lesson) after sciatectomy and induced chronic ammonia stress. The enzyme activity decreased in the tissues of the denervated frog. The activity of the enzyme increased in all the tissues of the normal and denervated frogs except in the denervated muscle when ammonium lactate was infused intraperitoneally.  相似文献

An ammonia limited chemostat culture of Gymnodinium simplex (Lohm.) Kofoid & Swezy was perturbed with ammonia and fluctuations in the free intracellular amino acid pools were followed 80 min. The steady-state value of glutamate was 2.07 ± 10-15 mol cell-1 and of glutamine was 0.31 ± 10-15 mol cell-1. Five minutes after the perturbation, a substantial rise in glutamine was observed with a corresponding decrease in glutamate. This is considered a result of glutamine synthetase acting as the primary ammonia assimilating enzyme. The level of ammonia and the major free amino acids reached a maximum 10 min after the perturbation and then slowly decreased.  相似文献

Laminaria longicrucis De la Pylaie took up exogenous nitrate under both summer and winter conditions. During July and August no NO3- was detected in the ambient water or in algal tissues although it was present in both in February. Discs (2.3 cm diam.) of thin blade tissue were incubated with NO3- at four temperatures, with and without illumination. Similar values Jor NO3- uptake were found for both summer and winter collected plants when measured in light at 0 C. An apparent K of 4–6 μM was recorded for both types of plants; the Vmax ranged from 7 to 10 μmol h-1 g-1 dry wt measured in ca. 1800 μW cm-2 of cool-white fluorescent light. Uptake rates at 5 C were 66%, and at 0 C 30% of those for controls run at 15 C. The alga scavenged NO3- from solutions <0.5 μM. Ammonia did not inhibit NO3- uptake. Antibiotic pretreatment reduced NO3- uptake by a maximum of 12%. Nitrite uptake was inhibited in proportion to the concentration of NO3- in the medium.  相似文献

从猪胰脏的酸醇提取液中纯化了一个新的活性多肽——胰岛素拮抗肽,它在整体和细胞水平上对胰岛素都有明显的拮抗作用。猪胰脏的酸醇提取液经CM-52、BioGel P-6、DEAE-52及RP-HPLC纯化后,可得到纯的胰岛素拮抗肽。它能剂量相关地抑制胰岛素在离体大鼠脂肪细胞中的促脂合成活性,抑制50%胰岛素活性时所需的胰岛素拮抗肽为2.0×10~(-10)mol/L与被拮抗的胰岛素剂量在同一水平上。该肽含有较多的碱性氨基酸,分子量的3 000,其N-末端是封闭的。胰岛素拮抗肽的上述理化特征及其对胰岛素的拮抗活性均不同于目前已知的胰脏活性多肽。它对脂肪细胞中胰岛素的拮抗作用可能具有重要的生理意义。  相似文献

The influence of ammonia and lactate on cell growth, metabolic, and antibody production rates was investigated for murine hybridoma cell line 163.4G5.3 during batch culture. The specific growth rate was reduced by one-half in the presence of an initial ammonia concentration of 4 mM. Increasing ammonia levels accelerated glucose and glutamine consumption, decreased ammonia yield from glutamine, and increased alanine yield from glutamine. Although the amount of antibody produced decreased with increasing ammonia concentration, the specific antibody productivity remained relatively constant around a value of 0.22 pg/cell-h. The specific growth rate was reduced by one-half at an initial lactate concentration of 55 mM. Although specific glucose and glutamine uptake rates were increased at high lacatate concentration, they showed a decrease after making corrections for medium osmolarity. The yield coefficient of lactate from glucose decreased at high lactate concentrations. A similar decrease was observed for the ammonia yield coefficient from glutamine. At elevated lactate concentrations, specific antibody productivities increased, possibly due to the increase in medium osmolarity. The specific oxygen uptake rate was insensitive to ammonia and lactate concentrations. Addition of ammonia and lactate increased the calculated metabolic energy production of the cells. At high ammonia and lactate, the contribution of glycolysis to total energy production increased. Decreasing external pH and increasing ammonia concentrations caused cytoplasmic acidification. Effect of lactate on intracellular pH was insignificant, whereas increasing osmolarity caused cytoplasmic alkalinization.  相似文献

Laboratory experiments have shown appreciable losses of ammonia after injection of anhydrous ammonia into dry and wet soils. In this study losses of ammonia injected into a moist (tension 10 kPa), dry (tension 160 kPa) and a wet (tension 1.6 kPa) sandy loam were measured under field conditions using wind tunnels. Losses were insignificant from a moist soil. However losses from a dry and a wet soil were 20% and 50% of injected ammonia, respectively. From the dry soil, losses of gaseous ammonia took place within the first hours after injection, which indicates a rapid transport through cracks and voids. From the wet soil, 20% of the injected ammonia was lost more gradually between 6 h and 6 d. This indicates that upward movement of water due to evaporation may be the cause of these ammonia losses which proceeded for longer periods.  相似文献

Cell wall lytic activity was detected in the culture medium and cell wall of 1AM Chlorella ellipsoidea C-87. The enzymes of both fractions had their highest activity at pH 5. The lytic activity bound to the cell wall consisted of a polysaccharide releasing enzyme, an exo-type enzyme releasing disaccharide, and glucosidase; but only the polysaccharide releasing enzyme was solubilized by lithium chloride. A polysaccharide releasing enzyme with a molecular weight around 40 kDa was isolated from the culture medium. Hemicellulose is degraded by the polysaccharide releasing enzyme, and the rigid wall by the exo-type enzyme.  相似文献
