分泌性蛋白酶是红色毛癣菌致病的潜在毒力因子。在构建红色毛癣菌6个不同时间段cDNA文库的基础上,共获得了9683条uniqueESTs,通过生物信息学分析从中得到了18个可能的分泌性蛋白酶的EST序列,包括4个分泌性肽酶、1个分泌性金属蛋白酶、2个细胞外丝氨酸蛋白酶、1个分泌性天冬氨酸蛋白酶、9个分泌性枯草杆菌蛋白酶样丝氨酸蛋白酶、1个空泡丝氨酸蛋白酶。这些分泌性蛋白酶在红色毛癣菌感染过程中可能分别与其获得营养、扩大侵袭范围及激起宿主免疫应答有关,这些结果为进一步研究红色毛癣菌感染和发病机制提供了重要的分子基础和线索。 相似文献

Interaction of ozone pollution and light effects on photosynthesis in a forest canopy experiment 总被引:3,自引:3,他引:3
Ozone pollution may reduce net carbon gain in forests, yet data from mature trees are rare and the effects of irradiance on the response of photosynthesis to ozone remain untested. We used an open-air system to expose 10 branches within the upper canopy of an 18-m-tall stand of sugar maple (Acer saccharum Marsh.) to twice-ambient concentrations of ozone (95nmol mol?1, 0900 to 1700, 1 h mean) relative to 10 paired, untreated controls (45nmol mol?1) over 3 months. The branch pairs were selected along a gradient from relatively high irradiance (PPFD 14.5 mol m?2 d?1) to deep shade (0.7mol m?2 d?1). Ozone reduced light-saturated rates of net photosynthesis (Asat) and increased dark respiration by as much as 56 and 40%, respectively. Compared to sun leaves, shade leaves exhibited greater proportional reductions in Asat and had lower chlorophyll concentrations, quantum efficiencies, and leaf absorptances when treated with ozone relative to controls. With increasing ozone dose over time, Asat became uncoupled from stomatal conductance as ratios of internal to external concentrations of carbon dioxide increased, reducing water-use efficiency. Ozone reduced net photosynthesis and impaired stomatal function, with these effects depending on the irradiance environment of the canopy leaves. In this paper a number of experiments with the purple bacteria Rhodospirillum rubrum and Rhodopseudomonas capsulata is described in which the total fluorescence yield and/or the total fraction of reaction centers closed after a picosecond laser pulse were measured as a function of the pulse intensity. The conditions were such that the reaction centers were either all in the open or all in the closed state before the pulse arrived. These experiments are analysed using the theoretical formalism discussed in the preceding paper (Den Hollander, W.T.F., Bakker J.G.C., and Van Grondelle, R., Biochim. Biophys. Acta 725, 492–507). From the experimental results the number of connected photosynthetic units, λ, the rate of energy transfer between neighboring antenna molecules, kh, and the rate of trapping by an open reaction center, kot, can be estimated. For R. rubrum it is found that λ = 14−17, kh = (1−2)·1012 s−1 and kot = (4−6)·1011 s−1, for Rps. capsulata λ ≈ 30, kh ≈ 4·1011 s−1 and kot ≈ 3·1011 s−1. The findings are discussed in terms of current models for the structure of the antenna and the kinetic properties of the decay processes occurring in these purple bacteria. 相似文献

ABSTRACT. Myrionecta rubra, a ubiquitous planktonic ciliate, has received much attention due to its wide distribution, occurrence as a red tide organism, and unusual cryptophyte endosymbiont. Although well studied in coastal waters, M. rubra is poorly examined in the open ocean. In the Irminger Basin, North Atlantic, the abundance of M. rubra was 0–5 cells/ml, which is low compared with that found in coastal areas. Distinct patchiness (100 km) was revealed by geostatistical analysis. Multiple regression indicated there was little relationship between M. rubra abundance and a number of environmental factors, with the exception of temperature and phytoplankton biomass, which influenced abundance in the spring. We also improve on studies that indicate distinct size classes of M. rubra; we statistically recognise four significantly distinct width classes (5–16, 12–23, 18–27, 21–33 μm), which decrease in abundance with increasing size. A multinomial logistic regression revealed the main variable correlated with this size distribution was ambient nitrate concentration. Finally, we propose a hypothesis for the distribution of sizes, involving nutrients, feeding, and dividing of the endosymbiont. 相似文献

目的探讨我国不同地域红色毛癣菌分离株的遗传多样性。方法采用随机扩增DNA多态性(RAPD)方法对来源于我国不同地域(江苏南京,山东济南,广东广州)的32株红色毛癣菌临床分离株进行DNA多态性分析。结果红色毛癣菌种内差异明显,根据遗传相似性分成三大聚类群,与地域差异及取材部位无明显相关性,而与表型具有一定相关性。结论随机扩增DNA多态性方法可用于红色毛癣菌的DNA分型,其DNA带型具有一定的遗传变异性,与菌株表型有一定关系,与地域差异、侵犯部位无明显相关性。 相似文献

Using the whole-vacuolar mode of the patch clamp technique, we studied the effect of the chaotropic anions thiocyanate and nitrate on the electric currents generated by the proton pumping tonoplast ATPase and pyrophosphatase (PPiase), respectively, in vacuoles from suspension cells of Chenopodium rubrum L. Addition of KNO3 (150–250 m M ) or KSCN (70–150 m M ), and ATP (5 m M , obligatory) irreversibly inhibited the subsequent electric current through the tonoplast ATPase driven by 1 m M ATP, whereas PPiase-activity by 50 μ M PPi remained unaffected. The kinetics of inhibition, indicative of ATPase disintegration by the chaotropic anions, follows a single exponential (τ= 3.44 min). However, apparent ATPase disintegration did not measurably increase the tonoplast conductance. We conclude that, by contrast to organellar F-ATPases, upon disintegration the transmembrane proteolipid of the V-ATPase does not act as a proton conductor which, in the presence of chaotropic anions, like chloride or nitrate, would severely perturb solute compartmentation in the plant cell. 相似文献
