研究了B型烟粉虱在4种葫芦科寄主植物黄瓜、节瓜、苦瓜和丝瓜上的发育和繁殖特性.结果表明,B型烟粉虱在节瓜上的世代发育历期最短,为19.3 d,在苦瓜上的世代发育历期最长,为29.0 d;世代存活率在黄瓜上最高,为92.85%,在苦瓜上最低,为53.08%;平均单雌产卵量在黄瓜上最多,为187.4粒,苦瓜上最少,为30.0粒;雌成虫寿命以在黄瓜上最长,为25.2 d,在苦瓜上最短,为10.9 d.B型烟粉虱在黄瓜、节瓜、苦瓜和丝瓜上的内禀增长率(rm)分别为0.1453、0.1429、0.0616和0.1055.综合比较4种葫芦科植物,黄瓜是B型烟粉虱的最适宜寄主. 相似文献

【目的】考察固态和液态两种培养方式对丝状真菌华根霉(Rhizopus chinensis)CCTCC M201021产胞外蛋白的影响,以加深对微生物固、液态培养特异性产酶的认识。【方法】利用成分相同的培养基对华根霉分别进行平板固态培养和液态培养,提取华根霉所产胞外蛋白,采用二维凝胶电泳(2-DE)结合质谱分析,分离鉴定差异蛋白。【结果】固态培养下华根霉所产胞外蛋白的蛋白酶活性是液态培养的9.2倍。胞外蛋白质组数据分析表明,华根霉固态和液态培养所产胞外蛋白存在显著差异,其中约70%是固态和液态培养下各自产生的特有蛋白。质谱鉴定进一步表明,固、液态培养对华根霉产胞外蛋白的种类和表达量都有显著影响,其中水解酶类所占比例较大,主要是与蛋白质降解相关的蛋白。【结论】固态和液态不同培养方式影响了华根霉产胞外蛋白的组成,有些基因只在特定培养方式下表达。固态培养下华根霉产胞外蛋白酶种类相对更多以及大部分蛋白酶的上调表达,可能是固态培养下胞外蛋白酶活性很高的原因。研究结果提示需要注意固液态不同培养方式下丝状真菌胞外酶的筛选和生产可能存在的不一致性。 相似文献

An effort has been made for the first time in Asia's largest brackish water lagoon, Chilika, to investigate the spatio-temporal variability in primary productivity (PP), bacterial productivity (BP), bacterial abundance (BA), bacterial respiration (BR) and bacterial growth efficiency (BGE) in relation to partial pressure of CO2 (pCO2) and CO2 air–water flux and the resultant trophic switchover. Annually, PP ranged between 24 and 376 µg C L?1 d?1 with significantly low values throughout the monsoon (MN), caused by light limitation due to inputs of riverine suspended matter. On the contrary, BP and BR ranged from 11.5 to 186.3 µg C L?1 d?1 and from 14.1 to 389.4 µg C L?1 d?1, respectively, with exceptionally higher values during MN. A wide spatial and temporal variation in the lagoon trophic status was apparent from BP/PP (0.05–6.4) and PP/BR (0.10–18.2) ratios. The seasonal shift in net pelagic production from autotrophy to heterotrophy due to terrestrial organic matter inputs via rivers, enhanced the bacterial metabolism during the MN, as evident from the high pCO2 (10,134 µatm) and CO2 air–water flux (714 mm m?2 d?1). Large variability in BGE and BP/PP ratios especially during MN led to high bacteria-mediated carbon fluxes which was evident from significantly high bacterial carbon demand (BCD >100% of PP) during this season. This suggested that the net amount of organic carbon (either dissolved or particulate form) synthesized by primary producers in the lagoon was not sufficient to satisfy the bacterial carbon requirements. Lagoon sustained low to moderate autotrophic–heterotrophic coupling with annual mean BCD of 231% relative to the primary production, which depicted that bacterioplankton are the mainstay of the lagoon biogeochemical cycles and principal players that bring changes in trophic status. Study disclosed that the high CO2 supersaturation and oxygen undersaturation during MN was attributed to the increased heterotrophic respiration (in excess of PP) fuelled by allochthonous organic matter. On a spatial scale, lagoon sectors such as south sector, central sector and outer channel recorded “net autotrophic,” while the northern sector showed “net heterotrophic” throughout the study period. 相似文献

本文旨在探讨母子分离应激对complexin Ⅱ基因缺陷(Cplx2-/-)小鼠快速移动行为的影响。实验分4组:应激Cplx2-/-组、应激Cplx2+/+组、无应激Cplx2-/-组和无应激Cplx2+/+组。无应激组小鼠正常饲养,应激组小鼠从出生后第2日至第21日每天施加母子分离应激。小鼠生长到第4周时,利用聚合酶链反应技术检测其基因型。4组小鼠腹腔注射激动剂去氧麻黄碱或生理盐水后,采用EthoVision系统对小鼠快速移动行为进行记录。结果显示,应激Cplx2-/-组、应激Cplx2+/+组、无应激Cplx2-/-组和无应激Cplx2+/+组注射去氧麻黄碱后,与生理盐水对照组相比,均出现快速移动行为不同程度的增多(P<0.01);与无应激Cplx2-/-组相比较,应激Cplx2-/-组注射去氧麻黄碱后快速移动行为增多更为明显(P<0.001);与应激Cplx2+/+组相比,应激Cplx2-/-组注射去氧麻黄碱后的快速移动行为增多更为明显(P<0.001)。本实验证实小鼠的行为可能受母子分离应激和Cplx2基因等因素的多重影响。 相似文献

The HB4C5 and HF10B4 cell lines are human-human hybridomas producing human IgM monoclonal antibodies (MAbs) reactive to porcine carboxypeptidase A (CPase), but not to double stranded DNA (ds DNA). We obtained G418-resistant HB4C5 and HF10B4 cells by an introduction of pSV2-neo DNA. Almost all of the G418-resistant clones produced MAbs reactive to not only the CPase but the ds DNA. The results of the inhibition ELISA suggested that the cross-reactivity of the antibodies from G418-resistant clones to CPase and ds DNA was responsible for the alteration on their antigen specificity. HB4C5 and HF10B4 cells and their G418-resistant clones produced antibodies having glycosylated chain. The antibodies produced by tunicamycin-treated G418-resistant subclones of HB4C5 and HF10B4 lost the ability to bind to ds DNA, but retained the ability to bind to CPase. These results suggest that an introduction of pSV2-neo DNA into these hybridomas alters the specificities of their MAbs, and that the alteration to antigen binding specificities of their MAbs may be associated with glycosylation of the MAbs by these hybridomas. 相似文献

Two pumilios, pum1 and pum2, were identified in medaka Oryzias latipes. Oryzias latipes pum1 and pum2 are ubiquitous in the adult tissues but with specific expression in the germ cells of gonads, ovary and testis. Pum1 is expressed in the spermatogonia to spermatocytes whilst pum2 presents in spermatocytes of testis only. Oryzias latipes pum1 and pum2 are maternally supplied RNA with ubiquitous expression in the early stages, and embryonic expression of pum1 and pum2 may begin from early gastrula. Both pum1 and pum2 are expressed in the tissues including brain, eye and trunk, and both are expressed in the gonads after hatching.
