Colonisation of extremely acidic waters (pH 3) by aquatic angiosperms occurs widely, but is poorly documented. Unlike acid rain affected and other naturally acidic aquatic ecosystems, waters with pH 3 usually have a high conductivity, with high concentrations of SO4 2- and often high concentrations of Fe3+, other heavy metal ions and Al3+. Where Fe3+ concentration is high, as in many mine waters, it provides a strong buffering system. In such waters, the biogeochemical Fe cycle exerts over water chemistry and the availability of nutrients and carbon for organisms. Biological activity is limited by low concentrations of phosphorus and inorganic carbon (DIC), which in this pH range is essentially all in the form of dissolved CO2. A number of angiosperms grow in such waters including Phragmites australis, Typha spp. and Juncus bulbosus, though the last is the only one reported to grow totally submerged in waters with pH 3 . J. bulbosus occurs in many lignite mining lakes in Lusatia (north eastern Germany) with pH 3. The characteristics and possible survival strategies for this and other species are discussed.  相似文献

Qualitative and quantitative estimates of gene flow were obtained for fourteen gymnosperm and seven angiosperm forest tree species. High levels of gene flow were prevalent among gymnosperms while these levels varied from high to low among angiosperms. In both groups, species with greater pollen dispersal abilities appear to maintain high levels of gene flow. A detailed analysis of population structure in relation to gene flow was carried out on a gymnosperm species (Pinus rigida) and two angiosperm subspecies (Eucalyptus caesia ssp. caesia and ssp. magna). The results suggested that populations of many species may be concatenated systems bound by gene flow, and the overall levels of gene flow may be influenced by either single or clusters of populations. Different levels of gene flow was found between two closely related species of E. caesia growing under similar ecological conditions, suggesting a plausible link between pollinator behaviour and pollen flow.  相似文献

目的:比较酶切信号放大法(Cervista)与导流杂交基因芯片技术(Hybri Ma)检测高危型人乳头状瘤病毒(HR-HPV)诊断宫颈上皮内瘤变2级或2级以上(CIN2+)的临床价值。方法:随机选择288例2012年3月至2013年1月在哈尔滨医科大学附属第一医院妇科门诊进行新柏氏液基细胞学检查的年龄在20~65岁的宫颈细胞学检测未明确意义的不典型鳞状细胞(ASCUS)的患者,采用Cervista技术与Hybri Ma技术进行高危型HPV检测,并对入组的患者行阴道镜下宫颈活组织检查。以病理学诊断结果为金标准,比较Cervista技术与Hybri Ma技术诊断宫颈上皮内瘤变2级或2级以上(CIN2+)的敏感度、特异度及ROC曲线。结果:在入组的288例患者中,Cervista技术和Hybri Ma技术检出高危型HPV的阳性率分别为49.31%和51.39%(P0.05),其诊断CIN2+的敏感度分别为95.65%和91.30%(P0.05),特异度分别为59.50%和56.20%(P0.05),阳性预计值分别为30.99%和28.38%(P0.05),阴性预计值分别为98.63%和97.14%(P0.05)。两组ROC曲线下面积分别为0.776和0.738(P0.05)。结论:Cervista技术与Hybri Ma技术诊断CIN2+的临床价值相当。  相似文献

We use a comprehensive subset of Canarian angiosperms corresponding to 23 families, 35 genera and 60 Canarian endemic taxa to test whether this flora is suitable to taxonomic identification with the two proposed plant DNA barcode sequences and whether these sequences may reveal the existence of cryptic species overlooked by morphology. The rate of discrimination success between the insular congeneric samples using the rbcL+matK combination and a ‘character‐based’ approach (where we use only the combination of nucleotide positions in an alignment that allows unambiguous species identification) is higher (82.29%) than that obtained with the ‘distance‐based’ approach (80.20%) used by the CBOL Plant Working Group in 2009 and also when compared with tests conducted in other floras. This suggests that the molecular identification of the Canarian endemic flora can be achieved as successfully as in other floras where the incidence of radiation is not as relevant. The facts that (i) a distance‐based criterion was unable to discriminate between congeneric and conspecific comparisons and (ii) only the character‐based discrimination criterion resolved cases that the distance‐based criterion did not, further support the use of a character discrimination approach for a more efficient DNA barcoding of floras from oceanic islands like the Canaries. Thus, a barcoding gap seems not to be necessary for the correct molecular characterization of the Canarian flora. DNA barcodes also suggest the possible existence of cryptic taxa to be further investigated by morphology and that the current taxonomic status of some of the taxa analysed may need revision.  相似文献

Neural stem cells (NSCs) are capable of self-renewal and differentiation into neurons, astrocytes and oligodendrocytes under specific local microenvironments. In here, we present a set of methods used for three dimensional (3D) differentiation and miRNA analysis of a clonal human neural stem cell (hNSC) line, currently in clinical trials for stroke disability (NCT01151124 and NCT02117635, Clinicaltrials.gov). HNSCs were derived from an ethical approved first trimester human fetal cortex and conditionally immortalized using retroviral integration of a single copy of the c-mycERTAMconstruct. We describe how to measure axon process outgrowth of hNSCs differentiated on 3D scaffolds and how to quantify associated changes in miRNA expression using PCR array. Furthermore we exemplify computational analysis with the aim of selecting miRNA putative targets. SOX5 and NR4A3 were identified as suitable miRNA putative target of selected significantly down-regulated miRNAs in differentiated hNSC. MiRNA target validation was performed on SOX5 and NR4A3 3’UTRs by dual reporter plasmid transfection and dual luciferase assay.  相似文献

Abstract Caytoniales are an important group of seed plants, and the nature of their female reproductive organ may influence interpretations of the seed plant phylogeny and the origin of angiosperms. Although not convincingly demonstrated by clear evidence, cupules on previously described specimens were interpreted as being distichously arranged, implying that the cupule‐bearing organ in Caytoniales was a pinnate megasporophyll. Here a female reproductive organ of Paracaytonia hongtaoi gen. et sp. nov. (Caytoniales) is reported from Liaoning, China. The well preserved specimen clearly shows a spiral arrangement of cupules along the reproductive axis, suggesting that the cupule‐bearing organ in Caytoniales is not a megasporophyll but a branch. This new information on the axial nature of the cupule‐bearing organ in Caytoniales has significant implications on the placement of Caytoniales in the seed plant phylogeny and interpretation of the relationship between Caytoniales and angiosperms.  相似文献

An important aspect of the evolution of carpel closure, or angiospermy, is the relationship between pollen tube growth patterns and internalization of the pollen‐tube pathway. True carpel closure, involving postgenital fusion of inner carpel margins, is inferred to have arisen once within the ancient order Nymphaeales, in the common ancestor of Nymphaeaceae. We studied pollen tube development, from pollination to fertilization, in a natural population of Nymphaea odorata, using hand pollinations and timed flower collections. Pollen germinates in stigmatic secretions within 15 min and pollen tubes enter subdermal transmitting tissue within an hour, following wide intercellular spaces towards the zone of postgenital fusion. At the zone of fusion they turn downwards to grow in narrow spaces between interlocked cells and then wander freely to ovules within ovarian secretions. The pollen‐tube pathway is 2–6 mm long and upper ovules are first penetrated 2.5 h after pollination. Pollen tubes grow at rates of approximately 1 mm/h whether in stigmatic fluid, transmitting tissues or ovarian secretions. Pollen‐tube pathways are structurally diverse across Nymphaeales, yet their pollen tubes have similar morphologies and rapid growth rates. The nucleotide composition and the contents of CpG and CpNpG in internal transcribed spacers 1 and 2 (ITS1 and ITS2) and the 5.8S rRNA gene of the nuclear genome were studied in two phylogenetic lineages of monocotyledonous angiosperms. The evolutionary advance of taxa by morphological characters proved to positively correlate with an increase in the contents of C, CpG, and CpNpG, contrasting the views that genome evolution in vertebrate and higher plants tends to decrease or, at least, preserve the amount of CpG and CpNpG, potentially subject to methylation, in nuclear DNA. Cryptaffinity taxa, which are intermediates between morphologically distinct taxonomic groups, displayed higher contents of CpG and CpNpG as compared with neighboring taxa. Changes in the contents of these elements in the regions of cryptaffinity taxa are intricate, suggesting a reciprocating character for their accumulation. Cryptaffinity taxa and their close phylogenetic relatives from the ancestral and descendant groups were assumed to reflect the key macroevolutionary changes and to correspond to saltatory periods separating the periods of gradual evolution.  相似文献

中华施迈斯内果在辽西的发现为被子植物的历史添上了重要的一笔。施迈斯内果是一个具有近170年研究历史的植物化石属。新近发现的中华施迈斯内果中发现了以前没被认识到的新特征(未成熟的雌性器官的中央结构有一个纵向的隔壁和封闭的顶端)。新的特征促使人们重新审视该化石的系统位置。王鑫等人通过仔细分析被子植物的定义、结合新的化石特征认为施迈斯内果代表了一个具有被子植物特征的新类群。按照现有的被子植物定义,施迈斯内果可以归入被子植物。这样一来,被子植物的历史就会被前推到早侏罗世。当然也不排除未来被子植物的定义改变后为施迈斯内果建立一个与被子植物平行的种子植物新类群的可能性。文章简要地介绍了施迈斯内果的由来、特征以及王鑫等人如何判定其为被子植物的逻辑思维过程,以便大家论评。  相似文献
