Summary Horizontal cells of rat retina were labeled intensely by a specific antibody to cerebellar calcium-binding protein. The amacrine cells stained very weakly. Abstract. In controlled environments, the interactive effects of warm (16: 8°C, day: night) and cool (12: 4°C, day: night) temperatures and long (13.5 h) and short (10 h) photoperiods on the dehardening of seedlings of Pinus radiata D. Don were investigated. In another experiment, the effect of four photoperiods from 9 to 14 h was examined. In a third, dehardening at constant temperatures from 5 to 17°C was followed. There was no evidence for an interaction between photoperiod and temperature. Dehardening was temporarily delayed by photoperiods below about 10 h, but there was no other quantitative effect of photoperiod. At constant temperatures, the rate of dehardening was initially constant but declined as the minimum summer frost hardiness was reached. In the initial phase the rate of dehardening was a linear function of temperature, increasing from 0.05°C day−1 at 8°C to 0.30 °C day−1 at 17°C. Temperature controlled the loss of frost hardiness by regulating the rate of dehardening.  相似文献

Summary Highly sensitive and specific synthetic substrates were used to quantitate cathepsin B and D activity in peritoneal macrophages in response to stimulation in vivo with mineral oil and thioglycollate. After intraperitoneal instillation of mineral oil the activity of cathepsin B increased significantly (to 15 300 units/mg protein versus 7 340 in saline controls), reaching values approaching those found in alveolar macrophages (18 400 units/mg protein). Significantly greater stimulation of enzyme activity was obtained after intraperitoneal instillation of thioglycollate (23 600 units/mg protein). Cathepsin D activity also increased significantly after both mineral oil and thioglycollate. However, the increase was moderate (from 806 to about 1 200 units/mg protein), remaining still more than six times lower-than in alveolar macrophages. The data are the first to demonstrate that cathepsin B activity can be stimulated in vivo in peritoneal macrophages by instillation of agents that induce acute inflammation. 本文报道蓖麻蚕蛹在室温28℃下的卵巢发育过程,以及外源20-羟基蜕皮酮对蚕蛹发育的影响。正常蛹在任何发育期内注射20-羟基蜕皮酮后,全部仍羽化成蛾,但蛹期延长约1至4天。无脑蛹经注射后出现蛹——蛾的变态,发育情况因剂量而不同:注射0.1微克后约有半数蛹发育成蛾;注射2微克羽化率较高,卵巢管的发育也最好;4微克或更高的注射量能使全部蛹发育成蛾,但卵巢管多少有些不正常。注射量超过5微克时,蛾体较小,颜色浅黄,没有或只有很少的鳞片。蛹的发育天数随剂量的增大而减少。经外源20-羟基蜕皮酮处理后,无论是有脑蛾或是无脑蛾的卵粒都明显地比正常蛾的卵粒大。当超过一定的注射量时,注射量越大,蚕蛾的自动蜕壳能力越差。  相似文献

棉株根系伤流有明显昼夜节奏,白天多,夜晚较少。白天中又以上午9~12时的伤流量为最高。伤流中CTK量及其浓度也以白天为高。在切除地上部4天以后,从蕾期和铃期棉株已不再能收集到伤流液,但盛花期棉株在切除地上部6天以后,仍产生了相当多的伤流,其中仍含有丰富的CTK类物质。盛花期棉株根系伤流量及其中CTK水平高于铃期棉株,提示铃期棉株根系活力已开始趋向衰老。根据Sephadex LH-20柱层析及高效液相层析鉴定出棉株根系伤流中的CTK类物质有Z,ZR,和IPA。  相似文献

5-methoxypsoralen (5-MOP) binds to human serum low density lipoproteins (LDL) according to a two-step process. Scatchard analysis of the first step yields K = 1.4 × 105 M?1 and 4 binding sites. It involves the LDL apoprotein. The second step corresponds to a solubilization, in the lipidic core, of ? 45 molecules of 5MOP per LDL molecule. It is accompanied by a large blue shift of the 5MOP fluorescence. The ability of LDL to bind 5MOP and to carry it into various cells may explain some biological effects sometimes encountered during PUVA therapy.  相似文献

To further analyze functionally important cholinergic receptors on lymphocytes, we studied the binding of the muscarinic antagonist Quinuclidinyl benzilate (QNB) to murine splenic lymphocytes. Studies of displacement of [3H]QNB by unlabelled QNB on lymphocytes revealed at least two binding sites. Scatchard analysis of equilibrium binding isotherms also distinguished two sites with apparent Kds of 480 nM and 16 μM. There was greater specific QNB binding to B cell-enriched lymphocyte fractions than to T cell fractions. Lymphocyte binding demonstrated temperature-dependent dissociability, and specific binding occurred on isolated lymphocyte membranes as well. Both muscarinic and nicotinic ligands competed for QNB binding to lymphocytes with low and nearly equal affinity. Therefore, QNB binding sites on lymphocytes appear to be of low affinity and of mixed muscarinic and nicotinic character.  相似文献
