利用G-CSF依赖型细胞株N8F-60,尝试将MTT比色法应用于基因工程产品重组人粒系集落刺激因子(rhG-CSF)的活性检测。多次实验探索出最适的细胞浓度和培养时间,细胞数为低于1×10 ̄4/孔,培养时间为30─48h。在宽的线性范围内检测G-CSF样品的生物活性,结果经传统的CFU-G方法验证可靠,且较后者更灵敏、简便、省时、省力,并可定性、定量地检测样品活性。特别适用于大量样品的同时检测。另外,此方法可避免接触放射性。 相似文献

A human myeloid leukemia cell line [HL-60] could be induced to differentiate into mature myeloid cells by 1α,25-dihydroxyvitamin D3 [1α,25(OH)2D3], the active form of vitamin D3. At 10?10–10?8 M, 1α,25(OH)2D3 suppressed cell growth in a dose-dependent manner and markedly induced phagocytosis and C3 rosette formation. The potency of 1α,25(OH)2D3 in inducing differentiation was nearly equivalent to that of known synthetic inducers such as dimethyl sulfoxide, actinomycin D or a phorbol ester (12-o-tetra-decanoyl-phorbol-13-acetate). These results clearly indicate that 1α,25(OH)2D3, besides its well known biological effect in enhancing intestinal calcium transport and bone mineral mobilization activities, is involved in the cell grwoth and differentiation of HL-60 cells. 相似文献

Some organisms, such as zebrafish, urodele amphibians, and newborn mice, have a capacity for heart regeneration following injury. However, adult mammals fail to regenerate their hearts. To know why newborn mice can regenerate their hearts, we focused on epigenetic factors, which are involved in cell differentiation in many tissues. Baf60c (BRG1/BRM‐associated factor 60c), a component of ATP‐dependent chromatin‐remodeling complexes, has an essential role for cardiomyocyte differentiation at the early heart development. To address the function of Baf60c in postnatal heart homeostasis and regeneration, we examined the detailed expression/localization patterns of Baf60c in both mice and axolotls. In the mouse heart development, Baf60c was highly expressed in the entire heart at the early stages, but gradually downregulated at the postnatal stages. During heart regeneration in neonatal mice and axolotls, Baf60c expression was strongly upregulated after resection. Interestingly, the timing of Baf60c upregulation after resection was consistent with the temporal dynamics of cardiomyocyte proliferation. Moreover, knockdown of Baf60c downregulated proliferation of neonatal mouse cardiomyocytes. These data suggested that Baf60c plays an important role in cardiomyocyte proliferation in heart development and regeneration. This is the first study indicating that Baf60c contributes to the heart regeneration in vertebrates. 相似文献

This report describes an outbreak of suppurative peritonitis caused by Klebsiella pneumoniae in an adult female of captive golden‐handed tamarin (Saguinus midas midas). Two virulent and multidrug‐resistant strains were isolated and classified through MLST as ST60 and ST1263. The microbiological diagnosis works as a support tool for preventive measures. 相似文献

The identification and characterisation of Cryptosporidiumgenotypes
and subtypes are fundamental to the study of cryptosporidiosis epidemiology, aiding
in prevention and control strategies. The objective was to determine the genetic
diversity ofCryptosporidium in samples obtained from hospitals of
Rio de Janeiro, Brazil, and Buenos Aires, Argentina. Samples were analysed by
microscopy and TaqMan polymerase chain reaction (PCR) assays
forCryptosporidium detection, genotyped by nested-PCR-restriction
fragment length polymorphism (RFLP) analysis of the 18S rRNA gene and subtyped by DNA
sequencing of the gp60 gene. Among the 89 samples from Rio de
Janeiro, Cryptosporidium spp were detected in 26 by
microscopy/TaqMan PCR. In samples from Buenos Aires,Cryptosporidium
was diagnosed in 15 patients of the 132 studied. The TaqMan PCR and the
nested-PCR-RFLP detected Cryptosporidium parvum,
Cryptosporidium hominis, and co-infections of both species. In
Brazilian samples, the subtypes IbA10G2 and IIcA5G3 were observed. The subtypes found
in Argentinean samples were IbA10G2, IaA10G1R4, IaA11G1R4, and IeA11G3T3, and mixed
subtypes of Ia and IIa families were detected in the co-infections. C.
hominis was the species more frequently detected, and subtype family Ib
was reported in both countries. Subtype diversity was higher in Buenos Aires than in

Heat shock proteins (Hsps) are a class of highly conserved proteins produced in virtually all living organisms from bacteria to humans. Hsp60 and Hsp10, the most important mitochondrial chaperones, participate in environmental stress responses. In this study, the full-length complementary DNAs (cDNAs) of Hsp60 (PmHsp60) and Hsp10 (PmHsp10) were cloned from Penaeus monodon. Sequence analysis showed that PmHsp60 and PmHsp10 encoded polypeptides of 578 and 102 amino acids, respectively. The expression profiles of PmHsp60 and PmHsp10 were detected in the gills and hepatopancreas of the shrimps under pH challenge, osmotic stress, and heavy metal exposure, and results suggested that PmHsp60 and PmHsp10 were involved in the responses to these stimuli. ATPase and chaperone activity assay indicated that PmHsp60 could slow down protein denaturation and that Hsp60/Hsp10 may be combined to produce a chaperone complex with effective chaperone and ATPase activities. Overall, this study provides useful information to help further understand the functional mechanisms of the environmental stress responses of Hsp60 and Hsp10 in shrimp. 相似文献

The present study was addressed to understand the interrelationship between Receptor-Ck induction-activation coupling; isoprenoids (derived from mevalonate pathway) and cyclin-dependent kinase inhibitors. The results reported here unambiguously reveal that isoprenoids regulate the expression of genes coding for CDK inhibited p16 and p27. Further, Receptor-Ck dependent signalling, known to control the mevalonate pathway, had a direct effect upon the p27 gene expression. Based upon these studies coupled with our earlier findings we propose that Receptor-Ck has an important role in the regulation of cell cycle machinery. 相似文献

Mimosine, a plant amino acid not found in proteins, has been widely used as a synchronizing agent, blocking the progression of cell cycle on the G1/S phase border. The mechanism by which this block is achieved is still unclear. We report that in HL60 cells the synchronization is related to an increase in apoptosis. Another human tumor cell line, K562, is insensitive to both phenomena thereby demonstrating that apoptosis observed in HL60 is line-specific. We hypothesize that the mimosine-induced apoptosis and alteration of the cell cycle is due to the inhibition of hypusine generation. 相似文献

The present study is on the growth inhibitory effect of Withania somnifera methanolic leaf extract and its active component, withanolide on HL-60 promyelocytic leukemia cells. The decrease in survival rate of HL-60 cells was noted to be associated with a time dependent decrease in the Bcl-2/Bax ratio, leading to up regulation of Bax. Both the crude leaf extract and the active component activated the apoptotic cascade through the cytochrome c release from mitochondria. The activation of caspase 9, caspase 8 and caspase 3 revealed that caspase was a key mediator in the apoptotic pathway. DNA fragmentation analysis revealed typical ladders as early as 12h indicative of caspase 3 role in the apoptotic pathway. Flow cytometry data demonstrated an increase of sub-G1 peak upon treatment by 51% at 24h, suggesting the induction of apoptotic cell death in HL-60 cells. 相似文献
