This study characterizes the colonization and composition of bacterial flora in dwarf Asian honeybee (Apis florea) larvae and compares bacterial diversity and distribution among different sampling locations. A. florea larvae were collected from 3 locations in Chiang Mai province, Thailand. Bacterial DNA was extracted from each larva using the phenol–chloroform method. Denaturing gradient gel electrophoresis was performed, and the dominant bands were excised from the gels, cloned, and sequenced for bacterial species identification. The result revealed similarities of bacterial community profiles in each individual colony, but differences between colonies from the same and different locations. A. florea larvae harbor bacteria belonging to 2 phyla (Firmicutes and Proteobacteria), 5 classes (Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, Bacilli, and Clostridia), 6 genera (Clostridium, Gilliamella, Melissococcus, Lactobacillus, Saccharibacter, and Snodgrassella), and an unknown genus from uncultured bacterial species. The classes with the highest abundance of bacteria were Alphaproteobacteria (34%), Bacilli (25%), Betaproteobacteria (11%), Gammaproteobacteria (10%), and Clostridia (8%), respectively. Similarly, uncultured bacterial species were identified (12%). Environmental bacterial species, such as Saccharibacter floricola, were also found. This is the first study in which sequences closely related to Melissococcus plutonius, the causal pathogen responsible for European foulbrood, have been identified in Thai A. florea larvae.  相似文献

We designed a single nucleotide primer extension (SNaPshot) assay for Pneumocystis jirovecii genotyping, targeting mt85 SNP of the mitochondrial large subunit ribosomal RNA locus, to improve minority allele detection. We then analyzed 133 consecutive bronchoalveolar lavage (BAL) fluids tested positive for P. jirovecii DNA by quantitative real‐time PCR, obtained from two hospitals in different locations (Hospital 1 [= 95] and Hospital 2 [= 38]). We detected three different alleles, either singly (mt85C: 39.1%; mt85T: 24.1%; mt85A: 9.8%) or together (27%), and an association between P. jirovecii mt85 genotype and the patient's place of hospitalization (= 0.011). The lowest fungal loads (median = 0.82 × 103 copies/μl; range: 15–11 × 103) were associated with mt85A and the highest (median = 1.4 × 106 copies/μl; range: 17 × 103–1.3 × 107) with mt85CTA (= 0.010). The ratios of the various alleles differed between the 36 mixed‐genotype samples. In tests of serial BALs (median: 20 d; range 4–525) from six patients with mixed genotypes, allele ratio changes were observed five times and genotype replacement once. Therefore, allele ratio changes seem more frequent than genotype replacement when using a SNaPshot assay more sensitive for detecting minority alleles than Sanger sequencing. Moreover, because microscopy detects only high fungal loads, the selection of microscopy‐positive samples may miss genotypes associated with low loads.  相似文献

2017年10月,在贵州省福泉县发现了桃花水母(Craspedacusta)。观察其形态结构,福泉的桃花水母与索氏桃花水母(C. sowerbyi)高度相似。采用PCR和DNA测序技术扩增和测定了福泉采集桃花水母的核糖体小亚基rRNA基因(18SrRNA)、核糖体RNA基因内转录间隔区(ITS)及线粒体细胞色素氧化酶亚基基因(COI),并与GenBank上已有的桃花水母18S rRNA、ITS、COI基因序列进行了比对。分子鉴定结果显示,福泉的桃花水母18SrRNA、ITS、COI基因序列与索氏桃花水母相似度分别为100%、92%、99%,确定贵州省福泉市发现的桃花水母样品在种分类水平上为索氏桃花水母。并就桃花水母的某些形态学分类指标的标准、不同伞径之间的差值是否能作为桃花水母形态分类的一个新指标这些问题做了讨论。  相似文献

目的 探索用有机溶剂/去污剂(solvent/detergent, S/D)病毒灭活处理对破伤风抗毒素质量的影响。方法 3批破伤风抗毒素样品,每批样品取3等份,向其中2份中分别加入磷酸三丁酯(tri- n -butylphosphate, TNBP)和吐温-80(Tween 80)至终质量分数为0.3%和1%;一份放置在(25±1)℃水浴中振摇6 h后取样,另一份放置在(30±1)℃水浴中振摇4 h后取样;向第3份破伤风抗毒素原液中加入等量的生理盐水混匀后室温放置作为对照。各样品经超滤后检测其效价和蛋白质含量,同时用SDS-PAGE电泳和凝胶过滤色谱柱测定。结果 破伤风抗毒素原液样品经过S/D病毒灭活处理后,其动物效价与对照相比差异无统计学意义( P >0.05),蛋白质含量、分子大小分布无明显变化,多聚体、二聚体及F(ab′) 2含量也无明显变化。结论 S/D病毒灭活处理对破伤风抗毒素的效价,蛋白质含量,分子大小分布,多聚体、二聚体及F(ab′) 2含量均无明显影响,可以作为破伤风抗毒素病毒灭活的候选方法。  相似文献

Fatty acid composition, especially oleic acid (C18:1), plays an important role in the eating quality of meat in Japanese Black cattle. Therefore, the objective of this study was to identify loci associated with C18:1 in the intramuscular fat of the trapezius muscles in Japanese Black cattle using the Illumina BovineSNP50 BeadChip whole genome single nucleotide polymorphism (SNP) assay. We also evaluated the relationship between C18:1 and three fatty acid synthesis genes, fatty acid synthase (FASN), stearoyl‐CoA desaturase and sterol regulatory element‐binding protein‐1. In this experiment, we applied a mixed model and Genomic Control approach using selective genotyping to perform a genome‐wide association study. A total of 160 animals (80 animals with higher values and 80 animals with lower values), selected from 3356 animals based on corrected phenotype, were genotyped using the Illumina BovineSNP50 BeadChip and three fatty acid synthesis genes, and the quality of these SNPs was assessed. In this study, a total of 38 955 SNPs, which included SNPs in the three fatty acid synthesis genes, were used, and the estimated inflation factor was 1.06. In the studied population, a total of 32 SNPs, including the FASN gene, had significant effects, and in particular 30 SNPs of all significant SNPs were located between 49 and 55 Mbp on chromosome 19. This study is one of the first genome‐wide association studies for fatty acid composition in a cattle population using the recently released Illumina BovineSNP50 BeadChip.  相似文献

以烟草赤星病菌为供试病原菌,采用对峙法、代谢液培养法对分离自大豆根瘤的内生细菌进行抑菌筛选,对筛选菌株作用的病原菌菌丝进行显微观察,研究筛选菌株细胞培养特征及生理生化特性、16S rDNA测序序列、系统发育,以及进行温室接种防病试验.结果表明: 大豆根瘤内生菌经初筛、复筛,对烟草赤星病菌抑菌率42%以上的有7株菌,分别属于芽孢杆菌属、假单胞菌属、中华根瘤菌属和寡养单胞菌属.显微观察表明,在拮抗性内生菌作用下病原菌菌丝末端出现明显畸形,呈珊瑚状分枝、球状膨大等;代谢液试验表明,内生菌对病原菌的抑制作用主要是内生菌代谢产生的胞外物质在发挥作用;对峙试验表明,芽孢杆菌可迅速形成生物薄膜,有效阻碍病原菌丝生长和延伸.盆栽防病试验表明,拮抗性内生菌处理下烟草病情指数显著低于对照,表明筛选菌株可作为烟草赤星病的生防菌株资源.  相似文献

Microbial communities are potential indicators for water quality as they respond rapidly to environmental changes. In the Whitsunday Islands, Australia, microbial biofilm communities from two offshore islands were compared to those from two inshore islands subjected to poor water quality. Biofilm community composition was characterized using three culture-independent molecular techniques. The clone libraries indicated high genetic diversity, with somewhat higher scores in the offshore sites (57%) compared to the inshore sites (41%). The majority of microbes in the biofilms were related to Alphaproteobacteria (39.8%), Gammaproteobacteria (14.1%), Bacteroidetes (13.2%), diatoms (8.3%) and Cyanobacteria (3.9%). Redundancy analysis (RDA) for the CARD-FISH data showed distinct microbial assemblages between offshore and inshore communities. Additionally, 5 out of 13 water quality parameters (DIN, Chla, POP, TSS and POC) explained a significant amount of variation in the microbial communities and high values of these were associated with inshore communities. Analysis of variance (ANOVA) indicated that Cyanobacteria (p = 0.01), Bacteroidetes (p = 0.04) and to some extent Alphaproteobacteria (p = 0.07), were significantly more abundant in the offshore biofilm communities. Principal Component Analysis (PCA) of DGGE data showed clear grouping of cyanobacterial communities into inshore and offshore communities. Reasons for community shifts in the bacterial lineages are currently not resolved. One possible causative factor may be that autotrophic primary producers are more dominant in offshore sites due to the higher light availability as well as the limitation by DIN. The trends found in this study are the bases for more detailed research on microbial indicator species for changes in water quality.  相似文献

Background

HbF-Monserrato-Sassari is a newly discovered abnormal fetal hemoglobin observed in an apparently normal newborn baby during a hemoglobinopathies survey at birth in North Sardinian population.

Methods

Electrophoretic analysis of the cord blood lysate evidenced for an abnormal tetramer due to a mutated fetal globin chain. Electrospray ionisation-mass spectrometry and gene sequencing were used to identify the mutation. Oxygen binding ability of the variant Hb was determined.

Results

Sequencing of the γ globin genes revealed the TGT → CGT transition at codon 93 in one of the two Gγ genes, which leads to the Arg for Cys amino acid replacement at position 9 of the F α-helix. The amino acid substitution was confirmed by mass spectrometric analysis of the globin chains. Since modifications or substitutions at position β93 are known to affect the arrangement of a salt bridge at the α1β2 sliding contacts that are crucial for subunit cooperativity, the functional properties of the variant were studied to evaluate the effect of the replacement at the same position in the γ globin chain. With respect to normal HbF, the variant showed a significant increase in oxygen affinity and a slight decrease of both Bohr effect and cooperativity.

General significance

Result indicates a key role of the Cys γ93 residue for subunit cooperativity in the T → R transition of the HbF tetramer. Substitutions at the F9 position of the Gγ globin may result in stabilization of the high affinity R-state of the Hb tetramer. Because of the loss of Cys γ93 residue, this variant is considered to be potentially compromised in nitric oxide transport.  相似文献

利用28S rDNA D1部分基因序列对直突摇蚊亚科代表性属级阶元进行了分子系统学研究。测定了12个内群属和2种外群的28SrDNAD1片段,并结合GenBank中3个同亚科种类该基因的同源序列进行了分析。采用2种建树方法(距离邻近法NJ和最大俭约法MP)分析了直突摇蚊亚科内属级分类单元的分子系统发育关系。结果表明,滨海摇蚊属Clunio位于系统发育树的基部,与该属营海洋生活的特殊性一致。心突摇蚊属和真开氏摇蚊属互为姐妹群,流环足摇蚊属和刀突摇蚊属互为姐妹群,此结果与基于形态学的系统发育研究相结果一致。其它属间的系统发育关系因尚无前人研究而有待做进一步研究。本研究同时证明28S rDNA D1基因片段在分析摇蚊科昆虫属级及属内阶元关系上具有一定的指导意义。  相似文献

从活性污泥中分离筛选得到一株能代谢甘油生产1,3-丙二醇(1,3-PD)的菌株2-1,通过形态学鉴定、生理生化试验、16S rRNA序列分析对菌株分类学地位进行鉴定,用MEGA 4.1软件构建的系统发育树显示菌株2-1与Klebsiella pneumoniae(CP001891)的亲缘关系最近。16S rDNA序列同源性比较发现,菌株2-1与模式菌株同源率为95.4%,疑似为新种。对菌株2-1在5 L发酵罐中进行发酵特性研究,分批补料发酵时得到较高的1,3-PD终浓度,达到63.5 g/L,此时生产强度为2.19 g/(L.h),底物转化率0.64 mol/mol。  相似文献
