The effects of quinine and 9-aminoacridine, two blockers of potassium conductance in islet cells, on 45Ca efflux and insulin release from perifused islets were investigated in order to elucidate the mechanisms by which glucose initially reduces 45Ca efflux and later stimulates calcium inflow in islet cells. In the absence of glucose, 100 μM quinine stimulated 45Ca net uptake, 45Ca outflow rate and insulin release. Quinine also dramatically enhanced the cationic and the secretory response to intermediate concentrations of glucose, but had little effect on 45Ca net uptake, 45Ca fractional outflow rate and insulin release at a high glucose concentration (16.7 mM). The ability of quinine to stimulate 45Ca efflux depended on the presence of extracellular calcium, suggesting that it reflects a stimulation of calcium entry in the islet cells. In the absence of extracellular calcium, quinine provoked a sustained decrease in 45Ca efflux. Such an inhibitory effect was not additive to that of glucose, and was reduced at low extracellular Na+ concentration. At a low concentration (5 μM), quinine, although reducing 86Rb efflux from the islets to the same extent as a non-insulinotropic glucose concentration (4.4 mM), failed to inhibit 45Ca efflux. In the presence of extracellular calcium, 9-aminoacridine produced an important but transient increase in 45Ca outflow rate and insulin release from islets perifused in the absence of glucose. In the absence of extracellular calcium, 9-aminoacridine, however, failed to reduced 45Ca efflux from perifused islets. It is concluded that quinine, by reducing K+ conductance, reproduces the effect of glucose to activate voltage-sensitive calcium channels and to stimulate the entry of calcium into the B-cell. However, the glucose-induced inhibition of calcium outflow rate, which may also participate in the intracellular accumulation of calcium, does not appear to be mediated by changes in K+ conductance.  相似文献

Anthocyanins, variously identified in inflorescence, fruit, leaf or petiole of 59 representative species of the Araccae, are of a simple type. The most common pigment is cyanidin 3-rutinoside, while pelargonidin 3-rutinoside and cyanidin 3-glucoside are regularly present. Two rare pigments are: cyanidin 3-gentiobioside in Anchomanes and Rhektophyllum, both in the subfamily Lasioideae; and delphinidin 3-rutinoside in Schismatoglottis concinna. In a leaf survey of 144 species from 58 genera, flavone C-glycosides (in 82%) and proanthocyanidins (in 35–45%) were found as the major flavonoids. In the subfamily Calloideae, subtribe Symplocarpeae, flavonols replace glycoflavones as the major leaf components but otherwise flavonols are uncommon in the family (in 27% of the sample) and more usually co-occur with flavone C-glycosides. Two new flavonol glycosides were characterized from Lysichiton camtschatcense: kaempferol 3-(6-arabinosylgalactoside)and kaempferol 3-xylosylgalactoside. Simple flavones, luteolin and chrysoeriol (in 6%) were found only in the subtribes Arinae and Cryptocoryninae, subfamily Aroideae. Flavonoid sulphates were identified in only four taxa: glycoflavone sulphates in two Culcasia species and Philodendron ornatum and a mixture of flavone and flavonol sulphates in Scindapsus pictus. Caffeic ester sulphates were more common and their presence in Anthurium hookeri was confirmed. These results show that the Araceae are unusual amongst the monocots in their simple and relatively uniform flavonoid profile; no one subfamily is clearly distinguished, although at tribal level some significant taxonomic patterns are observed. The best defined groups are the subfamilies Lasioideae and Monsteroideae, and the tribes Symplocarpeae and Arophyteae, and the subtribe Arinae. The greatest chemical diversity occurs in Anthurium and Philodendron, but this may only reflect the fact that these are the two largest genera in the family. The origin and relationship of the Araccae to other monocot groups are discussed in the light of the flavonoid evidence.  相似文献

目的:研究2型糖尿病患者Pentraxin-3和尿蛋白的相关性,以及糖尿病肾病患者中PTX3临床上所起的作用.方法:选取我院2010年4月-2012年6月间进行诊治的230例2型糖尿病患者,通过对白蛋白的排泄率进行测定,将其分为三组,分别是正常白蛋白尿组、微量白蛋白尿组以及临床白蛋白尿组,分别对三组患者的血糖、血脂、γ-谷氨酰转移酶、PTX3以及HS-CRP进行测量.结果:对PTX3水平测量时,微量白蛋白尿组明显比正常白蛋白尿组高(P<0.01),临床白蛋白尿组PTX3水平显著高于其它两组(P<0.01),PTX3与UAER呈正相关.各组患者的年龄、TG、HS-CRP、FPG、GGT比较,差异显著有统计学意义(P>0.05).结论:患者白蛋白尿和PTX3关系密切,因此测定PTX3的水平有助于早期对糖尿病肾病进行诊断,PTX3的水平尤其对2型糖尿病肾病患者是一个重要的衡量因素.  相似文献

目的: 探讨糖原合成酶激酶-3(GSK3β)/真核延伸因子激酶2(eEF2K)信号通路对肺纤维化进程的影响,为临床治疗肺纤维化寻找新的思路。方法: 采用一次性气管注射法构建C57BL/6雄性小鼠博莱霉素肺纤维化模型,造模14 d后将动物分成模型组、阴性抑制组与抑制组(n=5),另设空白组不作处理。抑制组使用腹腔注射TDZD-8(4 mg/kg),阴性抑制组腹腔注射二甲基亚砜(DMSO)溶液,28 d后处死采集指标。采用苏木精-伊红染色法检测小鼠肺脏病变情况;试剂盒水解法检测肺组织中羟脯氨酸(Hyp)的含量;采用Western blot法检测肺脏中GSK3β、磷酸化GSK3β(p-GSK3β)、eEF2K、p-eEF2K(Ser70)、p-eEF2K(Ser392)、p-eEF2K(Ser470)、基质金属蛋白酶-2前体蛋白(pro-MMP-2)、基质金属蛋白酶-2(MMP-2)蛋白表达水平,使用免疫组织化学法检测肺脏中MMP-2、胶原蛋白I(Col I)、胶原蛋白Ⅲ(Col Ⅲ)、α-平滑肌蛋白(α-SMA)的表达。结果: 与空白组相比,模型组中GSK3β、p-GSK3β、p-eEF2K(Ser70)、p-eEF2K(Ser392)、p-eEF2K(Ser470)、pro-MMP-2、MMP-2、Col I、Col Ⅲ、α-SMA蛋白表达水平升高,eEF2K蛋白表达水平降低(P<0.05);与模型组相比,抑制组GSK3β、p-GSK3β、p-eEF2K(Ser70)、p-eEF2K(Ser392)、p-eEF2K(Ser470)、pro-MMP-2、MMP-2、Col I、Col Ⅲ、α-SMA蛋白表达降低,eEF2K蛋白表达升高(P< 0.05)。结论: GSK3β能通过Ser70、Ser392、Ser470这3个位点磷酸化激活eEF2K,增加纤维化指标含量,促进肺纤维化形成,加重肺组织病变。  相似文献

为了探讨沙漠中固沙灌木种群共存和演替机制, 本文基于古尔班通古特沙漠东南缘固定沙丘上白梭梭(Haloxylon persicum)和梭梭(H. ammodendron)种群的地理位置和生长发育阶段信息(幼株、营养株、生殖株和死株), 采用点格局分析方法(g(r)函数)及Monte-Carlo随机模拟检验和零模型选取的方法, 分析了固沙灌木白梭梭和梭梭种群不同生长发育阶段在0-20 m尺度内的空间分布格局及种间关联性。结果表明: (1)两个种群在研究尺度范围内呈聚集分布, 随着尺度的增大, 其聚集强度逐渐减弱; (2)两个种群整体上呈负关联关系, 尺度越大负关联关系越显著; (3)白梭梭种群生长发育阶段相差越大, 个体间正关联关系越显著; 梭梭种群生长发育阶段越接近, 个体间正关联关系越显著; (4)两个种群中龄级较大的个体(如营养株、生殖株和死株)会对对方种群中龄级较小的幼株产生一定的抑制作用; 同时, 随着两个种群中个体的成长, 双方受到的抑制作用逐渐减弱, 主要表现为正关联和无关联。总体而言, 古尔班通古特沙漠固定沙丘白梭梭和梭梭种群的分布格局整体上为聚集分布, 随龄级增加聚集性减弱, 受生境异质性和扩散限制的影响明显。种间关系多为负相关, 种内不同生长发育阶段之间均为正关联关系  相似文献

Cytochrome c oxidase (COX) or complex IV of the mitochondrial respiratory chain plays a fundamental role in energy production of aerobic cells. In humans, COX deficiency is the most frequent cause of mitochondrial encephalomyopathies. Human COX is composed of 13 subunits of dual genetic origin, whose assembly requires an increasing number of nuclear-encoded accessory proteins known as assembly factors. Here, we have identified and characterized human CCDC56, an 11.7-kDa mitochondrial transmembrane protein, as a new factor essential for COX biogenesis. CCDC56 shares sequence similarity with the yeast COX assembly factor Coa3 and was termed hCOA3. hCOA3-silenced cells display a severe COX functional alteration owing to a decreased stability of newly synthesized COX1 and an impairment in the holoenzyme assembly process. We show that hCOA3 physically interacts with both the mitochondrial translation machinery and COX structural subunits. We conclude that hCOA3 stabilizes COX1 co-translationally and promotes its assembly with COX partner subunits. Finally, our results identify hCOA3 as a new candidate when screening for genes responsible for mitochondrial diseases associated with COX deficiency.  相似文献

The effects of experimental parameters including soil type, curing duration, inoculum size, and biomass and nutrients concentration on soil strengthening due to calcite precipitation by Sporosarcina pasteurii PTCC 1645 were investigated. The laboratory-scale mixing experiments on remolded samples were designed by the Taguchi method. Soil type proved to be the most incorporating factor, followed by curing time and nutrient concentration. The main effect and the interactions of the parameters were presented and the optimal conditions were obtained. This suggests the importance of local conditions including soil type on any future large-scale, in situ application.  相似文献
