The gastropod fauna of the Upper Devonian Baggy and Pilton formations in south‐west England is revised and includes some 30 taxa. The topmost part of the Upper Famennian succession in Devon is represented by clastic near‐shore and shallow shelf sediments, indicating a short‐term transgressive phase (‘Strunian Transgression’). The sequence yields a highly diverse fauna dominated by brachiopods and ostracodes, locally supplemented by crinoids, bryozoans, trilobites and molluscs. The taxa ‘Patellostium’britannicum sp. nov., Angyomphalus (Angyomphalus) junius sp. nov. and Dictyotomaria eurocapillaria sp. nov. are erected; a junior homonym is replaced by Macrochilina? piltonensis nom. nov. The gastropod fauna displays an independent character, where latest Devonian faunal elements overlap with Late Palaeozoic taxa expressing a transition similar to that of the bivalves, brachiopods, echinoderms and corals, without a sharp faunal break at the Devonian/Carboniferous boundary. Apart from the Caenogastropoda, all subclasses of gastropods are represented. Members of the bellerophontoids, pleurotomarioids and loxonematoids are most abundant, followed by murchisonioids, naticimorphs, euomphalomorphs and platyceratoids. The various gastropod groups represent different ecological demands and trophic categories, and together with the accompanying fauna indicate that nearly all habitats and niches were occupied in the shallow South Laurussian Shelf. 相似文献

Three features of chloroplast DNA (cpDNA) in plastids isolated from Acetabularia mediterranea (acetabulum) were analyzed after staining the organelles with the fluorochrome 4′6-diamidino-2-phenyl indole (DAPI): (1) number of chloroplasts exhibiting DNA fluorescence, (2) number of nucleoids per plastid, and (3) nucleoid morphology. In vegetative Acetabularia cells only half of the total chloroplast population comprising several millions displayed the whitish-blue fluorescence of the DNA/DAPI complex. This percentage remained stable independent of whether cells were grown in supplemented natural sea water or enriched synthetic sea water. A single nucleoid, widely differing in size and morphology among the organelles, was characteristic of 76–81% of chloroplasts with DNA. Less than 20% contained two nucleoids, and in rare cases three or four nucleoids were present. The pattern of nucleoid numbers followed a Poisson distribution in one experiment, if calculated with the intrinsic mean of the observed data. In two other experiments, however, a significant difference existed between observed and expected values for a Poisson distribution according to the Chisquared test. After secondary enlargement of portions of the negatives, the nucleoids’substructure was disclosed and found to consist of brightly fluorescent spots interspersed by unstained regions The lack of cpDNA in Acetabularia cells appears to be brought about by (1) the polarized pattern of growth and translation confined to the apical region of the single cell and (2) the cpDNA arrangement in a single nucleoid acentrically located in the organelle. A scheme for the evolution of a chloroplast population having plastids without DNA is proposed. In theory the lack of cpDNA could arise in each plant, since chloroplasts never evolved a mitotic-like spindle to ensure the equal distribution of genetic material. Different CD95 (Fas/APO-1) isoforms and phosphory lated CD95 species were identified in human T and B cell lines. We had shown
previously that the CD95 intracellular domain (IC), expressed as a glutathione S-transferase (GST) fusion protein in murine
L929 fibroblasts, was phosphorylatedin vivo. GST-CD95IC was phosphorylatedin vitro by a kinase present in extracts from the human lymphocytic cell lines Jurkat and MP-1 and from murine L929 cells. Phosphoamino
acid analysis indicated that phosphorylation occurred at multiple threonine residues and also at tyrosine (Tyr232 and Tyr291)
and serine. Amino acids 191 to 275 of CD95 were sufficient for phosphorylation at threonine, tyrosine and serine and also
mediated interaction with a 35 kDa cellular protein. Immuno-precipitation of CD95 and chemical cross-linking revealed CD95-associated
proteins of approximately 35, 45 and 75 kDa. GST-CD95IC affinity chromatography detected binding of the 35 and 75 kDa protein
species. The 75 kDa species may correspond to the CD95-associated proteins RIP or FAF1 and the 35 kDa protein may represent
a TRADD analogue. These data indicate that several cellular proteins interact with CD95, possibly in a multi-protein complex,
and that a kinase activity is associated with CD95 not onlyin vitro but alsoin vivo. Therefore, receptor phosphorylation may play a role in CD95 signal transduction.

Over-expression of p21 ras-related protein was determined in the plasma by immunoblotting and in the tissue by immuno-histochemistry in a cohort of patients undergoing colonoscopy. In the plasma samples, p21 ras over-expression was detected in: 9% (4/47) of normal controls; 21% (13/61) of individuals with normal colonoscopies but with a prior history of colonic neoplasia; 12% (4/33) of small adenoma patients, 29% (6/21) of large adenoma patients; 63% (5/8) of carcinoma-in-adenoma patients; 50% (2/4) of Dukes' A carcinoma patients; and 20% (2/10) of Dukes' B-D carcinoma patients. In the tissue samples, p21 ras over-expression was detected in: 25% (2/8) of small adenoma patients; 44% (4/9) of large adenoma patients; 100% (4/4) of carcinoma-in-adenoma patients; and 33% (1/3) of Dukes' B-C carcinoma patients. For matched plasma-tissue pairs, there was a statistically significant correlation for p21 ras over-expression (R = 0.47, p = 0.02). 相似文献

Portable meters and simplified gas Chromatographic (GC) techniques were investigated for monitoring volatile hydrocarbon (HC), CO2, and O2, concentrations in groundwater, exhaust gases, and soil vapor during in situ remediation using soil vapor extraction (SVE) and air sparging (AS). Results of groundwater samples analyzed in‐house using a headspace technique compared well to split samples analyzed by a certified analytical laboratory (r2 = 0.94). SVE exhaust gas HC and CO2 concentrations measured using a GT201 portable HC/O2 meter and a RA‐411A meter (GasTech), respectively, were highly correlated with in‐house laboratory GC analyses (r2 = 0.91). O2 concentrations fell in a small range and meter analyses were not well correlated with laboratory analyses. Results of soil gas monitoring were not as well correlated as those for exhaust gases for HC, CO2, or O2, perhaps due to environmental conditions such as changes in relative humidity or the wider range of soil gas values. Overall, the meters were good indicators of vapor contamination, they greatly simplified estimates of total HC mass removal, and they allowed estimates of the biological contribution to contaminant removal during the remediation process. 相似文献

The integration of Fas/Apo-1 (CD95) by Fas ligand or anti-Fas antibody induces apoptosis, and this system plays a pivotal role for the lysis of target cells by cytotoxic T lymphocytes. Fas-mediated apoptosis is also increased by a prior incubation of Fas-bearing cells with interferon(IFN)-. Interleukin-1- converting enzyme (ICE) and/or CPP32, or other members of ICE family act as direct cell death executors downstream of this mechanism, and a tetrapeptide inhibitor of these cysteine proteases blocks Fas-mediated apoptosis. In this study, we examined the effect of IFN- on Fas-mediated apoptosis in ACHN cells. IFN- augmented apoptosis in a dose dependent manner and reached a plateau at 400 U/ml when exposed for 48 h before the end of culture. The kinetics revealed a significant increase in apoptosis after 24 h. Exposing ACHN cells to IFN- increased pro-ICE expression accompanied with a decrease of pro-CPP32. These results suggest that direct enhancement of ICE expression and/or upregulation of conversion of pro-CPP32 to active form increases Fas-mediated apoptosis by IFN- in ACHN cells. 相似文献

Twenty five environmental isolates enriched for their ability to grow onN-acetylphenylalanine as sole carbon source were investigated for their hydrolytic action on (+)-lactam (2-azabicyclo[2.2.1]hept-5-en-3-one). Strain CMC 3060, a mucoidal Gram-negative rod identified as a strain ofPseudomonas fluorescens, produced high levels of (+)lactamase, and was subsequently found to produce two distinct intracellular enantiomer-selective, -lactamases, one for each isomer. The (+)lactamase was produced constitutively whereas the (-lactamase was produced only in the presence of the substrate. The (+)lactamase was stable when stored as a frozen cell paste but unstable as a protein solution, losing activity during purification and storage. This enzyme was highly selective for the (+)lactam and showed no activity against a wide range of similar compounds. By use of rapid purification techniques and the inclusion of protease inhibitors and protein stabilisers, the (+)lactamase was purified to homogeneity by FPLC and found to be a monomer of molecular weight 61000 Da. 相似文献
