Dogs demonstrate an age-related cognitive decline, which may be related to a decrease in the concentration of omega-3 polyunsaturated fatty acids (n-3 PUFA) in the brain. Medium chain triglycerides (MCT) increase fatty acid oxidation, and it has been suggested that this may raise brain n-3 PUFA levels by increasing mobilization of n-3 PUFA from adipose tissue to the brain. The goal of the present study was to determine whether dietary MCT would raise n-3 PUFA concentrations in the brains of aged dogs. Eight Beagle dogs were randomized to a control diet (n = 4) or an MCT (AC-1203) enriched diet (n = 4) for 2 months. The animals were then euthanized and the parietal cortex was removed for phospholipid, cholesterol and fatty acid determinations by gas-chromatography. Dietary enrichment with MCT (AC-1203) resulted in a significant increase in brain phospholipid and total lipid concentrations (P < 0.05). In particular, n-3 PUFA within the phospholipid, unesterified fatty acid, and total lipid fractions were elevated in AC-1203 treated subjects as compared to controls (P < 0.05). Brain cholesterol concentrations did not differ significantly between the groups (P > 0.05). These results indicate that dietary enrichment with MCT, raises n-3 PUFA concentrations in the parietal cortex of aged dogs.  相似文献

The present study was undertaken to observe the effects of chemical fertilizers and compost amendments on microbial community structure in rice based cropping system under long term fertilizer application experiment. Changes in microbial community structure assessed by the relative abundance of fatty acid methyl ester (FAME) profiles revealed that compost amendments led to promoting the abundance of Gram-positive bacterial FAMEs. Multivariate analysis of FAME profiles separated the treatments primarily on the basis of the amount of added compost. A link between different fertilizer application treatment and soil microbial components was tentatively established, but it needs to be verified in further studies.  相似文献

目的:探讨西安地区妊娠妇女不同孕期血清铁蛋白、叶酸和维生素B12的水平及临床意义。方法:收集2012 年8 月至2013年1 月在本院进行产前检查的750 例妊娠期妇女及86 例健康非妊娠妇女的血清标本。采用全自动化学发光免疫分析法测定其血清铁蛋白、叶酸和维生素B12的含量。结果:孕妇血清铁蛋白、叶酸和维生素B12的含量随孕期的增长而逐渐降低。早、中、晚孕期妇女叶酸的含量较对照组相比,并无显著差异(P〉0.05)。中、晚孕期妇女血清铁蛋白和维生素B12含量均显著低于对照组,差异有统计学意义(P〈0.05)。结论:西安地区中、晚孕期的孕妇体内存在铁蛋白和维生素B12缺乏,但并不缺乏叶酸。在重视补充叶酸的同时,也应该重视对铁和维生素B12的补充。  相似文献

水杨酸对不同灌水下限青花菜生理特性及产量品质的影响   总被引:1,自引:0,他引:1  
以青花菜为试材,采用大田试验研究了0.15mmol/L水杨酸(SA)对不同灌水下限(75%、60%和45%土壤相对含水量)青花菜生理特性及产量品质的影响。结果显示:(1)随灌水下限的降低,青花菜叶片相对含水量、水势及叶绿素含量均呈下降趋势,而其叶片电解质渗透率以及游离脯氨酸(Pro)、丙二醛(MDA)、可溶性蛋白(Pr)含量逐渐升高;青花菜的花球重及维生素C含量(Vc)随灌水下限的降低而降低,相反硝酸盐含量及可溶性固形物含量随之升高。(2)叶面喷施0.15mmol/L SA显著提高了青花菜叶片相对含水量、水势、渗透调节物质含量、产量及品质,且降低了膜脂过氧化产物MDA含量,其中以45%灌水下限处理效果最为显著,60%灌水下限处理次之,75%灌水下限处理最小。研究认为,适宜浓度外源SA通过改善青花菜叶片水分生理状况,提高叶片渗透调节物质含量,降低质膜透性来增强其对干旱胁迫的抗性,且以60%灌水下限配合叶面喷施0.15mmol/L SA处理较佳。  相似文献

Background

Electron paramagnetic resonance (EPR) spectroscopy (also known as electron spin resonance, ESR, spectroscopy) is widely considered to be the “gold standard” for the detection and characterisation of radicals in biological systems.

Scope of review

The article reviews the major positive and negative aspects of EPR spectroscopy and discusses how this technique and associated methodologies can be used to maximise useful information, and minimise artefacts, when used in biological studies. Consideration is given to the direct detection of radicals (at both ambient and low temperature), the use of spin trapping and spin scavenging (e.g. reaction with hydroxylamines), the detection of nitric oxide and the detection and quantification of some transition metal ions (particularly iron and copper) and their environment.

Major conclusions

When used with care this technique can provide a wealth of valuable information on the presence of radicals and some transition metal ions in biological systems. It can provide definitive information on the identity of the species present and also information on their concentration, structure, mobility and interactions. It is however a technique that has major limitations and the user needs to understand the various pitfalls and shortcoming of the method to avoid making errors.

General significance

EPR remains the most definitive method of identifying radicals in complex systems and is also a valuable method of examining radical kinetics, concentrations and structure. The proteomic response to bacterial infection in a teleost fish (Paralichthys olivaceus) infected with Streptococcus parauberis was analyzed using label-free protein quantitation coupled with LC-MS(E) tandem mass spectrometry. A total of 82 proteins from whole kidney, a major lymphoid organ in this fish, were found to be differentially expressed between healthy and diseased fish analyzed 6, 24, 72 and 120 h post-infection. Among the differentially expressed proteins, those involved in mediating immune responses (e.g., heat shock proteins, cathepsins, goose-type lysozyme and complement components) were most significantly up-regulated by infection. In addition, cell division cycle 48 (CDC48) and calreticulin, which are associated with cellular recovery and glycoprotein synthesis, were up-regulated in the universal protein group, whereas the other proteins in that group were down-regulated. There was continuous activation of expression of immune-associated proteins during infection, but there was also loss of expression of proteins not involved in immune function. We expect that our findings regarding immune response at the protein level would offer new insight into the systemic response to bacterial infection of a major immune organ in teleost fish.  相似文献

目的:探讨游离脂肪酸(FFA)作用下胰岛βTC3细胞双链RNA依赖性蛋白样内质网激酶(PERK)的表达以及利拉鲁肽(Lira)对其表达的干预作用。方法:以βTC3细胞为研究对象,分为对照组和FFA组(0.125,0.25,0.5及1 mmol/L)孵育24 h,Westernblot方法检测PERK的表达。然后,分为对照组,FFA组,和FFA+Lira组(0.5 mg/L和1 mg/L),Lira预孵育6 h后,1 mmol/L FFA继续孵育24 h,Western blot检测PERK的表达。结果:①不同浓度FFA孵育24 h后,与对照组相比,1 mmol/L FFA组PERK表达增加(P<0.05)。②与1 mmol/L FFA组相比,0.5mg/L Lira+1 mmol/L FFA和1 mg/L Lira+1 mmol/L FFA组PERK表达减少(P<0.05),两组之间有统计学差异(P<0.05)。结论:FFA作用能够上调βTC3细胞PERK的表达,而Lira在一定程度上逆转FFA水平异常导致的βTC3细胞PERK表达上调,减轻内质网应激反应。  相似文献
