Using a combination of gas-exchange and chlorophyll fluorescence measurements, low apparent CO2/O2 specificity factors (1300 mol mol?1) were estimated for the leaves of two deciduous tree species (Fagus sylvatica and Castanea sativa). These low values contrasted with those estimated for two herbaceous species and were ascribed to a drop in the CO2 mole fraction between the intercellular airspace (Ci) and the catalytic site of Rubisco (Cc) due to internal resistances to CO2 transfer. Cc. was calculated assuming a specificity of Rubisco value of 2560 mol mol?1. The drop between Ci and Cc was used to calculate the internal conductance for CO2 (gi). A good correlation between mean values of net CO2 assimilation rate (A) and gi was observed within a set of data obtained using 13 woody plant species, including our own data. We report that the relative limitation of A, which can be ascribed to internal resistances to CO2 transfer, was 24–30%. High internal resistances to CO2 transfer may explain the low apparent maximal rates of carboxylation and electron transport of some woody plant species calculated from A/Ci curves.  相似文献

Effects of salinity on seed set in rice   总被引:15,自引:1,他引:14  
Salinity reduces fertility in rice (Oryza sativa L.), but little is known of the underlying cause(s). In order to determine the relative importance of pollen viability and stigmatic receptivity for seed setting, plants of the rice cultivar IR36 were treated with ‘artificial’ sea water (0,10, 25 or 5Omol?3 with respect to NaCl) from 1 month after germination until the main tiller flowered. An increase in the salinity in the medium resulted in a decrease in the number of fertile florets and in the viability of pollen as determined both by pollen germination and by pollen staining with the tetrazolium salt 3-(4,5-dimethyl-ithyazolyl)-2,5-diphenyl monotetrazolium bromide. In order to assess the effects of salt on stigmas, seed production was measured for salt-grown and non-salt-grown female plants pollinated with viable pollen (from plants grown in the absence of salt). The percentage of seed set was reduced by 38% when the female plants were grown in 1Omol m?3 Na and by 72% at 25mol m?3 Na: no seed setting was recorded for plants grown in 50mol m?3 Na. Comparisons between crosses involving male and female parents grown at different salinities indicated that effects on the female plants dominated those on pollinator plants. Mineral analysis of leaves of different ages showed that there was a gradient of K concentration from leaf to leaf which was opposite to that of Na and Cl at all levels of applied salinity: K was maximal in the flag leaf, where Na and Cl were minimal. Analysis also revealed that there was an increase in the concentrations of Na and Cl and a decrease in the concentration of K in the pollen grains and stigmas of plants subjected to saline conditions. Correlations between the concentration of Na and Cl in pollen and pollen staining and pollen germination in vitro suggest that Na and Cl perse were responsible for the poor viability. The change in ionic concentrations in pollen and stigmas was much larger than that in the younger leaves, and in particular very much larger than that in the lemmas and paleas.  相似文献

Data on root parameters and distribution are important for an improved understanding of the factors influencing nutrient uptake by a crop. Therefore, a study was conducted on a Crowley silt loam at the Rice Research and Extension Center near Stuttgart, Arkansas to measure root growth and N, P and K uptake by three rice (Oryza sativa L.) cultivars at active tillering (36 days after emergence (DAE)), maximum tillering (41 DAE), 1.25 cm internode elongation (55 DAE), booting (77 DAE) and heading (88 DAE). Soil-root core samples were taken to a depth of 40 cm after plant samples were removed, sectioned into 5 cm intervals, roots were washed from soil and root lengths, dry weights and radii were measured. Root parameters were significantly affected by the soil depth × growth stage interaction. In addition, only root radius was affected by cultivar. At the 0- to 5-cm soil depth, root length density ranged from 38 to 93 cm cm-3 throughout the growing season and decreased with depth to about 2 cm cm-3 in the 35- to 40-cm depth increment. The increase in root length measured with each succeeding growth stage in each soil horizon also resulted in increased root surface area, hence providing more exposed area for nutrient uptake. About 90% of the total root length was found in the 0- to 20-cm soil depth throughout the season. Average root radius measured in the 0- to 5-cm and 35- to 40-cm depth increments ranged from 0.012 to 0.013 cm and 0.004 to 0.005 cm, respectively throughout the season. Total nutrient uptake by rice differed among cultivars only during vegetative growth. Differences in total nutrient uptake among the cultivars in the field appear to be related to absorption kinetics of the cultivars measured in a growth chamber study. Inheritance of foreign genes neo and gusA in rice (Oryza sativa L. cv. IR54 and Radon) has been investigated in three different primary (T0) transformants and their progeny plants. T0 plants were obtained by co-transforming protoplasts from two different rice suspension cultures with the neomycin phosphotransferase II gene [neo or aph (3) II] and the -glucuronidase gene (uidA or gusA) residing on separate chimeric plasmid constructs. The suspension cultures were derived from callus of immature embryos of indica variety IR54 and japonica variety Radon. One transgenic line of Radon (AR2) contained neo driven by the CaMV 35S promoter and gusA driven by the rice actin promoter. A second Radon line (R3) contained neo driven by the CaMV 35S promoter and gusA driven by a promoter of the rice tungro bacilliform virus. The third transgenic line, IR54-1, contained neo driven by the CaMV 35S promoter and gusA driven by the CaMV 35S.Inheritance of the transgenes in progeny of the transgenic rice was investigated by Southern blot analysis and enzyme assays. Southern blot analysis of genomic DNA showed that, regardless of copy numbers of the transgenes in the plant genome and the fact that the two transgenes resided on two different plasmids before transformation, the introduced gusA and neo genes were stably transmitted from one generation to another and co-inherited together in transgenic rice progeny plants derived from self-pollination. Analysis of GUS and NPT II activities in T1 to T2 plants provided evidence that inheritance of the gusA and neo genes was in a Mendelian fashion in one plant line (AR2), and in an irregular fashion in the two other plant lines (R3 and IR54-1). Homozygous progeny plants expressing the gusA and neo genes were obtained in the T2 generation of AR2, but the homozygous state was not found in the other two lines of transgenic rice.  相似文献

Rice seedlings accumulate stainable amounts of the 104 and 90 kDa polypeptides in response to high temperature stress. We have purified and raised highly specific polyclonal antisera against both of these polypeptides. In western blotting experiments, we find that these proteins are accumulated to different extents in rice seedlings subjected to salinity (NaCl), water stress, low-temperature stress and exogenous abscisic acid application. These proteins also accumulated when rice seedlings grown in pots under natural conditions were subjected to water stress by withholding watering. Seedlings of Triticum aestivum, Sorghum bicolor, Pisum sativum, Zea mays, Brassica juncea and mycelium of Neurospora crassa showed accumulation of the immunological homologues of both the 104 and the 90 kDa polypeptides, in response to high-temperature stress. We have earlier shown that shoots of rice seedlings exposed to heat shock accumulate a 110 kDa polypeptide which is an immunological homologue of the yeast HSP 104 (Singla and Grover, Plant Mol Biol 22: 1177–1180, 1993). Employing anti-rice HSP 104 antibodies and anti-yeast HSP 104 antibodies together, we provide evidence that rice HSP 104 is different from the earlier characterized rice HSP 110.  相似文献

A total of 687 DNA sequence accessions from the Mendel database (release 1.04, 3 November 1994) assigned standardized designations for plant genes and gene products were used in aBLAST similarity search of 7557 rice partial cDNA sequences and 287 other rice sequences from the Japanese Rice Genome Research Program. We describe procedures for data manipulation, import and export from and to Macintosh and Unix, and the use of 4th Dimension relational database management system (RDBMS) in data processing. Altogether 275 sequences showed strong similarity hits. Using the CPGN nomenclature, we assign putative designations for genes and gene products. Assignments include representatives of 26 gene products, including 58 cDNA sequences similar to α-tubulins (TubA), 23 similar to β-tubulins (TubB) and 51 similar to cytosolic subunit C of glyceraldehyde-3-phosphate dehydrogenase (NAD) (GapC). The results of the similarity searches are listed and are also available electronically. The assignments have been submitted to the CPGN working groups for verification and for later inclusion in the GenBank/EMBL/DDBJ sequence databases, which will include the standardized designations in the accession data fields. The results of studies on assimilate and water transport in the developing caryopsis of rice are summarised. Evidence is presented for a symplastic movement of solutes as far as the aleurone layer. However, transport into the apoplast at the nucellus/aleurone interface appears to be a necessary step due to the absence of plasmodesmata at this site. It is suggested that water leaves the caryopsis during grain filling by the isolated cell walls of the pigment strand, the suberised walls of these cells functioning to isolate the apoplast from the symplast and thereby allowing opposing fluxes of water and assimilates to occur in the dorsal region of the grain.  相似文献

Summary In order to examine changes in survival and mutation rates during a cell cycle in higher plant, fertilized egg cells of rice were irradiated with X-rays at 2 h intervals for the first 36 h after pollination, i.e., at different phases of the first and second cell cycles. The most sensitive phase in lethality was late G1 to early S, followed by late G2 to M, which were more sensitive than the other phases. In both M1 and M2 generations, sterile plants appeared most frequently when fertilized egg cells were irradiated at G2 and M phases. Different kinds of mutated characters gave rise to the respective maximum mutation rates at different phases of a cell cycle: namely, albino and viridis were efficiently induced at early G1, xantha at early S, short-culm mutant at mid G2, heading-date mutant at M to early G1. The present study suggests the possibility that the differential mutation spectrums concerning agronomic traits are obtained by selecting the time of irradiation after pollination.  相似文献

Summary The response of rice (Oryza sativa L.) embryos to X-rays (M1 to M3) was studied. By means of irradiating excised embryos, both chlorophyll and macromutation were successfully induced in three genotypes of rice. However, differential responses in terms of mutation frequency, mutation spectrum and optimal levels of X-rays required for induction of mutation (chlorophyll as well as morphological) were found to exist between cultivars. In Satika and Ashkhata, LD50 values and maximum induced seed sterility are concomitant to optimum level of radiation required for triggering chlorophyll mutation. However, optimum dose for induction of macromutation in Satika and Kerangserang is independent of either LD50 and/or induced seed sterility.Chances of obtaining both dominant and locus specific recessive mutations in the immediate X-ray treated generation (M1) are large. This indicates the very high degree of effectiveness of the excised embryo irradiation technique with rice.  相似文献
