PPD皮试引起的过敏性紫癜一例

患者,女,回族,15岁,永靖县回民中学初一年级学生。2008年10月27日县疾控中心组织的专业技术人员在永靖县回中开展正常学生体检时使用成都生物所生产的卡介苗纯蛋白衍生物（BCG—PPD）,批号为20080523（1-4）,有效期：2009．05．04,按常规给患者做了PPD试验（结素试验）。     

Structure variations of TBA G-quadruplex induced by 2＋-O-methyl nucleotide in K＋ and Ca2＋ environments

Thrombin binding aptamer （TBA）, a 15-mer oligonucleo- tide of d（GGTTGGTGTGGTTGG） sequence, folds into a chair-type antiparallel G-quadruplex in the K＋ environ- ment, and each of two G-tetrads is characterized by a syn-anti-syn-anti glycosidic conformation arrangement. To explore its folding topology and structural stability, 2＇-0- methyl nucleotide （OMe） with the C31-endo sugar pucker conformation and anti glycosidic angle was used to selectively substitute for the guanine residues of G-tetrads of TBA, and these substituted TBAs were characterized using a circular dichroism spectrum, thermally differential spectrum, ultra- violet stability analysis, electrophoresis mobility shift assay, and thermodynamic analysis in K＋ and Ca2＋ environments. Results showed that single substitutions for syn-dG residues destabilized the G-quadruplex structure, while single substi- tutions for anti-dG residues could preserve the G-quadruplex in the K＋ environment. When one or two G-tetrads were modified with OMe, TBA became unstructured. In contrast, in Ca2＋ environment, the native TBA appeared to be un- structured. When two G-tetrads were substituted with OMe, TBA seemed to become a more stable parallel G-4 structure. Further thermodynamic data suggested that OMe-substitu- tions were an enthalpy-driven event. The results in this study enrich our understanding about the effects of nucleo- tide derivatives on the G-quadruplex structure stability in different ionic environments, which will help to design G-quadruplex for biological and medical applications.     

含磷酸结合口袋的BRCT结构域功能位点预测

BRCT( BRCA1 C-terminus)是真核生物DNA损伤修复系统重要的信号传导和蛋白靶向结构域.为了探讨含磷酸结合口袋的BRCT与磷酸化配体结合的机制,对XRCC1 BRCT1、PTIP BRCT4、ECT2 BRCT1和TopBP1BRCT1进行了结构保守性和表面静电势分析.结果显示,4个BRCT的磷酸结合口袋周围所存在的结构保守并带正电势的沟槽很可能是其功能位点,并且类似的沟槽在含磷酸结合口袋的BRCT中普遍存在.沟槽两侧及底部均带有极性氨基酸残基,两侧带正电荷,而底部疏水.这说明沟槽与配体的结合以静电和疏水相互作用为主.沟槽主要位于单个BRCT中,而且4个BRCT的沟槽在形状和电荷分布上都不同,说确明BRCT配体特异性主要由单个BRCT决定.磷酸结合口袋位于沟槽中心,说明沟槽可能同时结合磷酸化残基的N端和C端附近残基.     

ATP竞争性mTOR抑制剂研究进展

哺乳动物雷帕霉素靶蛋白(mTOR)是一种丝氨酸/苏氨酸激酶,可集合胞内和胞外信号调节细胞生长、增殖、代谢和存活。在肿瘤的发生发展过程中,mTOR信号多处于异常激活状态,已被确证为肿瘤药物治疗的重要靶标。因雷帕霉素临床应用存在局限性,近期ATP竞争性mTOR抑制剂的研发十分活跃。本文综述了ATP竞争性mTOR抑制剂的研究进展,重点讨论药物的作用特点。     

125I-UdR提高内皮抑素基因的抑瘤作用

目的:探讨125I-UdR提高内皮抑素基因对大鼠种植癌模型的抑制效应。方法:制作肿瘤株Walker-256细胞大鼠皮下种植癌的动物模型并分为1-4组(对照组、125I-UdR组、Endostatin组和Endostatin+125I-UdR组),每组20只,通过实体瘤内注射,分别给与相同体积生理盐水125I-UdR、内皮抑素基因及125I-UdR和内皮抑素基因混合物,测量肿瘤治疗前体积(V0)和治疗后不同时间体积(Vt),计算10d、20d的肿瘤生长率(f=Vt/V0),观察各组肿瘤在光镜下的变化。结果:各组大鼠10d、20d肿瘤生长率为:(11.03±1.08、27.35±1.08),(4.02±0.79、7.58±2.98),(3.88±0.26、7.02±2.75),(2.72±1.01、2.94±1.26),2、3、4组的肿瘤生长率明显小于1组(P<0.001);2、3组之间肿瘤生长率差别不明显(P>0.05),4组肿瘤生长率小于2、3组(P<0.01)。结论:通过实体瘤内注射的方法给与125I-UdR、内皮抑素基因及两者混合物后,能够明显抑制肿瘤的生长,内皮抑素基因和125I-UdR联合治疗在抑制肿瘤生长方面作用更加显著。     

慢性乙型肝炎抗病毒治疗研究进展

乙肝病毒的慢性感染是肝纤维化、肝细胞癌的重要病因,积极有效地抗病毒治疗,可显著改善HBV感染者的生活质量。干扰素、核苷(酸)类药物的抗乙肝病毒疗效已得到全球公认,本文就上述两类药物的研究进展进行综述。     

骨髓瘤细胞中阿霉素诱导的ATP 变化与自噬的关系

目的:检测用阿霉素(doxorubicin DOXO)处理的骨髓瘤细胞株NCI-H929中ATP与自噬表达水平的变化,探讨两者之间的关联。方法:分别以DOXO 2umol/l 24h、DOXO 2umol/l联用自噬抑制剂3MA 10mmol/l 24h处理H-929细胞后,采用MTT法检测细胞存活率;ATP生物发光法检测ATP表达量;Western Blot检测靶细胞自噬标志分子LC3蛋白的表达。结果:各组相对未处理组存活率分别为54%、35%;相对未处理组%ATP分别为400%、150%;DOXO 24h LC3表达显著上调。结论:经DOXO处理H-929细胞系自噬形成,进而ATP上升以保护细胞。
