畜禽遗传资源系统保存的理论与模拟实验研究──I．畜禽遗传资源系统保存的理论分析

本文提出了畜禽遗传资源系统保存的概念、基本思想及其群体遗传结构变化的数学模型。该理论是将一定时空内某一畜种所拥有的全部基因作为保存对象,既将活体保存作为基本方法,又将其有机地与高新生物技术结合在一起;既追求系统地保存控制畜种特性的基因资源,又可达到保存地方品种的目的。在假定无世代重叠,群体内存在选择、突变、迁移,且考虑漂变效应的情况下,本文所构建的两个数学模型可分别用来描述一个座位上多个主基因频率或数量性状群体均数的动态变化。

陆地表层系统动力学机制研究Ⅰ．──陆地表层系统和动力学方程

对陆地表层动力学的研究对象、研究内容及其基本理论方法作了阐述,以多学科综合的观点定义了陆地表层系统,包括时空尺度范围的约定、状态变量的选择、物质循环以及外界因子等的讨论;依据物质和能量守恒原理建立了陆地表层系统的非线性控制方程组,表述了各圈层之间物质循环和能量循环过程、反馈关系及其动力学联系;讨论了控制方程组的整体运作功能以及人类活动对陆地表层系统的影响．     

中药材分类中的聚类分析

本文应用聚类分析中的系统聚类法与模糊聚类法,对植物药材百合的18个样品进行了聚类．结果不仅两法所得的结论相一致,而且与传统分类的情况相符合．     

A组轮状病毒SA11VP6基因的克隆和表达

从ＳＡ１１ＶＰ６基因全序列克隆开始,设计一对两端带有酶切位点的引物,逆转录ＰＣＲ扩增出ＶＰ６全基因ＣＤＮＡ。经酶切后插入ＰＵＣ１９,构建了ＶＰ６全基因克隆ＰＲＡ６。再经酶切后插入痘苗病毒载休质凿ＰＪＳＡ１１７５中。利用Ｌｉｐｏｆｅｃｔｉｎ导入ＴＫ１４３细胞,利用ＴＫ基因和Ｌａｃ基因作为重组病毒的筛选标记。表达产物用单克隆抗体ＥＬＩＳＡ法检测,发现细胞培养上清和细胞裂解液都是阳性。Ｗｅｓｔｅｒｎ ｂ     

天花粉凝集素的固定化及其应用

制备了天花粉凝集素ＴＫＬ－Ｓｅｐｈａｒｏｓｅ４Ｂ的亲和柱,用来分离其它来源物质中与ＴＫＬ相互作用的糖复合物,发现从兔红细胞的血影细胞的抽提物中可以得到分子量为６２ｋＤ,４０ｋＤ等糖蛋白,这可能就是ＴＫＬ凝集兔红细胞的分子基础,从人及骆驼,绵羊血浆中也能得到与ＴＫＬ结合的,分子量不等的糖蛋白,进一步鉴定人血浆ＴＫＬ结合物中含有α１－抗胰蛋白酶等,为从血浆中分离这些糖蛋白提供了较为便利的方法,在寻找天     

Physical mapping of three fruit ripening genes：Endopolygalacturonase,ACC oxidase and ACC synthase from apple（Malus x domestica） in an apple rootstock A106（Malus sieboldii

The apple rootstock,A106(Malus sieboldii),had 17 bivalents in pollen mother cells at meiotic metaphase 1,and 17 chromosomes in a haploid pollen cell.Karyotypes were prepared from root-tip cells with 2n=34 chromosomes,Seven out of 82 karyotypes(8.5%) showed one pari of satellites at the end of the short arm of chromosome 3.C-bands were shown on 6 pairs of chromosomes 2,4,6,8,14,and 16 near the telomeric regions of short arms.Probes for three ripening-related genes from Malus x domestica:endopolygalacturonase(EPG,0.6kb),ACC oxidase(1.2kb),and ACC synthase(2kb)were hybridized in situ to metaphase chromosomes of A106.Hybridization sites for the EPG gene were observed on the long arm of chromosome 14 in 15 out of 16 replicate spreads and proximal to the centromere of chromosomes 6 and 11.For the ACC oxidase gene,hylridization sites were observed in the telomeric region of the short arm of chromosomes 5 and 11 in 87% and 81% of 16 spreads respectively,proxiaml to the centromere of chromosome 1 in 81% of the spreads,and on the long arm of chromosome 13 in 50% of the spreads. Physical mapping of three fruit ripening genes in an apple rootstock A106.Twenty five spreads were studied for the ACC synthase gene and hybridization sites were observed in the telomeric region of the short arm of chromosome 12 in 96% of the spreads.chromosomes 9 and 10 in 76% of the spreads,and chromosome 17 in 56% of the spreads.     

Chromosomal localization of ribosomal DNA sequences in an apple rootstock using a digoxygenin detection system

A 6kb rDNA probe comprising the 18S coding plus spacer sequences has been hybridized to the metaphase chromosomes of apple rootstock cultivar MM106 demonstrating the localization of ribosomal gene arrays in the vicinity of the telomeric regions of the short arms of chromosomes 6 and 14.The in situ results using digoxygenin labelling coupled to an alkaline phosphatase immunoassay were confirmed by silver staining for NORs and nucleoli.This study demonstrates the feasibility of molecular cytogenetic analysis of very small chromosomes(1.0-2.7μm） of apple.
