浅析汤剂药物有效成分的影响因素

汤剂是我国应用历史最悠久的剂型,已有三千多年的历史.因为汤剂比其它中药剂型易于吸收而奏效迅速,更重要的是,它的处方组成以及用量均可针对病情随时加减,适应中医"辨证论治"的要求.     

Redox status of thioredoxin-1 （TRX1） determines the sensitivity of human liver carcinoma cells （HepG2） to arsenic trioxide-induced cell death

lntracellular redox homeostasis plays a critical role in determining tumor cells＇ sensitivity to drug-induced apoptosis. Here we investigated the role of thioredoxin-1 （TRX1）, a key component of redox regulation, in arsenic trioxide （AS2O3）-induced apoptosis. Over-expression of wild-type TRX1 in HepG2 cells led to the inhibition of As2O3-induced cytochrome c （cyto c） release, caspase activation and apoptosis, and down-regulation of TRX1 expression by RNAi sensitized HepG2 cells to As2O3-induced apoptosis. Interestingly, mutation of the active site of TRX1 from Cys^32/35 to Ser^32/35 converted this molecule from an apoptotic protector to an apoptotic promoter. In an effort to understand the mechanisms of this conversion, we used isolated mitochondria from mouse liver and found that recombinant wild-type TRX1 could protect mitochondria from the apoptotic changes. In contrast, the mutant form of TRX1 alone elicited mitochondria-related apoptotic changes, including the mitochondrial permeability transition pore （mPTP） opening, loss of mitochondrial membrane potential, and cyto c release from mitochondria. These apoptotic effects were inhibited by cyclosporine A （CsA）, indicating that mutant TRX1 targeted to mPTP. Alteration of TRX1 from its reduced form to oxidized form in vivo by 2,4-dinitrochlorobenzene （DNCB）, a specific inhibitor ofTRX reductase, also sensitized HepG2 cells to As203-induced apoptosis. These data suggest that TRX1 plays a central role in regulating apoptosis by blocking cyto c release, and inactivation of TRX1 by either mutation or oxidization of the active site cysteines may sensitize tumor cells to As2O3-induced apoptosis.     

杜仲抗真菌肽对黄曲霉的抑制作用(英文)

用纸片琼脂扩散法,研究了杜仲抗真菌肽EAFP3对黄曲霉(Aspergillus flavus)的抗真菌活性.研究结果表明,EAFP3对黄曲霉有明显抑制作用,最小抑菌浓度为125 μg/mL.     

人参EST资源的SSR信息分析

从7055条人参EST序列中搜索出791个SSR,其出现频率为11．21％,平均长度为21．37bp,平均分布频率为1／5．7kb。二核苷酸重复是主要的重复类型,占全部EST-SSR的56．89％,其次是三核苷酸重复的占全部SSR的21．11％。AT、GAA是二核苷酸和三核苷酸中出现次数最多的重复基元类型,分别占28．89％和10．18％。     

津田芜菁和赤丸芜菁苯丙氨酸解氨酶基因(PAL)的克隆和表达

以UV—A处理津田芜菁和赤丸芜菁块根24h后提取总RNA,再用RT—PCR方法分别克隆BrPAL1和BrPAL2基因的结果表明,BrPAL1和BrPAL2的开放读码框为2169bp,编码722个氨基酸。氨基酸序列分析显示,BrPAL1和BrPAL2与甘蓝型油菜苯丙氨酸解氨酶（PAL）的同源性达99％,第61～559的肽段具有PAL结构域。BrPAL1和BrPAL2的核苷酸序列在9个位置上存在差异,而推导的氨基酸序列仅在3个位置上有差异。BrPAL1和BrPAL2基因有高度同源性。Northern杂交结果显示,UV—A可以诱导BrPAL1和BrPAL2表达,基因的表达量与处理时间呈相关。     

基于叶绿体DNA trnL-F序列研究肋果沙棘的谱系地理学

由于青藏高原的地理效应,第四纪冰期气候的反复变化应对现今该地区生物的地理分布及其居群遗传结构产生重大影响。肋果沙棘Hippophae neurocarpa是青藏高原地区的一个特有种,根据叶上的附属物（星状鳞毛或者鳞片状鳞毛）分为两个亚种：肋果沙棘亚种subsp.neurocarpa和密毛肋果沙棘亚种subsp.stellatopilosa。依据母系遗传的叶绿体DNA片段对该物种谱系地理学进行研究不仅能阐明该物种冰期避难所,而且对于理解两个亚种的母系分化也具有重要意义。共对两个亚种14居群的70个个体的trnL-F序列进行了测序,共发现8种单倍型,其中5种单倍型出现在肋果沙棘亚种中,4种出现在密毛肋果沙棘亚种中,两个亚种共享一种单倍型。种内谱系分化与两个亚种形态上的分化不一致。嵌套进化分支把8种单倍型分为三支：一支为肋果沙棘亚种,其他两支中两个亚种单倍型嵌套组成,且肋果沙棘亚种处于进化末端。我们的研究结果还表明肋果沙棘在冰期可能在高海拔地区存在多个避难所,并且存在来自避难所的邻域扩张。     

四川泗耳自然保护区及邻近地区两栖爬行动物初步调查

2002年9月4～16日对四川省平武县泗耳自然保护区及邻近地区的两栖爬行动物进行了调查。共发现16种,隶属12属7科。动物地理区系组成分析表明该地以东洋界西南区成分为主,同时周围各动物地理区的某些物种在这里交汇。建议适当扩大该保护区的面积,使其包括一些较低海拔的生境,以利对一些重点保护物种的有效保护。     

生长猪蛋白质周转代谢三室模型建立及参数求解

介绍了用房室分析方法研究蛋白质动态代谢规律的数学模型,建立了蛋白质动态代谢的三室模型并给出了用单剂量终末产物法求解改模型参数的具体方法。     

新疆北部森林生态系统地衣植物生活型分析

Life forms of lichen of forest ecosystem in Northern Xinjiang were researched in the Kanas National Natural Reservation Area.They have been grouped into 6 types:Epipetria,Soil-epipetria,Epiphytia arboricosa,Epiphytia moss,Epiphytis grass and Dry-radicantia,in which the species of Epiphytia arboricosa account for 36.89% of the total species in this area,the species of Epipetria for 22.33% and Epiphytia grass the least.The results show that life forms of lichem at the srea in Northem Xinjiang are relatively rich,which indicates the biodiversity characteristics of lichen.     

尿素改善SDS-PAGE分离小分子肽的效果

目的：探讨尿素对Tricine—SDS—PAGE系统分离小分子蛋白的影响。方法：利用常规SDS-PAGE和Tricine-SDS-PAGE分离小分子肽,比较不同组成的分离胶的分离效果。结果：调整聚丙烯酰胺凝胶的分子组成,使丙烯酰胺和甲叉双丙烯酰胺的交联度为5．05％,能够改善小分子肽的分离效果。加入36％的尿素比加入甘油可以更有效分离1kD的小肽。但尿素胶聚合太快影响分离效果。结论：尿素改善SDS—PAGE分离小分子肽的效果,制作尿素胶时,宜采用低浓度的AP和TEMED使凝胶慢速聚合。
