水稻TFⅢA型锌指蛋白基因ZFP207的克隆和表达分析 总被引:2,自引:0,他引:2
锌指蛋白是一类重要的转录因子, 广泛参与植物的生长发育和胁迫应答过程。文章使用生物信息学方法从水稻中预测并克隆了一个TFⅢA型锌指蛋白基因ZFP207(GenBank 登陆号: AK063147.1), 该基因包含一个567 bp的开放阅读框, 编码一条188个氨基酸组成的多肽,其编码产物的预测分子量为20.72 kDa, 等电点为9.67。锌指蛋白ZFP207含有一个典型的TFⅢA型锌指结构, 并且在C端含有一个可能具有转录抑制功能的EAR-motif, 但不具有任何已知的核定位信号。系统发生分析结果表明, ZFP207与植物中已经发现的单锌指蛋白亲缘关系较近。通过RT-PCR方法分析了ZFP207基因在成株期水稻不同组织中的表达, 发现ZFP207在茎和叶中表达量较高, 而在穗和根中表达量较低。在酵母中的转录激活实验结果显示, ZFP207在酵母中不具有转录激活功能。 相似文献

心脏毒素cardiotoxinⅢ(CTXⅢ)是中华眼镜蛇毒主要组分之一,根据Genbank中已有的CTXs序列设计特异性引物,从中华眼镜蛇毒中克隆出208bp的CTXⅢ片段,并将该基因片段分别克隆至大肠杆菌His—patch ThioredoxinB载体中,重组CTXⅢ经渗透休克分泌至胞外。将CTXⅢ基因插入酵母pPIC9K分泌型表达载体重组表达,N端带有3个氨基酸(GYT)残基的重组CTXⅢ(rCTXⅢ)分泌量达9.5mg/L,经一步凝胶过滤纯化,其纯度达90%以上,纯化率达65%。经细胞毒性实验检测,纯化的rCTXⅢ 12h的IC50为4.66μg/ml,证实重组蛋白具有良好的生物学活性。 相似文献

Carboxypeptidase Ⅲ from germinating triticale grains was purified 434.2-fold with a six-step procedure including: homogenization, ammonium sulfate precipitation, cation-exchange chromatography on CM-cellulose, gel filtration chromatography on Sephadex G-150, cation-exchange chromatography on SP8HR column (HPLC), and affinity chromatography on CABSSepharose 4B. Triticale carboxypeptidase Ⅲ is a monomer with a molecular weight of 45 kDa, which optimally hydrolyzes peptides at temperature 30-50℃ and pH 4.6. N-CBZ-Ala-Phe, N-CBZ-Ala-Leu, and N-CBZ-AIa-Met are hydrolyzed at the highest rates. Amino acids with aromatic or large aliphatic side chains are preferred in position P1', whereas the presence of these types of groups in position P1 of the substrate results in a lower rate of hydrolysis. Peptides containing glutamic acid in positions P1 are poor substrates for the enzyme. This phenomenon suggests the hydrophobic substrate-binding sites S1 and S1'. The active site contains serine since diisopropylfluorophosphate and phenylmethanesulfonyl fluoride reduce the activity by 89.9% and 81.5%, respectively. Moreover, the activity of triticale carboxypeptidase Ⅲ is reduced by mercury ions and organomercurial compounds, which suggests the presence of a sulfhydryl group adjacent to the active site of the enzyme. Identification of purified enzyme by mass spectrometry method demonstrated that the enzyme is a homolog of barley carboxypeptidaseⅢ. 相似文献

BackgroundAs a newly discovered muscle factor secreted by skeletal muscle cells, irisin is a polypeptide fragment formed from hydrolysis of fibronectin type Ⅲ domain-containing protein 5 (FNDC5). Irisin can promote beigeing of white adipose tissue (WAT) and regulate glucose and lipid metabolisms. However, the functions of irisin in skeletal muscle development remain largely unknown. In order to characterize the expression of irisin, this study investigated the expression of irisin precursor FNDC5 in myoblasts and skeletal muscles during different developmental stages of SPF mice.ResultsThe Western blot, quantitative real-time PCR (qRT-PCR), and immunofluorescence assay results showed that FNDC5 was expressed in all the developmental stages of myoblasts and gastrocnemius, but its expression differed at different stages. FNDC5 protein exhibited the highest expression in gastrocnemius of sexually mature mice, followed by elderly mice and adolescent mice, and it displayed the lowest expression in pups. Additionally, FNDC5 protein was mainly expressed in cytoplasm, and it had the highest expression in primary myoblasts, followed by the myotubes with the lowest expression in C2C12 myogenic cells.ConclusionsOverall, FNDC5 was mainly expressed in cytoplasm and extracellular matrix with different expression levels at different developmental stages of skeletal muscle cells and tissues in mice. This study will provide new strategies for promoting skeletal muscle development and treating muscle- and metabolism-related disease by using irisin. 相似文献

Laccases (benzenediol: oxygen oxidoreductases, EC1.10.3.2) can oxidize various substrates, and those which are tolerant to and even activated by salts have attracted a lot of attention due to their application potential in certain industries. The mechanism of the salt activation of laccases is awaiting to be elucidated yet. Our previous study (Li, Xie et al. 2018) supposed that the salt activation of marine laccase Lac15 might be attributed to Cl- ion specifically binding to some local sites to interfere substrate binding and/or electron transfer. In this study, we found two sites whose mutations resulted in elimination of the salt activation of Lac15’s activity towards catechol and dopamine respectively, and revealed that the mutations affected the activity by altering both Em and kcat, demonstrating the supposed mechanism. A model for the salt activation of laccases was accordingly proposed, albeit some details are to be elucidated. 相似文献
