For the purpose of describing the pathway by which estrogens are synthesized in the rhesus monkey (Macacamulatta) corpus luteum (CL), CL were obtained during the midluteal phase of the menstrual cycle and fragments incubated with equimolar amounts of [7-3H]pregnenolone plus [4-14C]progesterone. Metabolites including 3H-progesterone, 3H, 14C-20α-dihydroprogesterone, 3H, 14C-17-hydroxyprogesterone, 3H-estrone and 3H-estradiol-17β appeared in the medium during the first 20 minutes of incubation, 3H, 14C-Androstenedione was not consistently noted until after 60 minutes. Despite the fact that the 14C/3H-17-hydroxyprogesterone ratio quickly approached a constant value in the medium, 14C-estrogens were not detected in the medium or tissue fragments suggesting that progesterone was not a principal precursor for estrogen synthesis. As evidenced by the observation that the 14C/3H-progesterone ratio was significantly higher in luteal fragments than the 17-hydroxyprogesterone ratio, 17-hydroxyprogesterone appeared to be synthesized from pregnenolone both by way of progesterone and by another route which did not include progesterone. C21- and C18-Steroids were more concentrated in tissue fragments after 120 minutes of incubation than in the medium indicating that these steroids were sequestered by luteal tissue.  相似文献

Summary Seeds from 27 wild strains (18 tetraploids and 9 diploids) of Glycine weightii were germinated at a pH range of 5 to 8. The differences in germination (%) between all the strains were highly significant but between pH levels they were only nearly significant (P=0.067) with no interaction between pH levels and strains. Mean germination (%) for all tetraploids seems to be slightly higher ( 2%) than that for all diploids, especially at pH's 5, 7 and 8 but this may be due to the significantly longer time ( one day) it took tetraploids to complete germination. The apparent inverse relationship between seed weight and germination (%) was not significant.Mean germination time was highly significant for strains, pH's and their interaction. Increasing mean germination (%) resulted in decreasing mean germination time among strains. Large seeds took less time to germinate especially those from some of the tetraploid strains. This indicates that it is possible to produce a variety with high germination (%), fast germination rate and possibly large seeds. If the marked difference in pH tolerance among strains will prove to be mainly hereditary, then it will be also possible to select for either specific pH tolerance or tolerance at a wide range of pH.  相似文献

The synthesis and processing of the major storage proteins in soybean cotyledons was studied both in vivo and in vitro. The and subunits of 7S as well as the 11S proteins are synthesized as higher molecular weight-precursors on membrane-bound polysomes. The initial translation products of the 7S are proteolytically cleaved during translation suggesting the removal of a signal peptide as evidenced by the presence of 2 and 2 peptides immunoreactive with 7S antibody in the in vitro chain completion products of the membrane-bound polysomes. This is followed or accompanied by cotranslational glycosylation, which increases their size equivalent to that of initially-synthesized precursors. In vivo pulse-labelled 7S and products are of slightly higher molecular weights than the immunoprecipitable chain-completion products, indicating further post-translational modifications. A slow post-translational processing during a period of 1.5 to 16 h yields the final 7S and glycoproteins.Acidic and basic subunits of the 11S protein appear to be synthesized from common large molecular weight (60K-59K) precursors. Antibodies to the 11S acidic component recognize both acidic and basic domains in the precursor while those raised against basic subunits appear to be specific for that region only. The processing of the 11S precursor is also very slow and occurs post-translationally. This slow rate of processing, coupled with a temporal difference in the synthesis of 7S and 11S components, suggests a highly coordinated mechanism for synthesis and packaging of these proteins into protein bodies during seed development.  相似文献

Soybean (Glycine max (L.) Merr.) genotypes varying in area per nodal unit (usually a trifoliolate) and maturity class were grown in plots at the University of Illinois experimental farm. Leaf CO2-exchange rates per unit area (CER) were measured under sunlight on intact plants. In addition to previously reported correlations with specific leaf weight and chlorophyll, CER was positively correlated with ribulose bisphosphate carboxylase (RuBPcase) activity, specific activity, and soluble protein, and was negatively correlated with area per leaf unit. The CER: chlorophyll correlation was destroyed by high CER values in 2 chlorophyll-deficient lines. CER values for 27 of the 35 lines tested fell within the range of those for isolines of cultivar Clark varying in leaf characteristics. The CER values were highest for fully expanded leaves during rapid pod fill. These results suggested that photoperiod (maturity) genes and genes for leaf area growth interact with genes controlling photosynthetic CO2-exchange to produce the major differences in CER values among soybean genotypes.  相似文献

A high level of binding of [3H]methyltrienolone (R1881 = 17beta-hydroxy-17alpha-methyl-estra-4, 9, 11-trien-3-one) was found in cytosol prepared from adrenals of castrated male rats. Binding of [3H]R1881 was of high affinity (DK = 6.2 nM) and highly specific for androgens. The [3H]R1881 complex migrates at 7-9S on sucrose gradients in low ionic strength buffer and at 4-5S in buffer containing 0.4M KC1. All binding studies have been performed in parallel with rat ventral prostate and adrenal cytosol. The present data suggest the presence of an androgen binding component in rat adrenal tissue.  相似文献

The uptake of glycine in rabbit renal brush border membrane vesicles was shown to consist of glycine transport into an intravesicular space. An Na+ electrochemical gradient (extravesicular>intravesicular) stimulated the initial rate of glycine uptake and effected a transient accumulation of intravesicular glycine above the steady-state value. This stimulation could not be induced by the imposition of a K+, Li+ or choline+ gradient and was enhanced as extravesicular Na+ was increased from 10 mM to 100 mM. Dissipation of the Na+ gradient by the ionophore gramicidin D resulted in diminished Na+-stimulated glycine uptake. Na+-stimulated uptake of glycine was electrogenic. Substrate-velocity analysis of Na+-dependent glycine uptake over the range of amino acid concentrations from 25 μM to 10 mM demonstrated a single saturable transport system with apparent Km = 996 μM and Vmax = 348 pmol glycine/mg protein per min. Inhibition observed when the Na+-dependent uptake of 25 μM glycine was inhibited by 5 mM extravesicular test amino acid segregated dibasic amino acids, which did not inhibit glycine uptake, from all other amino acid groups. The amino acids d-alanine, d-glutamic acid, and d-proline inhibited similarly to their l counterparts. Accelerative exchange of extravesicular [3H]glycine was demonstrated when brush border vesicles were preloaded with glycine, but not when they were preloaded with l-alanine, l-glutamic acid, or with l-proline. It is concluded that a single transport system exists at the level of the rabbit renal brush border membrane that functions to reabsorb glycine independently from other groups of amino acids.  相似文献

1. Modification of the Class II sulphydryl groups on the (Na+ + K+)-ATPase from rectal glands of Squalus acanthias with N-ethylmaleimide has been used to detect conformational changes in the protein. The rates of inactivation of the enzyme and the incorporation of N-ethylmaleimide depend on the ligands present in the incubation medium. With 150 mM K+ the rate of inactivation is largest (k1 = 1.73 mM?1 · min?1) and four SH groups per α-subunit are modified. The rate of inactivation in the presence of 150 mM Na+ is smaller (k1 = 1.08 mM?1 · min-1) but the incorporation of N-ethylmaleimide is the same as with K+. 2. ATP in micromolar concentrations protects the Class II groups in the presence of Na+ (k1 = 0.08 mM?1 · min?1 at saturating ATP) and the incorporation id drastically reduced. ATP in millimolar concentrations protects the Class II groups partially in the presence of K+ (k1 = 1.08 mM?1 · min?1) and three SH groups are labelled per α subunit. 3. The K+ -dependent phosphatase is inhibited in parallel to the (Na+ + K+)-ATPase under all conditions, and the ligand-dependent incorporation of N-ethylmaleimide was on the α-subunit only. 4. It is shown that the difference between the Na+ and K+ conformations sensed with N-ethylmaleimide depends on the pH of the incubation medium. At pH 6 there is a very small difference between the rates of inactivation in the presence of Na+ and K+, but at higher pH the difference increases. It is also shown that the rate of inactivation has a minimum at pH 6.9, which suggests that the conformation of the enzyme changes with pH. 5. Modification of the Class III groups with N-ethylmaleimide-whereby the enzyme activity is reduced from about 16% to zero-shows that these groups are also sensitive to conformational changes. As with the Class II groups, ATP in micromolar concentrations protects in the presence of Na+ relative to Na+ or K+ alone. ATP in millimolar concentrations with K+ present increases the rate of inactivation relative to K+ alone, in contrast to the effect on the Class II groups. 6. Modification of the Class II groups with a maleimide spin label shows a difference between Class II groups labelled in the presence of Na+ (or K+) and Class II groups labelled in the presence of K + ATP, in agreement with the difference in incorporation of N-ethylmaleimide. The spectra suggest that the SH group protected by ATP in the presence of K+ is buried in the protein. 7. The results suggest that at least four different conformations of the (Na+ + K+)-ATPase can be sensed with N-ethylmaleimide: (i) a Na+ form of the enzyme with ATP bound to a high-affinity site (E1-Na-ATP); (ii) a Na+ form without ATP bound (E1-Na); (iii) a K+ form without ATP bound (E2-K); and (iv) an enzyme form with ATP bound to a low-affinity site in the presence of K+, probably and E1-K-ATP form.  相似文献

The influence of canopy development in soybean on the survival of corn earworm, Heliothis zea (Boddie) (Lepidoptera: Noctuidae), egg and larval stages and population dynamics of arthropod fauna were evaluated in field trials during 1986–88 in eastern North Carolina. Soybean canopy size decreased as soybean cyst nematode, Heterodera glycines Ichinohe (Nematoda: Heteroderidae), initial population densities increased. Plant species composition of the soybean canopy was affected by weed population densities. Mortality of H. zea larvae due to parasitism and infection with entomopathogens was greater in closed canopy and (or) weedy soybeans than in very open and (or) weed free soybeans. Predation and parasitism of corn earworm eggs were similar across nematode and weed density treatments. Natural enemy populations increased to highest levels during July in closed canopy and (or) weedy soybeans, coinciding with availability of largest prey population reservoirs. A delay in colonization of very open and (or) weed free soybeans by beneficial arthropods until mid to late August allowed greater H. zea larval survival than in closed canopy and (or) weedy soybeans. Arthropod species richness was generally greatest in closed canopy and (or) weedy soybeans during mid to late July, with differences becoming nonsignificant in August and early September. Mean and maximum ambient temperatures were higher and relative humidities lower in open canopy than in closed canopy plots. These conditions were less favorable for development of pathogens and natural enemies.  相似文献

Heterodera glycines was identified in North Carolina in 1954, although symptoms of the disease were noted in the state at least 8 years earlier. Crop rotation experiments designed to develop management systems were initiated in 1956. Two or more years in production of a nonhost crop resulted in decreases of the nematode to low or undetectable levels with acceptable subsequent yields of soybean (Glycine max). Because of almost complete dependence on resistant cultivars and (or) nematicides for nematode control, crop rotation experiments were not conducted from 1962 to 1980. Research on control of H. glycines, beginning in 1981, emphasized biological and ecological aspects of the nematode in order to determine cropping systems that restrict the nematode to nondamaging levels. Mortality during embryogenesis was high at temperatures above 30 C. Hatching of eggs occurs readily in May and June. Postinfection development takes 2-3 weeks at weekly mean temperatures of 22-29 C and is slow above and below those temperatures. Egg production is high during the late growing season. Some cultural practices such as planting early maturing cultivars in mid-to-late June and rotation with a nonhost effectively keeps populations at low levels.  相似文献
