The expression of an Arabidopsis acyl carrier protein (ACP) gene promoter has been examined in transgenic tobacco plants by linking it to the reporter gene -glucuronidase (GUS). Fluorometric analysis showed that the ACP gene promoter was most active in developing seeds. Expression was also high in roots, but significantly lower in young leaves and downregulated upon their maturation. Etiolated and light-grown seedlings showed the same level of GUS activity, indicating that this promoter is not tightly regulated by light. Histochemical studies revealed that expression was usually highest in apical/ meristematic zones of vegetative tissues. Young flowers (ca. 1 cm in length) showed GUS staining in nearly all cell types, however, cell-specific patterns emerged in more mature flowers. The ACP gene promoter was active in the stigma and transmitting tissue of the style, as well as in the tapetum of the anther, developing pollen, and ovules. The results provide evidence that this ACP gene is regulated in a complex manner and is responsive to the array of signals which accompany cell differentiation, and a demand for fatty acids and lipids, during organogenesis. 相似文献

There is an urgent need to develop approaches for repairing the damaged heart, discovering new therapeutic drugs that do not have toxic effects on the heart, and improving strategies to accurately model heart disease. The potential of exploiting human induced pluripotent stem cell (hiPSC) technology to generate cardiac muscle “in a dish” for these applications continues to generate high enthusiasm. In recent years, the ability to efficiently generate cardiomyogenic cells from human pluripotent stem cells (hPSCs) has greatly improved, offering us new opportunities to model very early stages of human cardiac development not otherwise accessible. In contrast to many previous methods, the cardiomyocyte differentiation protocol described here does not require cell aggregation or the addition of Activin A or BMP4 and robustly generates cultures of cells that are highly positive for cardiac troponin I and T (TNNI3, TNNT2), iroquois-class homeodomain protein IRX-4 (IRX4), myosin regulatory light chain 2, ventricular/cardiac muscle isoform (MLC2v) and myosin regulatory light chain 2, atrial isoform (MLC2a) by day 10 across all human embryonic stem cell (hESC) and hiPSC lines tested to date. Cells can be passaged and maintained for more than 90 days in culture. The strategy is technically simple to implement and cost-effective. Characterization of cardiomyocytes derived from pluripotent cells often includes the analysis of reference markers, both at the mRNA and protein level. For protein analysis, flow cytometry is a powerful analytical tool for assessing quality of cells in culture and determining subpopulation homogeneity. However, technical variation in sample preparation can significantly affect quality of flow cytometry data. Thus, standardization of staining protocols should facilitate comparisons among various differentiation strategies. Accordingly, optimized staining protocols for the analysis of IRX4, MLC2v, MLC2a, TNNI3, and TNNT2 by flow cytometry are described. 相似文献

The present work is based on semi-quantitative study carried on detailed sampling (samples are spaced by 5, 10 and 15 cm close to the boundary) of an essentially continuous and expanded section crossing the Cretaceous–Paleogene (K/Pg) boundary in Iran. By this work, we attempt to detail biostratigraphy based on planktonic foraminifera biozones and correlate biozones and subzones with dinocyst events. The entire Cretaceous–Paleogene interval contains rich, diversified and well-preserved planktonic foraminifera and dinoflagellate cyst assemblages. Four planktonic foraminiferal biozones have been recognized across the Cretaceous–Paleogene transition (K/Pg): Abathomphalus mayaroensis Biozone including Plummerita hantkeninoides Subzone from the Late Maastrichtian and Guembelitria cretacea (including Hedbergella holmdelensis and Parvularugoglobigerina longiapertura subzones), Parvularugoglobigerina eugubina Biozone and Parasubbotina pseudobulloides Biozone belonging to the Early Danian. These biozones have been correlated with four dinocyst biozones: the Manumiella seelandica Biozone belonging to the Late Maastrichtian and the Alisocysta reticulata, Senoniasphaera inornata and Damassadinium californicum biozones from the Early Danian. At this section, like at the El Kef section (GSSP for the K/Pg) and the auxiliary sections, an Ir anomaly is detected indicating the K/Pg boundary. This geochemical anomaly coincides also with mass extinctions of planktonic foraminifera species. The extinct species are in particular the large, complex tropical and subtropical taxa dwelling in subsurface and lower photic water. The mass extinctions at the Izeh section occurred over a succinct period of time similar to the K/Pg type section at El Kef (Tunisia). These sudden mass extinctions indicate a catastrophic pattern event occurring at the Maastrichtian/Danian boundary. In contrast the organic-walled dinocysts were less affected by the mass extinction and most species crossed the K/Pg boundary without showing mass and sudden extinctions. Nevertheless, they showed changes in their assemblages’ structure beyond the K/Pg boundary. Especially, Manumiella seelandica and M. druggii, typical species of Antarctic Maastrichtian dinocysts assemblages, occur in coeval deposits at the Izeh section; they persist through the Lower Danian and, like in Tunisia (e.g., El Kef section, Ellès section) show an obvious increase in relative abundance. 相似文献

Summary A series of phosphopeptides Tyr(PO3H2)-Val-Pro-Xxx-Leu (Xxx=Met, Met(O), Nle, Dab or Cys), derived from the native platelet-derived growth factor- receptor (PDGF-) sequence, has been prepared to study their interaction with the src-homology 2 (SH2) domains of the p85 subunit of PI3 kinase. The phosphopeptides were synthesized using Fmoc methodology incorporating N-Boc dibenzyl-protected phosphotyrosine (Boc-Tyr[PO3(Bzl)2]) as the N-terminal amino acid, since the benzyl groups can be removed during resin cleavage with TFA. Only peptides containing methionine were found to exist partially as S-benzyl sulfonium salts after TFA cleavage from the resin. Our interest in thecis-acting elements that promote the up-regulation of the globin gene has led to a systematic deletion analysis of portions of the globin gene in the context of the HS2 and globin gene using transgenic mice. In constructs that delete the 5 region to only 265 bp, high-level erythroid-specific expression was observed. Further deletion to 122 bp, however, results in significantly reduced expression levels A substitution of a minilocus control region for the single HS2 site was also produced, resulting in increased globin expression over that seen with the HS2 alone. These results are consistent with the presence of an enhancer-like element between –122 and –265. In addition, a construct in which the entire globin gene promoter was replaced by a thymidine kinase promoter was tested. Interestingly, no expression was detected in these transgenic mice. This may indicate the requirement for an erythroid-specific promoter to drive this gene. Finally, the 3 region of the globin gene was deleted in order to examine the effect of a previously defined 3 enhancer region. With deletion of this region, the expression of the human globin gene in transgenic mice is unchanged relative to the parental constructs. 相似文献

In several psammophine snake species, small sense-organ-like pits have been discovered occurring on the top of the head. These pits show a heretofore non-described structure. In numbers of one to four per head, they are present in Dromophis lineatus, Malpolon monspessulanus, several Psammophis species and Rhamphiophis rubropunctatus, but not in all individuals which is unrelated to sex. The reason for their sporadic occurrence remains obscure. Their equally obscure function is discussed in relation to the presumed functions of already known upper-head 'pits' in snakes.
