The effects of application of two arbuscular mycorrhizal (AM) fungi Glomus macrocarpum and G. fasciculatum on shoot biomass and concentration of essential oil in Anethum graveolens L. and Trachyspermum ammi (Linn.) Sprague fruits were evaluated. Results revealed significant variation in effectiveness of the two AM fungal species. AM fungal inoculation in general improved the growth of the plants. On mycorrhization, the concentration of essential oil increased up to 90% in dill and 72% in carum over their respective controls. Glomus macrocarpum was more effective than G. fasciculatum in enhancing the oil concentration. The constituents of the essential oils were characterized by gas liquid chromatography. The levels of limonene and carvone were enhanced in essential oil obtained from G. macrocarpum-inoculated dill plants, while G. fasciculatum inoculation resulted in a higher level of thymol in carum.  相似文献

Summary Arbuscular mycorrhizal (AM) fungi significantly improve plant growth in soils with low phosphorus availability and cause many changes in root morphology, similar to those produced by increased P nutrition, mainly depending on root apex size and activity. The aim of this work was to discriminate between the morphogenetic role of AM fungi and P in leek (Allium porrum L.) by feeding mycorrhizal and nonmycorrhizal plants with two nutrient solutions containing 3.2 or 96 M P and examining specific parameters related to adventitious root apices (apex size, mitotic cycle, and RNA synthesis). The results showed that AM fungi blocked meristem activity as indicated by the higher percentages of inactive apices and metaphases in the apical meristem of mycorrhizal plants, whereas the high P supply lengthened the mitotic cycle without blocking the apices, resulting in steady, slow root growth. Pisum sativum L. myc mutants which fail to form arbuscular mycorrhiza have recently been identified amongst nod mutants (Duc et al., 1989, Plant Sci. 60, 215–222). The reason for this resistance to symbiotic fungi has been investigated in the case of a locus a mutant (P2) inoculated with Glomus mosseae (Nicol. and Gerd.) Gerd, and Trappe. The fungal symbiont formed viable appressoria in contact with the root surface but its development was stopped at the root epidermis. Abundant material was deposited on the inner face of root cell walls adjacent to the appressoria in the P2 mutant, but not in the wild-genotype parent cultivar (Frisson) forming a symbiotic mycorrhizal infection. Fluorescence, histochemical, cytochemical and immunocytological approaches were used to characterize the paramural deposits in epidermal and hypodermal cells of the mutant. Strong fluorescence under blue light indicated the accumulation of phenolic compounds although polymers like lignin or suberin were not localized. Proteins and glycoproteins were homogeneously distributed within the paramural deposits. In the latter, the periodic acid-thiocarbohydrazide-silver proteinate (PATAg) reaction for 1,4-polysaccharide detection showed a heterogeneous composition with electron-dense points surrounded by non-reactive material, but cytological tests for cellulose and pectin gave weak responses as compared to epidermal and hypodermal walls of the wild genotype. -1,3-Glucans indicative of callose were detected by in-situ immunolocalization in the paramural deposits below appressoria on mutant roots, but not in walls of the wild genotype. Thus, appressorium formation by G. mosseae on roots of the locus a P. sativum mutant elicits wall modifications usually associated with activation of defence responses to pathogens. It is proposed that this locus must be involved in a key event in symbiotic infection processes in P. sativum, and the possible role of complex regulatory interactions between symbiosis and defence genes in endomycorrhiza development is discussed.Abbreviations DAPI 4,6-diamino-2-phenylindole - FDA fluo-rescein diacetate - PATAg periodic acid-thiocarbohydrazide-silver proteinate The authors are grateful to C. The effects of inoculum forms (single-spore, multi-spores, or colonized root pieces) and host plants (Nicotiana tabacum L., Sorghum sudanense(Piper) Stapf, and Trifolium repens L. ) on the development and inoculum potential (IP) of the arbuscular mycorrhizal fungi (AMF): Glomus macrocarpum Tul & Tul, Glomus mosseae (Nicol & Gerd. ) Gerdemann & Trappe, Glomus versiforme (Karsten) Berch, and Sclerocystis sinuosa Gerdemann & Bakhi cultured in pots were investigated. The lag phase of treatment with 50 spores or 0.5 g (fresh weight) of colonized root pieces was 4 weeks, much shorter than that of the treatment with 1 spore (8 weeks); the value of IP(VIP) and percentage of root colonization(PRC) of the former were greater than those of the latter. Only on the early stages of colonization was there difference between the 50 spores and the 0.5 g (fresh weight) of colonized root piece inoculation treatments. The IP per plant inoculated with 0. 5 g (fresh weight) of colonized mot pieces of AMF was greater than that of the other two treatments except G. vers/forme on Nicotiana tabacum, while the PRC of the plants inoculated with 50 spores and 0. 5 g (fresh weight) of colonized root pieces of AMF was higher than that of the 1 spore inoculation after 10 weeks. The VIP of AMF on Trifolium repens was significantly higher than that on the other two hosts. The VIP of G. mosseae, G. versiforme, and S. sinuosa was respectively greater than that of G. macrocarpum. This suggested that different species of AMF produced different VIP of the inoculum. Nicotiaha tabacum was much better than the other host plants which used to be inoculated with single spore, and to produce inocula of AMF.  相似文献

In split-root systems of alfalfa (Medicago sativa L.), already existing nodules or arbuscular mycorrhizal roots suppress further establishment of symbiosis in other root parts, a phenomenon named autoregulation. Roots treated with rhizobial nodulation signals (Nod factors) induce a similar systemic suppression of symbiosis.In order to test the hypothesis that flavonoids play a role in this systemic suppression, split-root systems of alfalfa plants were inoculated on one side of the split-root system with Sinorhizobium meliloti or Glomus mosseae or were treated with Nod factor. HPLC-analysis of alfalfa root extracts from both sides of the split-root system revealed a persistent local and systemic accumulation pattern of some flavonoids associated with the different treatments. The two flavonoids, formononetin and ononin, could be identified to be similarily altered after rhizobial or mycorrhizal inoculation or when treated with Nod factor.Exogenous application of formononetin and ononin partially restored nodulation and mycorrhization pointing towards the involvement of these two secondary compounds in the autoregulation of both symbioses.  相似文献

Our goal was to evaluate the mycorrhizal colonization, as well as the density of arbuscular mycorrhizal (AM) fungal spores, in Eucalyptus camaldulensis and E. grandis monocultures at 2 years in a semiarid part of Brazil. Soil and root samples were collected in 2 consecutive years. Eucalyptus camaldulensis showed varied AM colonization level according to season of sampling, and Glomus was dominant in spore numbers. Eucalyptus grandis showed dominant ectomycorrhizal (ECM) colonization and lower AM fungal spore density. Overall results suggest that E. camaldulensis has both AM and ECM dependencies, whereas E. grandis is solely ECM dependent in the monocultures.  相似文献

 Mycorrhizal and nonmycorrhizal roots of Allium schoenoprasum were tested for activities of α-mannosidase, β-glucosidase and arabinosidase. Mannosidase activity was higher by a factor of two in mycorrhizal than in nonmycorrhizal root extracts. The apparent molecular weight of the enzyme was 152 kDa and its KM was 1.25 mM in colonized roots and 1.85 mM in uncolonized roots. α-Mannosidase activity was further characterized by an acid pH optimum and Zn2+ dependency. No significant differences could be found between mycorrhizal and nonmycorrhizal roots for β-glucosidase and arabinosidase activities. Accepted: 28 August 1995  相似文献

In late‐successional environments, low in available nutrient such as the forest understory, herbaceous plant individuals depend strongly on their mycorrhizal associates for survival. We tested whether in temperate European forests arbuscular mycorrhizal (AM) woody plants might facilitate the establishment of AM herbaceous plants in agreement with the mycorrhizal mediation hypothesis. We used a dataset spanning over 400 vegetation plots in the Weser‐Elbe region (northwest Germany). Mycorrhizal status information was obtained from published resources, and Ellenberg indicator values were used to infer environmental data. We carried out tests for both relative richness and relative abundance of herbaceous plants. We found that the subset of herbaceous individuals that associated with AM profited when there was a high cover of AM woody plants. These relationships were retained when we accounted for environmental filtering effects using path analysis. Our findings build on the existing literature highlighting the prominent role of mycorrhiza as a coexistence mechanism in plant communities. From a nature conservation point of view, it may be possible to promote functional diversity in the forest understory through introducing AM woody trees in stands when absent.  相似文献

Arbuscular mycorrhizal fungi (phylum Glomeromycota) are among the oldest and most successful symbionts of land plants. With no evidence of sexual reproduction, their evolutionary success is inconsistent with the prediction that asexual taxa are vulnerable to extinction due to accumulation of deleterious mutations. To explore why Glomeromycota defy this prediction, we studied ribosomal RNA (rRNA) gene evolution in the Claroideoglomus lineage and estimated effective population size, Ne, in C. etunicatum. We found that rRNA genes of these fungi exhibit unusual and complex patterns of molecular evolution. In C. etunicatum, these patterns can be collectively explained by an unexpectedly large Ne combined with imperfect genome‐wide and population‐level rRNA gene repeat homogenization. The mutations accumulated in rRNA gene sequences indicate that natural selection is effective at purging deleterious mutations in the Claroideoglomus lineage, which is also consistent with the large Ne of C. etunicatum. We propose that in the near absence of recombination, asexual reproduction involving massively multinucleate spores typical for Glomeromycota is responsible for the improved efficacy of selection relative to drift. We postulate that large effective population sizes contribute to the evolutionary longevity of Glomeromycota.  相似文献
