Candidate plus trees (CPTs) of Pongamia pinnata , a potential biodiesel plant occurring across 10 locations in North Guwahati, were identified based on morphological markers (vegetative and reproductive) using combined analysis over locations. Identified CPTs were then multiplied using seed propagation technique in a nursery bed. The performance of the candidate trees with respect to seed and pod traits, the two most important characters with regard to oil, were evaluated using CROPSTAT software for inferring potential genotypes that can be included in programmes aimed at genetic improvement of the species. Total oil content from the seeds of plus trees was also analysed using solvent extraction procedure at their boiling points. Hexane extraction yielded maximum oil content from seeds (33%) compared with petroleum ether (30%). When the seed to solvent ratio varied, no significant difference was noticed on the total oil yield for an individual tree, although the recovery of solvent and the time taken for oil extraction were significantly reduced at higher ratios of solvent used.  相似文献

We investigated the relationships between vegetation area, edge length, and mammals in the urban woods of Daejeon Metropolitan City, South Korea. The vegetation patches included in this study varied from 2.1 to 501.0?ha in size. Surveys were conducted monthly between February and October 2015, with a survey route measuring 1?km in length and 10?m width established in each forest patch. Field signs of 14 species of mammals were recorded in the 33 chosen forest patches over the course of the study period, and the number of species in each patch varied from 2 to 11. Mammal species richness was positively correlated with vegetation area, and field sign frequency was positively correlated with vegetation area and negatively correlated with edge length. The field sign frequencies of large moles Mogera robusta, Siberian chipmunks Tamias sibiricus, leopard cats Prionailurus bengalensis, Korean hares Lepus coreanus, water deer Hydropotes inermis, and wild boars Sus scrofa were positively correlated with vegetation area. Moreover, that of large moles, leopard cats, Korean hares, and water deer were negatively correlated with edge length. Remnant vegetation area and edge length are the primary determinants of mammal species richness and field sign frequency in urban woods, highlighting the importance of vegetation patch size for mammal conservation in fragmented urban landscapes.  相似文献

Having confirmed that adenovirus-mediated overexpression of NH2-tau fragment lacking the first 25 aminoacids evokes a potent neurotoxic effect, sustained by protracted stimulation of NMDA receptors, in primary neuronal cultures we investigated whether and how chemically synthesized NH2-derived tau peptides, i.e. NH2-26-44 and NH2-1-25 fragments, affect mitochondrial function. We tested both fragments on each step of the processes leading to ATP synthesis via oxidative phosphorylation: i) electron flow via the respiratory chain from physiological substrates to oxygen with the activity of each individual complex of the respiratory chain investigated in some detail, ii) membrane potential generation arising from externally added succinate and iii) the activity of both the adenine nucleotide translocator and iv) ATP synthase. Oxidative phosphorylation is not affected by NH2-1-25 tau fragment, but dramatically impaired by NH2-26-44 tau fragment. Both cytochrome c oxidase and the adenine nucleotide translocator are targets of NH2-26-44 tau fragment, but adenine nucleotide translocator is the unique mitochondrial target responsible for impairment of oxidative phosphorylation by the NH2-26-44 tau fragment, which then exerts deleterious effects on cellular availability of ATP synthesized into mitochondria.  相似文献

Skeletal muscle can regenerate repeatedly due to the presence of resident stem cells, called satellite cells. Because satellite cells are usually quiescent, they must be activated before participating in muscle regeneration in response to stimuli such as injury, overloading, and stretch. Although satellite cell activation is a crucial step in muscle regeneration, little is known of the molecular mechanisms controlling this process. Recent work showed that the bioactive lipid sphingosine-1-phosphate (S1P) plays crucial roles in the activation, proliferation, and differentiation of muscle satellite cells. We investigated the role of growth factors in S1P-mediated satellite cell activation. We found that epidermal growth factor (EGF) in combination with insulin induced proliferation of quiescent undifferentiated mouse myoblast C2C12 cells, which are also known as reserve cells, in serum-free conditions. Sphingosine kinase activity increased when reserve cells were stimulated with EGF. Treatment of reserve cells with the D-erythro-N,N-dimethylsphingosine, Sphingosine Kinase Inhibitor, or siRNA duplexes specific for sphingosine kinase 1, suppressed EGF-induced C2C12 activation. We also present the evidence showing the S1P receptor S1P2 is involved in EGF-induced reserve cell activation. Moreover, we demonstrated a combination of insulin and EGF promoted activation of satellite cells on single myofibers in a manner dependent on SPHK and S1P2. Taken together, our observations show that EGF-induced satellite cell activation is mediated by S1P and its receptor.  相似文献

Background

Extracorporeal photopheresis that exposes isolated white blood cells to 8-methoxypsoralen (8-MOP) and ultraviolet-A (UV-A) light is used for the management of cutaneous T-cell lymphoma and graft-versus-host disease. 8-MOP binds to DNA of both tumor and normal cells, thus increasing the risk of carcinogenesis of normal cells; and also kills both tumor and normal cells with no selectivity after UV-A irradiation. Hexaminolevulinate (HAL)-induced protoporphyrin-IX is a potent photosensitizer that localizes at membranous structures outside of the nucleus of a cell. HAL-mediated photodynamic therapy selectively destroys activated/transformed lymphocytes and induces systemic anti-tumor immunity. The aim of the present study was to explore the possibility of using HAL instead of 8-MOP to kill cells after UV-A exposure.

Methods

Human T-cell lymphoma Jurkat and Karpas 299 cell lines were used to evaluate cell photoinactivation after 8-MOP and/or HAL plus UV-A light with cell proliferation and long term survival assays. The mode of cell death was also analyzed by fluorescence microscopy.

Results

Cell proliferation was decreased by HAL/UV-A, 8-MOP/UV-A or HAL/8-MOP/UV-A. At sufficient doses, the cells were killed by all the regimens; however, the mode of cell death was dependent on the treatment conditions. 8-MOP/UV-A produced apoptotic death exclusively; whereas both apoptosis and necrosis were induced by HAL/UV-A.

Conclusion

8-MOP can be replaced by HAL to inactivate the Jurkat and Karpas 299 T-cell lymphoma cells after UV-A irradiation via apoptosis and necrosis. This finding may have an impact on improved efficacy of photopheresis.  相似文献

白菜叶裂数性状主基因+多基因遗传分析   总被引:1,自引:0,他引:1  
以叶片全缘的大白菜自交系(P1)和叶缘深裂的欧洲白菜型油菜自交系(P2)杂交所获得的6个基本世代(P1、P2、F1、B1、B2、F2)为材料,应用主基因+多基因混合遗传模型对白菜叶裂数进行遗传分析。结果表明,白菜的叶裂数受2对加性-显性-上位性主基因+加性-显性多基因的控制,第1对主基因加性效应为-1.154 7,显性效应为-1.516 8;第2对主基因的加性效应为-1.154 8,显性效应为1.034 9;多基因加性效应为0.591 9,显性效应为1.145 2,2对主基因间存在明显的交互作用。B1、B2和F2世代叶裂数的主基因遗传率分别为88.48%、90.40%、93.03%;多基因遗传率分别为4.114%、0、0。B1、B2、F2世代叶裂数表现出较高的主基因遗传率,受环境影响较小。在白菜叶裂数性状的改良中应以主基因为主,并适于早代选择。  相似文献

A desensitized aspartate kinase (AK) gene has been developed as a non-antibiotic selection marker for use in the production of transgenic chickpea (Cicer arietinum L.). Transgenic shoots regenerated from embryo explants bombarded with the desensitized AK gene were selected on media containing two amino acids, lysine and threonine (LT). Approximately 15% of the putative transgenic shoots of vars. P-362 and P-1042 survived after 4 weeks of growth on MSB5 medium (MS mineral salts and B5 vitamins) containing 2 µM thidiazuron (TDZ) and 2 mM lysine and 2 mM threonine. These shoots were subsequently grown on MSB5 medium supplemented with 2 µM TDZ and 5 mM lysine and 5 mM threonine, and nearly 1% continued to grow after 16 weeks of selection. A phosphinothricin (PPT) selection system for Agrobacterium-mediated chickpea transformation was also developed. Three varieties of chickpea, P-362, P-1042 and P-1043, were successfully used for Agrobacterium transformation. Following Agrobacterium infection, 3-8% of the regenerated shoots remained green and continued to grow on MSB5 medium supplemented with 2.5 mg l-1 PPT. Increasing the concentrations of PPT to 15 mg l-1 reduced transgenic shoot production in P-362, P-1042 and P-1043 to 0.7%, 1.2% and 1.1%, respectively. Selected putatively transformed shoots of all three varieties were rooted and grown to maturity. Southern hybridization analysis revealed single as well as multiple integration of genes in selected transgenic lines. 以果实性状差异较大的一年生辣椒材料C.annuum B9431(P1)和灌木辣椒材料C.frutescens H108(P2)为亲本,构建4世代群体(P1、P2、F1、F2),应用数量性状主基因+多基因混合遗传模型方法对辣椒6个果实性状进行遗传分析,为辣椒果实性状QTL定位及分子标记辅助育种研究奠定理论基础。结果表明:6个果实性状均符合2对主基因+多基因遗传模型。其中,单果重量、果实纵径、果实横径、果形指数和果肉厚度5个性状均符合E-0模型,即2对加性-显性-上位性主基因+加性-显性-上位性多基因混合遗传模型;果柄长度最佳遗传模型为E-5,即2对完全显性主基因+加性-显性多基因模型。单果重量、果实纵径、果实横径、果形指数、果肉厚度和果柄长度主基因遗传率分别为87.64%、37.67%、82.46%、94.82%、83.33%和40.00%,多基因遗传率分别为7.50%、54.56%、10.53%、0.27%、12.96%和37.78%。  相似文献
