Alcohol-extractable, hydrophobic zein proteins contaminate starch granule surfaces and can be removed by enzymatic digestion with thermolysin. The goal of this research was to find practical alternatives to thermolysin that might be used during the corn wet-milling process. All of the commercial thermostable alkaline proteases studied (SP 709, Neutrase, and Spezyme FAN) removed the zein proteins from various types of cornstarch, as demonstrated by the lack of protein bands below 30 kDa under the reducing conditions of SDS-PAGE gel. Each enzyme removed the zein proteins as effectively as thermolysin removed them. However, the removal of the zein protein did not reduce the quantity of free fatty acids associated with the starch. Eighty-two cultures of Fusarium species isolated in 1986 from moldy maize in Minnesota were each cultured on rice for 4 weeks and found to produce the following mycotoxins: F. graminearum isolates, deoxynivalenol (DON, 4–225 g/g), 3-acetyldeoxynivalenol (3-ADON, 2–4g/g), 15-acetyldeoxynivalenol (15-ADON, 1–35 g/g) and zearalenone (ZEA, 5–4350 g/g); F. moniliforme, fusarin C (detectable amounts to 1000 g/g); F. mòniliforme, F. oxysporum, F. proliferatum and F. subglutinans isolates, moniliformin (15–6775 g/g); F. moniliforme, F. proliferatum, and F. subglutinans isolates, fusaric acid (detectable amounts). Other mycotoxins screened for in each rice sample and not detected were T-2 toxin, HT-2 toxin, neosolaniol, T-2 tetraol, nivalenol, fusarenon-X, scirpenols, alpha and beta trans-zearalenols, wortmannin, and fusarochromanone. The rat feeding bioassay indicated that other, unidentified toxins may be present.  相似文献

Roots of ten-days-old seedlings obtained from a maize hybrid grown in complete or in sulphate-deprived medium were used to extract Poly(A)+RNA. The response to sulphate deprivation, which is known to increase the uptake capacity up to ten times, was manifested also by the expression of three mRNA species, as shown by the in vitro translation of the mRNA population. Studies were undertaken to evaluate the effects of mechanical impedance on root exudation by maize (Zea mays L., var Dea) and to examine the importance of these effects in relation to the stage of plant development. Plants were grown under sterile and hydroponic conditions. Mechanical impedance was simulated using glass beads of 1 mm diameter. This treatment was compared with a control without beads. Results demonstrated that plant growth was influenced by mechanical impedance. Mechanical impedance markedly affected the growth of the shoot, whether this was measured as leaf area or total dry matter. Besides increasing root/shoot biomass ratios, mechanical impedances also stimulated root exudation of organic and inorganic compounds. Stressed plants lost more nitrogenous compounds than control plants. Otherwise, the percentage of released carbon decreased. Depending on the developmental stage of the plant, there was a large variation in the magnitude and time course on mechanical impedance effects. The effects of mechanical impedance persist and accentuate with time.  相似文献

Ammonium sulfate (5 mM) had no effect on nitrate reductase activity during a 3 hr dark incubation, but the enzyme was increased 2.5-fold during a subsequent 24 hr incubation of the maize leaves in light. The enzyme activity induced by ammonium ion declined at a slower rate under non-inducing conditions than that induced by nitrate. The decline in ammonium stimulated enzyme activity in the dark was also slower than that with nitrate. Further. cycloheximide accelerated the dark inactivation of the ammonium-enzyme while it had no effect on the nitrate-enzyme. The experiments demonstrate that increase in nitrate reductase activity by ammonium ion is different from the action of nitrate action.  相似文献

Generalized estimating equations (GEE) are extension of generalized linear models (GLM) widely applied in longitudinal data analysis. GEE are also applied in spatial data analysis using geostatistics methods. In this paper, we advocate application of GEE for spatial lattice data by modeling the spatial working correlation matrix using the Moran's index and the spatial weight matrix. We present theoretical developments and results for simulated and actual data as well. For the former case, 1,000 samples of a random variable (response variable) defined in (0, 1) interval were generated using different values of the Moran's index. In addition, 1,000 samples of a binary and a continuous variable were also randomly generated as covariates. In each sample, three structures of spatial working correlation matrices were used while modeling: The independent, autoregressive, and the Toeplitz structure. Two measures were used to evaluate the performance of each of the spatial working correlation structures: the asymptotic relative efficiency and the working correlation selection criterions. The results showed that both measures indicated that the autoregressive spatial working correlation matrix proposed in this paper presents the best performance in general. For the actual data case, the proportion of small farmers who used improved maize varieties was considered as the response variable and a set of nine variables were used as covariates. Two structures of spatial working correlation matrices were used and the results showed consistence with those obtained in the simulation study.  相似文献

We have studied hydrotropism and its interaction with gravitropism in agravitropic roots of a pea mutant and normal roots of peas (Pisum sativum L.) and maize (Zea mays L.). The interaction between hydrotropism and gravitropism in normal roots of peas or maize were also examined by nullifying the gravitropic response on a clinostat and by changing the stimulus-angle for gravistimulation. Depending on the intensity of both hydrostimulation and gravistimulation, hydrotropism and gravitropism of seedling roots strongly interact with one another. When the gravitropic response was reduced, either genetically or physiologically, the hydrotropic response of roots became more unequivocal. Also, roots more sensitive to gravity appear to require a greater moisture gradient for the induction of hydrotropism. Positive hydrotropism of roots occurred due to a differential growth in the elongation zone; the elongation was much more inhibited on the moistened side than on the dry side of the roots. It was suggested that the site of sensory perception for hydrotropism resides in the root cap, as does the sensory site for gravitropism. Furthermore, an auxin inhibitor, 2,3,5-triiodobenzoic acid (TIBA), and a calcium chelator, ethyleneglycol-bis-(-aminoethylether)-N,N,N,N- tetraacetic acid (EGTA), inhibited both hydrotropism and gravitropism in roots. These results suggest that the two tropisms share a common mechanism in the signal transduction step.  相似文献

The effect of intercropped legumes and three N fertilizer rates in a continuous maize (Zea mays L.) cropping system on the physical properties of two soils were investigated for three years. The legumes, being a mixture of alfalfa, clover and hairy vetch, had a significant cumulative effect on some physical properties of both soil. The lowest stability and smallest mean weight diameter of soil aggregates were associated with monoculture maize plots. Aggregate size and stability were not affected by N fertilization at any of the rates of 0, 70, and 140 kg ha-1 in intercropped plots, except that aggregate stability was actually reduced by N fertilization in one soil, the Ste. Rosalie clay. In maize plots in both soils, stability and size of soil aggregates were significantly increased with increased added N. Intercropped legumes significantly decreased dry bulk density and soil penetration resistance. Added N had no measurable influence on these compaction factors. Soil water properties were not significantly affected by either intercropping or N fertilization. Positive effects noted on soil aggregation and other physical properties in intercropped plots are the result of enhanced root activity, or incorporation of legumes as green manure, or both. Improvement of soil structure in maize plots associated with increasing N application was the result of increased maize-root residues.  相似文献
