展示酶的酵母细胞既具有固定化酶的优点,又有制备简单、成本较低的特点.采用表面展示南极假丝酵母脂肪酶B (Candida antarctica lipase B,CALB)的毕赤酵母细胞催化合成己二酸二异辛酯(Diisooctyl adipate,DIOA),对该反应体系进行优化,并实现了初步工艺放大制备.经条件优化后,在10mL反应体系中,DIOA的产率可达85.0%.该工艺放大到200mL反应体系时,DIOA产率可达97.8%.经减压蒸馏,DIOA纯度可达到98.2%.该酵母表面展示脂肪酶在合成绿色润滑油己二酸二异辛酯中具有良好应用前景. 相似文献

An oil-degrading bacterium, Acinetobacter lwoffi, isolated by elective culture from the Medway estuary, utilized an ester-based synthetic lubricating oil EMKARATE DE 155 as sole carbon and energy source. Analysis of culture supernatants by gas chromatography showed the accumulation of a nondegradable metabolite 1,1,1 Tris (hydroxymethyl) propane in addition to two metabolizable fatty acids, octanoic and decanoic acids as products of the synthetic oil degradation. Esterase activities were subsequently demonstrated in oil and acetate-grown cells. The synthetic oil therefore appears to be partially biodegradable in the environment. 相似文献

Preliminary characterization of a biosurfactant-producing Azotobacter chroococcum isolated from marine environment showed maximum biomass and biosurfactant production at 120 and 132 h, respectively, at pH 8.0,
38°C, and 30‰ salinity utilizing a 2% carbon substrate. It grew and produced biosurfactant on crude oil, waste motor lubricant
oil, and peanut oil cake. Peanut oil cake gave the highest biosurfactant production (4.6 mg/mL) under fermentation conditions.
The biosurfactant product emulsified waste motor lubricant oil, crude oil, diesel, kerosene, naphthalene, anthracene, and
xylene. Preliminary characterization of the biosurfactant using biochemical, Fourier transform infrared spectroscopy, and
mass spectral analysis indicated that the biosurfactant was a lipopeptide with percentage lipid and protein proportion of

Criteria selected for screening of biosurfactant production by Bacillus megaterium were hemolytic assay, bacterial cell hydrophobicity and the drop-collapse test. The data on hemolytic activity, bacterial
cell adherence with crude oil and the drop-collapse test confirmed the biosurfactant-producing ability of the strain. Accordingly,
the strain was cultured at different temperatures, pH values, salinity and substrate (crude oil) concentration in mineral
salt medium to establish the optimum culture conditions, and it was shown that 38°C, 2.0% of substrate concentration, pH 8.0
and 30‰ of salt concentration were optimal for maximum growth and biosurfactant production. Laboratory scale biosurfactant
production in a fermentor was done with crude oil and cheaper carbon sources like waste motor lubricant oil and peanut oil
cake, and the highest biosurfactant production was found with peanut oil cake. Characterization of partially purified biosurfactant
inferred that it was a glycolipid with emulsification potential of waste motor lubricant oil, crude oil, peanut oil, diesel,
kerosene, naphthalene, anthracene and xylene. 相似文献

高效降解机油微生物的筛选及除油效果初探 总被引:7,自引:0,他引:7
从多处受石油及其制品污染的土壤中筛选到六株对高浓度机油等相关石油制品具有降解能力的微生物菌种。对该六株菌种的单独和混合降解机油的效果进行了研究,并考察了营养成分对机油降解的影响。实验表明:Px01菌种具有最强的除油能力,在合适条件下,Px01培养液的机油降解速率可达0.502g/L.d。混合菌的除油效果明显好于单菌,最高降解速率可到0.64g/L.d。混合菌的营养要求较低,添加有机营养元素对混合微生物的生长和机油降解没有显著的促进作用。由于所筛选驯化得到的微生物菌种针对高浓度含机油废水的降油效果显著,因此具有良好的应用前景。 相似文献

The purpose of this work was to evaluate and compare the functionality of bovine fatty acids-derived (MgSt-B) and vegetable
fatty acids-derived (MgSt-V) magnesium stearate powders when used for the lubrication of granules prepared by high-shear (HSG)
and fluid bed (FBG) wet granulation methods. The work included evaluation of tablet compression and ejection forces during
tabletting and dissolution testing of the compressed tablets. Granules prepared by both granulation methods required significantly
lower ejection force (p < 0.01) when lubricated with the MgSt-V powder as compared to those lubricated with the MgSt-B powder. Granules prepared
by the HSG method and lubricated with the MgSt-V powder also required significantly lower compression force (p < 0.01) to produce tablets of similar weight and hardness as compared to those lubricated with the MgSt-B powder. The dissolution
profiles were not affected by these differences and were the same for tablets prepared by same granulation method and lubricated
with either magnesium stearate powder. The results indicate significant differences (p < 0.01) between lubrication efficiency of the MgSt-B and the MgSt-V powders and emphasize the importance of functionality
testing of the MgSt powders to understand the impact of these differences.

This work investigates the effect of excipient particle size on compaction properties of brittle, plastic and viscoelastic
materials with and without added lubricants. Sieve cuts of Microcrystalline cellulose (MCC), Starch and Dibasic calcium phosphate
dihydrate were obtained by sieving, then samples were tested without lubrication or with added lubricant (0.5% Mg stearate
mixed for either 5 or 30-min). Compacts were left overnight before testing. It was found that in the absence of lubricant,
compact tensile strength (TS) was dependent on particle size only for starch. With Mg stearate, lubricant sensitivity shows
a strong dependence on excipient particle size for both starch and MCC, where smaller particles are less affected by lubricant. Dibasic calcium phosphate dihydrate was not sensitive to lubricant
even after 30 min mixing. This study highlights that in the absence of lubricant, initial particle size of excipients has
no impact on compact strength not only for Dibasic calcium phosphate dihydrate (brittle), but also for MCC (plastic). On the
other hand, TS is dependent on particle size both with or without added lubricant for starch (viscoelastic). 相似文献

Objectives: To assess the extent of lubricant use by smear-takers and the effect of lubricant contamination of ThinPrep® processed cervical cytology samples.
Methods: All primary care smear-takers were sent a questionnaire on lubricant type and frequency of use. Fifty cervical cytology samples were then contaminated with incremental amounts of K-Y® jelly, 50 samples contaminated with incremental amounts of Aquagel® and ten non-contaminated vials were processed using the ThinPrep® T2000 processor followed by Papanicolaou staining. The morphological appearances of lubricant contamination were described microscopically and formal cell counts performed on all slides.
Results: Seventy of 94 (74.5%) primary care smear-takers indicated lubricant use of whom 9/70 (12.8%) used Aquagel® and 61/70 (87.2%) used K-Y® jelly. K-Y® jelly appeared as mucoid blue deposits in the slide background whereas Aquagel® appeared as pink stringy background material. Cell counting showed a significant difference between Aquagel® and K-Y® jelly contaminated slides compared to the original non-contaminated preparations for all fields and the average fields ( P < 0.001) with a significantly higher count for the original non-contaminated slides than the lubricant contaminated groups.
Conclusion: Lubricant contamination of ThinPrep® cervical cytology samples may result in reduced cellularity of the subsequent slide. This study provides evidence-based data to support British Society for Clinical Cytology recommendations for no lubricant use when taking cervical samples. 相似文献
