A simple apparatus was developed to allow 12 petri plates to be poured simultaneously by hand. It was used when screening bacterial isolates from sewage and dog feces for their ability to detect phages from these sources. This was done to assess the ease with which source-specific phage hosts can be isolated from these sources of fecal pollution. Host bacteria that consistently detected phages from sewage were easily isolated from sewage. These bacterial isolates did not detect phages from dog feces. Host bacteria were not isolated from dog feces even after screening hundreds of colonies from fecal samples from six dogs. Journal of Industrial Microbiology & Biotechnology (2000) 24, 124–126. A photosensitive chemical oscillating reaction, i.e., the Briggs-Rauscher (B.R.) reaction, exhibiting a wealth of nonlinear behavior, when performed in a continuous-flow stirred-tank reactor, and subjected to periodic light irradiation, is studied as an experimental example of entrainment phenomena observable in biological systems. The adaptation patterns under periodic light irradiation are elucidated by means of the response of the system to continuous and single-pulse light irradiation. It is shown that self-oscillating states, excitable steady states and bistable systems can exhibit the same types of synchronization patterns when submitted to periodic external forces with appropriate amplitude and time scale conditions.  相似文献

The influence of short draft tubes covered by perforated plates on gas-liquid mass transfer was examined in external-loop airlift bioreactors. The volumetric mass transfer coefficients in a model external-loop airlift bioreactor were measured with water and non-Newtonian media. It was found that introduction of draft tubes covered with perforated plates in the riser significantly improved the mass transfer rate, particularly in higher viscous non-Newtonian fermentation media. The enhancement of mass transfer rate might be due mainly to an increase in bubble coalescence and redispersion. (c) 1994 John Wiley & Sons, Inc.  相似文献

本文主要阐述了一种具有纤溶活性的枯草杆菌(Bacillussubtilis)蛋白激酶产生菌株的筛选与鉴定的研究结果。作者从初筛的12株Bacillussublilis菌中,通过对固体发酵和液体发酵所产生的枯草杆菌蛋白激酶,用琼脂糖-纤维蛋白平板法测其活性,经比较不同菌株的活性,筛选出两株高产酶菌株:B.subtilisHW—12和B.subtilisHW—3。同时对菌体和菌落形态特点、生理生化反应进行了鉴定,认为B.SubtilisHW-12菌株可用来做为发酵生产该酶的菌种。  相似文献

The specification of anterior neuroectoderm is controlled by a highly conserved molecular mechanism in bilaterians. A forkhead family gene, foxQ2, is known to be one of the pivotal regulators, which is zygotically expressed in this region during embryogenesis of a broad range of bilaterians. However, what controls the expression of this essential factor has remained unclear to date. To reveal the regulatory mechanism of foxQ2, we performed cis‐regulatory analysis of two foxQ2 genes, foxQ2a and foxQ2b, in a sea urchin Hemicentrotus pulcherrimus. In sea urchin embryos, foxQ2 is initially expressed in the entire animal hemisphere and subsequently shows narrower expression restricted to the anterior pole region. In this study, as a first step to understand the foxQ2 regulation, we focused on the later restricted expression and analyzed the upstream cis‐regulatory sequences of foxQ2a and foxQ2b by using the constructs fused to short half‐life green fluorescent protein. Based on deletion and mutation analyses of both foxQ2, we identified each of the five regulatory sequences, which were 4–9 bp long. Neither of the regulatory sequences contains any motifs for ectopic activation or spatial repression, suggesting that later mRNA localization is regulated in situ. We also suggest that the three amino acid loop extension‐class homeobox gene Meis is involved in the maintenance of foxQ2b, the expression of which is dominant during embryogenesis.  相似文献

目的:比较锁定钢板(Locking plate,LP)与半肩关节置换(semi-shoulder arthroplasty,SSA)治疗老年肱骨近端NeerⅢ、Ⅳ型骨折临床疗效。方法:对我院骨科2009年5月至2013年5月收治的61例老年肱骨近端NeerⅢ、Ⅳ型骨折的患者资料进行回顾性分析,男性18例,女性43例,年龄60~84岁,平均69.3岁。其中采用LP治疗者40例,采用SSA治疗者21例,观察两组患者的手术时间、失血量、Neer评分情况,记录手术并发症情况,并进行统计学比较。结果:所有患者均获得随访,平均随访时间14.5个月(12~20个月)。LP组手术时间(105.6±20.4 min vs 80.6±18.2 min,t=2.650,P=0.01)多于SSA组,两组在手术出血量(188.5±25.2 ml vs 200.5±31.6 m L,t=1.666,P=0.1)、Neer评分优良率(92.5%vs 90.5%,X2=0.075,P=0.784)和并发症发生率(4.8%vs12.5%,X2=1.351,P=0.245)无明显差异。结论:在老年肱骨近端NeerⅢ、Ⅳ型骨折的治疗上,SSA手术时间短,但手术并发症发生率以及疗效优良率与LP相似,临床应根据病情及需要灵活选择手术方式。  相似文献

In recent years, many fungal genomes have become publicly available. In combination with novel gene editing tools, this allows for accelerated strain construction, making filamentous fungi even more interesting for the production of valuable products. However, besides their extraordinary production and secretion capacities, fungi most often exhibit challenging morphologies, which need to be screened for the best operational window. Thereby, combining genetic diversity with various environmental parameters results in a large parameter space, creating a strong demand for time-efficient phenotyping technologies. Microbioreactor systems, which have been well established for bacterial organisms, enable an increased cultivation throughput via parallelization and miniaturization, as well as enhanced process insight via non-invasive online monitoring. Nevertheless, only few reports about microtiter plate cultivation for filamentous fungi in general and even less with online monitoring exist in literature. Moreover, screening under batch conditions in microscale, when a fed-batch process is performed in large-scale might even lead to the wrong identification of optimized parameters. Therefore, in this study a novel workflow for Aspergillus niger was developed, allowing for up to 48 parallel microbioreactor cultivations in batch as well as fed-batch mode. This workflow was validated against lab-scale bioreactor cultivations to proof scalability. With the optimized cultivation protocol, three different micro-scale fed-batch strategies were tested to identify the best protein production conditions for intracellular model product GFP. Subsequently, the best feeding strategy was again validated in a lab-scale bioreactor.  相似文献
