煤附生真菌产漆酶菌株的分离鉴定及产酶特性研究   总被引:3,自引:1,他引:2  
从煤炭样品中筛选到一株产漆酶活性菌株,经菌体形态观察和ITS序列分析,鉴定为Trichoderma asperellum W03。菌株所产漆酶的最适反应pH为3.5-4.5,最适反应温度45℃,类似于白腐真菌漆酶。液态发酵条件的均匀设计实验表明,适宜的发酵培养基组成为:土豆200.00g/L、葡萄糖9.36g/L、米糠粉37.44g/L、硝酸钾4.00g/L、KH2PO43.20g/L、MgSO4·7H2O2.00g/L、CuSO4·5H2O0.005g/L、初始pH8.0;在33℃、180r/min、50mL/250mL的摇瓶培养条件下,棘孢木霉W03在孢子接种培养后48h、84h产酶量较高,分别处在菌体的快速生长期和衰亡期;菌体产酶受Cu2+、联苯胺诱导,而受1-萘酚、愈创木酚和2,4-D抑制。  相似文献

以木质素磺酸钠为唯一碳源的培养基对带岭凉水自然保护区土壤样品进行富集培养,涂布于愈创木酚-PDA平板。经2,2′-连氮-双(3-乙基苯并噻唑-6-磺酸)(ABTS)和丁香醛联氮(SGZ)平板检测初筛,ABTS法测定摇瓶发酵液酶活力复筛,筛选到一株漆酶高产真菌NF-05。形态学观察结合rDNA-ITS序列分析,鉴定该菌为半知菌疣孢漆斑菌Myrothecium verrucaria。该菌株在液体产酶培养基中生物量积累与产酶基本同步,发酵第5天达到产酶高峰,最高酶活力为8,375.87U/L。纯化漆酶对偶氮染料脱色研究结果表明,该酶在96h对甲基橙脱色率达到90%以上,以2,2,6,6-四甲基哌啶氧化物(TE)为介体时,48h脱色率即达90%以上;该酶在24h对橙黄Ⅰ的脱色率即达90%以上;以TE为介体时,该酶在24h即使橙黄G6完全脱色。  相似文献

Conidia of Metarhizium flavoviride grown in diphasic culture with sporulation on rice, were extracted in kerosene and formulated with peanut oil. The formulation led to high mortality when applied to field populations of Zonocerus variegatus; between 70 and 95% of insects in samples collected from the field 0, 3 or 7 days after spraying died. Sporulation of M. flavoviride was observed in 70–80% of the dead‐treated insects, which compares favourably with results obtained in laboratory bioassays. Many dead Z. variegatus, some with internal sporulation, were found in shaded niches and oviposition sites in the treated plots. No sporulation was observed in samples collected prior to application, and both sporulation and mortality differed significantly between control and treated plots. Some infections were observed in insects collected 16 and 23 days after treatment; these late infections may be a consequence of contact with spores surviving in the environment.  相似文献

At present knowledge of fungal biodiversity in North America is scattered in diverse sources ranging from well-reviewed, comprehensive databases to unedited databases of reports from the literature, information on file cards, and uncomputerized reference collections. Resources available electronically were used to determine their relative importance in evaluating the plant-associated fungi known from the United States and to a lesser extent Canada. The results demonstrate that the literature provides the greatest information but that reference collections in the US National Fungus Collections contribute between 22–31% additional data. Little overlap exists between fungi isolated as endophytes and those reported in the literature or as reference collections. Eighty to 100% of the plant-associated fungi reported from Canada are accounted for in comprehensive resources for the United States. A comprehensive database of plant-associated fungi in Canada, the United States, and eventually Mexico would serve as a valuable resource for those making plant quarantine decisions.  相似文献

【目的】揭示胶州湾典型污染海域半知菌群体结构多样性动态变化与生境的关系。【方法】根据以往对近岸污染调查记录,分别确定胶州湾海泊河、李村河入海口为中度、重度污染采样站位,石老人潮间为轻度污染对照站位,利用PCR-DGGE指纹图谱方法,研究该生境半知菌群体结构多样性季节动态变化与污染的关系。【结果】结果证明,污染指标除总氮含量外,其他各项指标含量在三站位的变化趋势为石老人潮间带最低,海泊河居中,李村河含量最高;与石老人站位相比,海泊河、李村河站位污染指标在四季变化较显著。首次证明在重金属和氮磷严重超标的海域,仍存在数量和种类较多的半知菌。PCR-DGGE指纹图谱结果显示,同一站位相似污染程度采样点之间DGGE指纹图谱比较一致,不同站位、不同季节之间指纹图谱存在明显的差异。优势条带割胶测序分析显示,3个站位存在大量的不可培养真菌,青霉属为半知菌优势种,重度污染的李村河入海口除半知菌外,存在较大量的阿米巴虫、腐霉等动物病原菌。【结论】污染海域的半知菌群体结构与生境污染程度及季节具有较密切的联系。  相似文献

The entomopathogenic entomophthoraceae (zygomycotina) Erynia rhizospora, Erynia dipterigena, and Erynia neoaphidis and the deuteromycete Aspergillus flavus produced single novel endoproteases (pI ca. 9) with activity against trypsin and chymotrypsin substrates. In contrast, the deuteromycete Paecilomyces farinosus produced a chymotrypsin (pI ca. 10).Inhibitor studies confirmed that the mixed activities (purified by isoelectric focusing) were derived from single endoproteases. The most potent inhibitor was Chicken ovoinhibitor. Little or no inhibition was observed for P. farinosus endoprotease by any of the chemicals tested.Although the different fungi possessed a broad spectrum of aminopeptidase activity, 3 species (E. rhizospora, E. dipterigena, and A. flavus) showed a preference for leucine at the N-terminal position and 2 species (E. neoaphidis and P. farinosus) showed maximal activity against arginine. Inhibitor studies confirmed these aminopeptidases as metallo-enzymes.  相似文献

Phialocephala fortinii is among the few identified hyphomycetes belonging to the Mycelium radicis atrovirens complex. This 'dark-septate endophyte' has a global distribution and colonizes a wide variety of host plants. In this study, the spatial distribution of discrete genets of P. fortinii on the forefront of a receding glacier was analysed using the randomly amplified polymorphic DNA (RAPD) technique to determine plants colonized and patterns of colonization. In two consecutive years of sampling, a total of 74 isolates of P. fortinii were obtained from nine plant species, typically ectomycorrhizal, ericoid mycorrhizal or non-mycorrhizal. The isolates showed substantial variation, sharing on average approx. half the RAPD markers. In the first year, three isolates belonging to a single genet were obtained from two plants separated by a distance of nearly 1.5 m. The continuity of this genet was assessed by a sampling the following year. No isolates similar to that, or any of the genets collected the year before were observed. Consequently, the identical isolates from the previous year were concluded to represent discontinuous ramets. Two additional large genets were observed during the second year of sampling, which inhabited roots of several plants representing three different species. These data suggest that the sharing of P. fortinii genets among plant species might play a fundamental role in adaptation and interaction within the whole plant community in a system undergoing primary succession.  相似文献

Dark septate root endophytes (DSE) are conidial or sterile fungi (Deuteromycotina, Fungi Imperfecti) likely to be ascomycetous and colonizing plant roots. They have been reported for nearly 600 plant species representing about 320 genera and 100 families. DSE fungi occur from the tropics to arctic and alpine habitats and comprise a heterogeneous group that functionally and ecologically overlaps with soil fungi, saprotrophic rhizoplane-inhabiting fungi, obligately and facultatively pathogenic fungi and mycorrhizal fungi. Numerous species of undescribed sterile and anamorphic taxa may also await discovery. Although DSE are abundant in washed root and soil samples from various habitats, and are easily isolated from surface-sterilized roots of ecto-, ectendo-, endo- and non-mycorrhizal host species, their ecological functions are little understood. Studies of DSE thus far have yielded inconsistent results and only poorly illustrate the role of DSE in their natural habitats. These inconsistencies are largely due to the uncertain taxonomic affinities of the strains of DSE used. In addition, because different strains of a single anamorph taxon seem to vary greatly in function, no clear generalizations on their ecological role have been drawn. This paper reviews the current literature on DSE and the ecology and discusses the need for and direction of future research.  相似文献
