目的: 原核表达盐穗木(Halostachys caspica C. A. Mey.)金属硫蛋白HcMT并探究其抗氧化活性。方法: 构建原核表达载体pET-32a-HcMT,转化至大肠杆菌Escherichia coli BL21,加入Zn2+胁迫培养(终浓度为200 μmol/L),分离纯化得到Zn-HcMT,测定Zn-HcMT自由基清除活性和总抗氧化能力,制备复合物Zn-HcMT/TiO2并做FTIR表征。结果: 通过原核表达获得融合蛋白Zn-HcMT,对·OH、O2·-、DPPH自由基具有较强的清除活性,对·OH、O2·-的IC50分别为0.386 mg/mL、0.038 mg/mL。融合蛋白浓度为0.01 mg/mL时,对DPPH清除率达(37.43 ± 0.006 8)%,浓度为0.3mg/mL时TEAC(trolox-equivalent antioxidant capacity)值为(1.023 ± 0.01)mmol/L,融合蛋白还原力A700为0.142 ± 0.055,FTIR图谱同时表现了Zn-HcMT和TiO2吸收特性。结论: Zn-HcMT具有良好的清除ROS活性及较强的抗氧化能力,在化妆品领域有潜在应用前景。  相似文献

The induction of shinorine (mycosporine-like amino acid (MAA)) in Anabaena variabilis PCC 7937 was studied in various culture media (BGA, BGA+, BG11 and Allen and Arnon’s). The objective was to select the most appropriate medium that can support the highest induction of MAAs and can be used for industrial production of these UV protective substances from cyanobacteria. Also, in vivo photosynthetic activity was measured under shinorine inducing conditions in all media. The shinorine content and photosynthetic activity (yield) were highest (2948.73 ± 61.13 nmol/g dry wt and 0.47 ± 0.01, respectively) in BG11 medium in comparison to others after 72 h of UV radiation. After the same duration of irradiation shinorine content was 1076.08 ± 21.77, 1320.07 ± 98.19 and 554.64 ± 16.47 nmol/g dry wt in BGA, BGA+, and Allen and Arnon’s media, respectively. Thus, BG11 medium can be used for mass production of MAAs from cyanobacteria.  相似文献

This paper reports on the synthesis and properties of a new UV-absorber (OC-NO) based on the most popular UV filter worldwide, ethylhexyl methoxycinnamate (OMC) in which the methoxy group has been replaced with a pyrrolidine nitroxide bearing antioxidant activity. This sunscreen active has therefore both UV-absorbing and antioxidant properties which could ideally address both the UV-B and UV-A skin photo-damage. For broad-spectrum coverage, the combinations of OC-NO with two commonly used UV-A absorbers (BMDBM and DHHB) were also studied. The results obtained reveal that OC-NO: (a) is as photostable as OMC after UV-A exposure; (b) acts as free radical scavenger as demonstrated by EPR and chemical studies; (c) reduces UV-A and UV-A+BMDBM induced lipid peroxidation in liposomes and cells, measured as reduced TBARS levels and increased C11-BODIPY red fluorescence, respectively; (d) has comparable antioxidant activity to that of vitamin E and BHT commonly used in skin care formulations; (e) is non-cytotoxic to human skin fibroblasts as assessed with the MTT assay when exposed to increasing doses of UV-A; and (f) OC-NO+DHHB is a promising, photostable broad spectrum UV-filter combination that concomitantly reduces UV-induced free radical damage. These results suggest that nitroxide/antioxidant-based UV-absorbers may pave the way for the utilization of 'multi-active' ingredients in sunscreens thereby reducing the number of ingredients in these formulations.  相似文献

The exposure to sun radiation is indispensable to our health; however, a long-term and high exposure could lead to cell damage, erythema, premature skin aging, and promotion of skin tumors. An underlying pathomechanism is the formation of free radicals which may induce oxidative stress at elevated concentrations. Different skin models, such as porcine-, murine-, human- ex vivo skin, reconstructed human skin (RHS) and human skin in vivo, were investigated during and after irradiation using X- and L-band EPR spectroscopy within different spectral regions (UVC to NIR). The amount of radical formation was quantified with the spin probe PCA and the radical types were measured ex vivo with the spin trap DMPO. The radiation dose influences the types of radicals formed in the skin. While reactive oxygen species (ROS) are always pronounced at low doses, there is an increase in lipid oxygen species (LOS) at high doses. Furthermore, the radical types arise independent from the irradiation wavelength, whereas the general amount of radical formation differs with the irradiation wavelength. Heat pre-stressed porcine skin already starts with higher LOS values. Thus, the radical type ratio might be an indicator of stress and the reversal of ROS/LOS constitutes the point where positive stress turns into negative stress.Compared to light skin types, darker types produce less radicals in the ultraviolet, similar amounts in the visible and higher ones in the infrared spectral region, rendering skin type-specific sun protection a necessity.  相似文献

The effects of artificial ultraviolet radiation [UVR; 8 W m−2 ultraviolet-A (UVA), 0.4 W m−2 ultraviolet-B (UVB)] on photosynthetic performance, growth and the capability to synthesise mycosporine-like amino acids (MAAs) was investigated in the aeroterrestrial green algae Stichococcus sp. and Chlorella luteoviridis forming biofilms on building facades, and compared with the responses of two green algae, from soil (Myrmecia incisa) and brackish water (Desmodesmus subspicatus). All species exhibited decreasing quantum efficiency (F v/F m) after 1–3 days exposure to UVR. After 8–12 days treatment, however, all aeroterrestrial isolates exhibited full recovery under UVA and UVA/B. In contrast, D. subspicatus showed only 80% recovery after treatment with UVB. While Stichococcus sp. and C. luteoviridis exhibited a broad tolerance in growth under all radiation conditions tested, M. incisa showed a significant decrease in growth rate after exposure to UVA and UVA/B. Similarly D. subspicatus grew with a reduced rate under UVA, but UVA/B led to full inhibition. Using HPLC, an UV-absorbing MAA (324 nm-MAA) was identified in Stichococcus sp. and C. luteoviridis. While M. incisa contained a specific 322 nm-MAA, D. subspicatus lacked any trace of such compounds. UV-exposure experiments indicated that all MAA-containing species are capable of synthesizing and accumulating these compounds, thus supporting their function as an UV-sunscreen. All data well explain the conspicuous ecological success of aeroterrestrial green algae in biofilms on facades. Biosynthesis and accumulation of MAAs under UVR seem to result in a reduced UV-sensitivity of growth and photosynthesis, which consequently may enhance survival in the environmentally harsh habitat.  相似文献

The impact of sunscreen formulations on the barrier properties of human skin are often overlooked leading to formulations with components whose effects on barrier mechanical integrity are poorly understood. The aim of this study is to demonstrate the relevance of carrier selection and sunscreen photostability when designing sunscreen formulations to protect the biomechanical barrier properties of human stratum corneum (SC) from solar ultraviolet (UV) damage. Biomechanical properties of SC samples were assayed after accelerated UVB damage through measurements of the SC's mechanical stress profile and corneocyte cohesion. A narrowband UVB (305–315 nm) lamp was used to expose SC samples to 5, 30, 125, and 265 J cm?2 in order to magnify damage to the mechanical properties of the tissue and characterize the UV degradation dose response such that effects from smaller UV dosages can be extrapolated. Stresses in the SC decreased when treated with sunscreen components, highlighting their effect on the skin prior to UV exposure. Stresses increased with UVB exposure and in specimens treated with different sunscreens stresses varied dramatically at high UVB dosages. Specimens treated with sunscreen components without UVB exposure exhibited altered corneocyte cohesion. Both sunscreens studied prevented alteration of corneocyte cohesion by low UVB dosages, but differences in protection were observed at higher UVB dosages indicating UV degradation of one sunscreen. These results indicate the protection of individual sunscreen components vary over a range of UVB dosages, and components can even cause alteration of the biomechanical barrier properties of human SC before UV exposure. Therefore, detailed characterization of sunscreen formulation components is required to design robust protection from UV damage.  相似文献

1,3-Diferuloyl-sn-glycerol is found ubiquitously throughout the plant kingdom, possessing ultraviolet adsorbing and antioxidant properties. Diferuloyl glycerol was synthesized and isolated as a byproduct in up to 5% yield from a pilot plant scale packed-bed, biocatalytic transesterification of ethyl ferulate with soybean oil or mono- and diacylglycerols from soybean oil. The yield of the diferuloyl glycerol byproduct was directly proportional to the overall water concentration of the bioreactor. The isolated diferuloyl glycerol exhibited good ultraviolet adsorbing properties, 280–360 nm with a λmax 322 nm, and compared well to the efficacy of commercial sunscreen active ingredients. The antioxidant capacity of diferuloyl glycerol (0.25–2.5 mM) was determined by its ability to scavenge 2,2-diphenyl-1-picrylhydrazyl radicals and was comparable to that of ferulic acid. At current pilot plant scale production capacity, 120 kg diferuloyl glycerol byproduct could be isolated per year. Product names are necessary to report factually on available data; however, the USDA neither guarantees nor warrants the standard of the product, and the use of the name by the USDA implies no approval of the product to the exclusion of others that may be suitable.  相似文献

The synthesis of vitamin D in skin is a two-stage process that begins with the production of previtamin D after irradiation of 7-dehydrocholesterol by ultraviolet (UV) radiation. A number of personal and environmental factors control the probability of a suitable UV photon reaching a molecule of 7-dehydrocholesterol in the skin. These are astronomical factors that govern the solar zenith angle (SZA), and the local state of the atmosphere, determining the available solar UV radiation; skin pigmentation and age, determining competing absorbers of UV radiation and available 7-dehydrocholesterol; individual behaviour in the local surroundings, determining exposure of unprotected skin to available UV radiation. The only one of these influences that can be determined unequivocally for any situation is the SZA. The other influences must be considered either as individual case studies, or be represented by "typical" and "idealised" situations for the weather, skin and behaviour. At large SZAs there is insufficient solar UV radiation to initiate significant vitamin D synthesis. At smaller SZAs assessment of solar exposure necessary for vitamin D synthesis can only be indicative and application of any such assessment necessarily requires awareness of both self- and the local environment.  相似文献

Sunscreens have different levels of protection, measured most commonly with the sun protection factor (SPF). Students initially believed higher SPF factors mean greater sun protection and learned through this activity that higher SPF does not mean greater protection. Students analyzed the amount of ultraviolet radiation (UVR) protection and composition of different sunscreens, first by UVR sensors in an outdoor activity and then by thin layer chromatography. These activities demonstrated that even some sunscreens provide protection from harmful UVR, and that different sunscreens, despite varying SPF factors and brand names, have similar composition. Students can apply their knowledge outside the classroom on outdoor safety and solar protection.  相似文献

Background information on the inefficacy of sunscreens to provide free radical protection in skin, despite their usefulness in preventing sunburn/erythema, prompted us to synthesize a compound which would display in the same molecule both UV-absorbing and antioxidant capacities. For this purpose, the UVB absorber, 2-ethylhexyl-4-methoxycinnamate (OMC) was combined with the piperidine nitroxide TEMPOL, which has antioxidant properties. The spectral properties of the new nitroxide-based sunscreen (MC-NO) as well as its efficacy to prevent photo-oxidative damage to lipids induced by UVA, natural sunlight and 4-tert-butyl-4-methoxydibenzoylmethane (BMDBM), a photo-unstable sunscreen which generates free radicals upon UV radiation, was studied. The results obtained demonstrate that MC-NO: (a) absorbs in the UVB region even after UVA irradiation; (b) acts as free radical scavenger as demonstrated by EPR experiments; (c) strongly reduces both UVA-, sunlight- and BMDBM-induced lipid peroxidation in liposomes, measured as reduced TBARS levels; and (d) has comparable antioxidant activity to that of commonly used vitamin E and BHT in skin care formulations. These results suggest that the use of the novel sunscreen-antioxidant or of other nitroxide-based sunscreens in formulations aimed at reducing photoinduced skin damage may be envisaged.  相似文献
