A laboratory calibration study was undertaken with juvenile Sepia officinalis (80-85 g initial wet weight) to investigate the effects of different food rations and different starving intervals on RNA/dry weight (DW) ratios and RNA/DNA ratios in cephalopod mantle muscle at two different temperatures. The digestive gland index was also used as an additional indicator of recent growth. High food rations and low temperature went along with high RNA/DW ratios and high RNA/DNA ratios. Starving resulted in a linear decline in growth performance and a concomitant decrease in RNA/DW and RNA/DNA ratio, with RNA/DNA ratios representing the growth data better. RNA/DNA ratios decreased faster at higher temperatures. A fluorimetric assay for nucleic acid analysis was optimized for cephalopod mantle tissues and yielded reproducible RNA/DNA ratios with a relative variance below 10%. Thus, it may be possible to use this estimator of recently encountered feeding regime for the evaluation of mortality rates of early teuthid paralarvae to eventually support stock management. Also, log relative digestive gland weight showed a strong relationship with starving time, but, surprisingly, not with temperature. Data from the two temperatures analyzed could be combined to form a common regression line of relative digestive gland index with starving time. This indicator for recent growth might be especially suitable for large specimens with a well-developed digestive gland.  相似文献

Potentials were recorded from the epidermal head lines and from the CNS of young cuttlefish, Sepia officinalis, in response to weak water movements. 1. Within the test range 0.5-400 Hz a sinusoidal water movement elicits up to 4 components of response if the electrode is placed on a headline: (i) a positive phasic ON response; (ii) a tonic frequency-following microphonic response; (iii) a slow negative OFF response; and (iv) compound nerve impulses. 2. The amplitude of both the ON wave and the microphonic potential depends on stimulus frequency, stimulus amplitude and stimulus rise time. Frequencies around 100 Hz and short rise times are most effective in eliciting strong potentials. The minimal threshold was 0.06 microns peak-to-peak water displacement at 100 Hz (18.8 microns/s as velocity). 3. Change of direction of tangential sphere movement (parallel vs. across the head lines) has only a small effect on the microphonic and the summed nerve potentials. 4. Frequency and/or amplitude modulations of a carrier stimulus elicit responses at the onset and offset of the modulation and marked changes in the tonic microphonic response. 5. Evoked potentials can be recorded from the brain while stimulating the epidermal lines with weak water movements. The brain potentials differ in several aspects from the potentials of the head lines and show little or no onset or offset wave at the transitions of a frequency and amplitude modulation.  相似文献

The cuttlefish, Sepia officinalis, provides a fascinating opportunity to investigate the mechanisms of camouflage as it rapidly changes its body patterns in response to the visual environment. We investigated how edge information determines camouflage responses through the use of spatially high-pass filtered 'objects' and of isolated edges. We then investigated how the body pattern responds to objects defined by texture (second-order information) compared with those defined by luminance. We found that (i) edge information alone is sufficient to elicit the body pattern known as Disruptive, which is the camouflage response given when a whole object is present, and furthermore, isolated edges cause the same response; and (ii) cuttlefish can distinguish and respond to objects of the same mean luminance as the background. These observations emphasize the importance of discrete objects (bounded by edges) in the cuttlefish's choice of camouflage, and more generally imply that figure-ground segregation by cuttlefish is similar to that in vertebrates, as might be predicted by their need to produce effective camouflage against vertebrate predators.  相似文献

We have determined the content of free l-amino acids and d-aspartate in the nervous tissue of three representative cephalopods: Sepia officinalis, Octopus vulgaris, and Loligo vulgaris, and the optic lobes of adult and embryo Sepia officinalis. Taurine is the most abundant amino acid in the cephalopod nervous tissue. Its content amounts to more than 50% of the total free amino acids. The other most concentrated amino acids are Glu, Ala, Asp, and GABA. High concentrations of d-aspartate were found in the nervous tissue of all cephalopods examined (7–12 μmol/g wet tissue) which represents 50–80% of the total aspartate (d + l), depending on the animal. Among the various regions of the brain of Octopus vulgaris, d-aspartate was found to be evenly distributed in the various regions of the brain. In nerve tissue of Sepia officinalis, there is no significant difference in the pattern of free l-amino acids, in particular of the d-aspartate concentration, between adults and embryos, except for GABA, Gly, His and Thr. This suggests that d-aspartate in nerve tissue of the Cephalopoda is of endogenous origin and not a product of accumulation from exogenous sources. From a comparative study of the content of d-aspartate in the nervous tissue of different animals, we found that protostomia contain a significantly higher amount than deuterostomia. Thus, d-aspartate could be a criterion to distinguish the protostomia phyla from the deuterostomia phyla.  相似文献

The ovoid cells of the branchial heart complex of Sepia officinalis L. were investigated with respect to their role in detoxification processes. The electron microscopical localization of in situ injected ferritin in the endocytotic-lysosomal system, and the fluorescence microscopical localization of protein-bound Evans blue in the ovoid cells of in vivo incubated animals, indicate that foreign materials are eliminated from the hemolymph by the branchial heart tissue. In addition to the non-circulating ovoid cells of the branchial heart, hemocytes in the circulating blood and in the wall of the branchial heart are also involved in the incorporation of allogeneic substances and bacteria, or their debris. Based on these observations, we propose that the ovoid cells, together with circulating and adhesive hemocytes in the branchial hearts, are an important component of a more comprehensive defence and detoxification system in dibranchiate cephalopods that prevents contamination of the whole organism by endocytotic removal of noxious substances from the hemolymph.  相似文献

Sperm displacement behavior of cuttlefish (Sepia esculenta) was observed in a tank. Before ejaculation, male cuttlefish used their arms III to scrape out sperm masses attached to the buccal membranes of females. The removed sperm mass debris was directly visible and countable. Active sperm were present within the removed sperm debris, implying that the aim of this behavior is to remove competing male sperm. However, many sperm masses remained on the female buccal membrane even after the removal behavior, showing that sperm removal in S. esculenta is incomplete. The duration of sperm removal (an indicator of male investment in that process) was unaffected by the body sizes of mated pair, the duration of spermatangia placement at the current mating (for the hypothesis that the sperm removal serves to creat attachment space of spermatophores), or the estimated amount of sperm masses deposited from previous matings. Moreover, male S. esculenta performed sperm removal regardless of whether the last male to mate with the partner was himself, suggesting males remove not only the sperm of rivals but also their own. Although the number of removed sperm masses increased with the time spent on removal of sperm, male cuttlefish may shorten the duration of sperm removal to avoid the risk of mating interruption. We conclude that this time restriction would likely influence the degree of partial sperm removal in S. esculenta. The protein p27BBP (alias eIF6) occurs in yeast and mammalian epithelial cells. It is essential for ribosome genesis and has also been implicated in the functionality of integrins and intermediate filaments. By immunoblot, we show that homogenized integument from Sepia officinalis (Cephalopoda, Mollusca) contains a protein with immunological properties that closely resemble those of p27BBP. We also demonstrate, by immunogold electron microscopy with an indirect immunoreaction technique on ultrathin sections of human skin and Sepia integument, that p27BBP is constantly present in both species in epithelial cells, fibroblasts, and muscle fibers. It is found in the vicinity of intermediate filaments, in nucleoli, along the internal wall of the nuclear membrane, and in association with desmosomes and hemidesmosomes and occasionally occurs extracellularly. Thus, the structure and function of p27BBP seem to have been highly conserved throughout evolution; the protein appears to be essential in eukaryotic cells in which it interacts with several ultrastructural components of diverse function.Financial support was provided by funds from FIRST.  相似文献

Hatchlings cuttlefish were reared in the laboratory from hatching until 30 days old, fed with live shrimp, frozen shrimp or fish oil-enriched frozen shrimp. Survival of cuttlefish fed with oil-enriched frozen shrimp was better than in animals receiving live shrimp. However, there was no difference with cuttlefish fed with frozen shrimp, even if survival of those receiving oil-enriched frozen shrimp was always higher all along the experiment. Lower survival in animals fed with live shrimp represented the problem of using such food and confirms the necessity to elaborate an artificial food. Utilization of artemia was detrimental to growth and induced low values of instantaneous growth rate (IGR) and conversion rate even after feeding cuttlefish with shrimp. Nevertheless, growth parameters evolutions generally corresponded to those observed by other researchers. The profile noticed at the end of the experiment is typically observed when cuttlefish acquire their adult digestive system. Main differences were observed between groups fed with live shrimp or oil-enriched frozen shrimp. Enrichment did not induce same growth as in cuttlefish receiving live prey. However, at 20 and 25 days after hatching (DAH), in cuttlefish fed with oil-enriched frozen shrimp, ration was lower for the same growth than in other groups.These data showed capacity of juvenile cuttlefish to adjust their digestive enzyme activities according to the diet and the stage of development. Indeed, chymotrypsin was strongly influenced by enrichment, while other enzymes showed difference between live and frozen preys. Trypsin exhibited regulation by diet after 20 DAH. Freezing seemed to delay development as acid phosphatases, characteristic of first stages of cuttlefish, had lower activity in cuttlefish fed with live shrimp at 10 DAH. Moreover, influence of the stage of development was strong as activities between 20 and 30 DAH were different in all groups. This was in relation with evolution of the digestive system. These data illustrated the difficulty to elaborate optimal diet as digestive system evolves.  相似文献
