Essential genes were identified in the 1.5-map unit dpy-5 unc-13 region of chromosome I in the Caenorhabditis elegans genome by rescuing lethal mutations using the duplication sDp2. In this paper, we report the mapping and complementation testing of lethal mutations, 45 of which identify 18 new, essential genes. This analysis brings the number of essential genes defined by the sDp2 rescue of lethal mutants to 97; 64 of these map between dpy-5 and unc-13. 61% of these essential genes are identified by more than one allele. Positioning of the mutations was done using the breakpoints of six duplications. The mutant phenotypes of 14 loci essential for fertility were characterized by Nomarski microscopy and DAPI staining. None of the mutants were rescued by wild-type male sperm. The cytological data showed that four genes produced mutants with defects in gonadogenesis, let-395, let-603, let-605 and let-610. Mutations in seven genes, let-355, let-367, let-384, let-513, let-544, let-545 and let-606, affected germ cell proliferation or gametogenesis. Mutants for the remaining three genes, let-370, let-599 and let-604, produced eggs that failed to develop or hatch, thereby acting as maternal effect lethals. ABSTRACT

Background: The disruption of circadian rhythm has been found to associate with obesity in vivo and in vitro. Sleep duration, eating habits, total feeding time, and nightshift work can also affect circadian rhythms. This study investigated the association between misalignment of circadian rhythm and obesity in Korean men, using a cross-sectional database.

Methods: This study used data from the Korean National Health and Nutrition Examination Survey (KNHANES), whose study population was 3,658 men aged 18 to 60 years. General and abdominal obesity was defined as a body mass index (BMI) ≥ 25 kg/m2 and waist circumference ≥ 90 cm, respectively. Circadian rhythm factors were determined with a self-report questionnaire and included breakfast frequency, sleep duration, and work time. Frequency of breakfast was divided into regular breakfast (five to seven times a week) and irregular breakfast (less than five times a week). Sleep duration was divided into less than 7 hours, 7–9 hours, and over 9 hours. Working time was defined as day/evening, night shift, and other type. The adjusted odds ratios (aORs) and 95% confidence intervals (CIs) for general and abdominal obesity were calculated using multivariable logistic regression according to the number of factors that disturb the circadian rhythm.

Results: Participants with 1 (aOR 1.34, 95% Cl 1.10–1.61) and ≥2 (aOR 1.62, 95% Cl 1.29–2.05) factors disturbing circadian rhythms were associated with elevated risk for general obesity. Similarly, those with 1 (aOR 1.33, 95% Cl 1.09–1.63) and ≥2 (aOR 1.70, 95% Cl 1.32–2.20) factors had elevated risk for abdominal obesity.

Conclusions: Factors disturbing the circadian rhythm were associated with general and abdominal obesity. Additional studies are needed, and associations with metabolic diseases should be investigated.  相似文献

Summary In some strains of mice, eggs when X irradiated during the pronuclear stage, undergo a mitotic block in the G2 phase of the first cell cycle and cleave when the second division takes place in controls. The importance of this effect varies considerably with the strain and depends exclusively on the maternal genotype. In previous work, two-dimensional electrophoresis showed that eggs blocked at the one-cell stage after irradiation, undergo the same modifications in polypeptide synthesis as two-cell controls of the same age, except at the time of normal first mitosis, where three polypeptide sets of 30, 35 and 45 kDa appear only in cleaving controls. In the present study, we have found phosphorylations in dividing controls, on polypeptides of 30, 35 and 45 kDa. These phosphorylations are not seen in blocked irradiated eggs.  相似文献

母性行为是动物以维持幼崽的生存及生理健康为主要目的的一种基本行为,母性行为作为重要的早期经历对动物的个体发展有深远影响。动物的行为在时间和环境中具有一致性,多个行为特征的一致性加权被称为气质特征,气质特征的差异是犬(Canis lupus familiaris)能否顺利通过培训成为导盲犬的决定性因素。其中,胆量是决定导盲犬培训成功与否的重要气质特征。本研究以中国导盲犬大连培训基地的拉布拉多种犬及幼犬为研究对象,探究母性行为水平对幼犬胆量的影响。本研究通过视频观察记录拉布拉多犬哺乳期前21 d的母性行为变量时长,对在哺乳区内、身体接触、哺乳和舔舐幼犬4项变量进行主成分分析后将7只实验犬分为母性行为高水平与低水平两组。对两组犬生产的共54只幼犬于6 ~ 8周龄时进行幼犬胆量行为测试,根据胆量行为测试的评分标准对幼犬的行为表现进行评分,统计分析母性行为高水平组与低水平组其幼犬的胆量是否存在差异。本研究结果表明,母性行为低水平组的幼犬在胆量测试中面对陌生环境、突然出现的响声刺激、突然打开的雨伞刺激以及陌生人的游戏邀请时均表现出更大的胆量。在被动测试中,母性行为低水平组幼犬的探索潜伏时长显著短于母性行为高水平组幼犬(P < 0.05),探索范围显著大于母性行为高水平组幼犬(P < 0.05),紧张程度极显著低于母性行为高水平组幼犬(P < 0.01);在金属响声测试中,母性行为低水平组幼犬的惊吓反应(P < 0.01)和紧张程度(P < 0.01)均极显著低于母性行为高水平组幼犬;在雨伞测试中,母性行为低水平组幼犬的紧张程度显著低于母性行为高水平组幼犬(P < 0.05);在玩具测试中,母性行为低水平组幼犬的玩耍兴趣显著高于母性行为高水平组幼犬(P < 0.05),紧张程度显著低于母性行为高水平组幼犬(P < 0.05);在斜坡隧道测试中,母性行为低水平组幼犬的紧张程度显著低于母性行为高水平组幼犬(P < 0.05),通过斜坡的用时短于母性行为高水平组幼犬,但经统计检验无显著差异(P > 0.05)。本研究的结论为低母性行为水平带给幼犬强度适当的早期生活压力,使幼犬面对新环境刺激时表现出更好的适应能力和较大的胆量。本研究为工作犬种犬的筛选提出新的建议:母性行为水平低的种犬对幼犬胆量的发展有更好的影响。  相似文献

The fatty acid composition of constituent phospholipids and the cholesterol content of rat liver plasma membranes were determined subsequent to maternal alcohol ingestion during pregnancy and lactation. The alcoholic group was given a liquid Metrecal diet containing 37% ethanol-derived calories. The control group was pair-fed an isocaloric sucrose/Metrecal diet. Litters were killed for lipid analyses at days 5, 15 and 25 after birth. These studies revealed that the total phospholipid phosphorus was similar and increased significantly with age in both groups. Cholesterol also increased significantly with age in both groups but was greater in the alcoholic pups, resulting in a higher cholesterol/phospholipid molar ratio. While the phosphatidylethanolamine (PE) content increased with age in both groups, that of sphingomyelin decreased. Phosphatidylserine + phosphatidylinositol (PS + PI) was significantly higher in the control group at all ages studied. A consistent increase of C22:6 in phosphatidylcholine (PC), sphingomyelin, PS + PI and in the total phospholipid fraction from alcoholic pups was observed. Although other fatty acid changes were found in PC, PS + PI and sphingomyelin, PE was not affected. These results suggest that specific adaptive changes were induced in the liver plasma membrane lipids of the progeny from alcoholic rats.  相似文献

We previously assessed the motivational properties of pups relative to those of cocaine in parturient female rats (dams) across the postpartum period and demonstrated that the larger subset of dams in early postpartum (PPD8) preferred the pup-associated chamber, whereas the majority of dams tested in late postpartum (PPD16) preferred the cocaine-associated chamber [Mattson, B.J., Williams, S., Rosenblatt, J.S., Morrell, J.I. 2001. Comparison of two positive reinforcing stimuli: pups and cocaine throughout the postpartum period. Behav. Neurosci., 115, 683-694; Seip, K.M., Morrell, J.I. 2007. Increasing the incentive salience of cocaine challenges preference for pup- over cocaine-associated stimuli during early postpartum: place preference and locomotor analyses in the lactating female rat. Psychopharmacology 194, 309-319]. The present study uses a dual-choice conditioned place preference to ask how the progression of the postpartum period, including natural pup development, influences maternal motivation for pups. Preferences for cued chambers associated with pups that were age-matched to the postpartum stage of the dam in contrast to a stimulus with little incentive salience were higher during the early than the late postpartum, suggesting that the incentive salience of pups diminishes as the postpartum period progresses. Preferences of the early postpartum dams deprived of pups for 15 min, 2, 6, 12 or 22 hrs prior to conditioning and testing did not differ statistically but there was a trend of more pup preference after 22 hr deprivation; pup age was not an important factor in early postpartum. In marked contrast, late postpartum dams only exhibited robust pup-associated place preference when they were conditioned with young (4-7 day-old) pups or after a 22 hr period of deprivation from contemporaneous pups. Together these results suggest that both forces are at work in the mother-pup dyad, changes in the pups as they develop and changes in the physiological and endocrine state of the female as she progresses through the postpartum period.  相似文献

When sows kept under commercial conditions were put into crates in the early 1960s, the neuro-endocrine regulation of the maternal behavior in these domestic animals was disputed. Thus, the study of sow maternal behavior intensified and today a significant body of knowledge has accumulated to support the hormonal regulation of sow maternal behavior. The onset of nest building is associated with a periparturient decline in progesterone, an increase in prolactin and a major rise in plasma concentrations of PGF2alpha the day before parturition. Some nest building behaviors, such as pawing and gathering straw, have been found to correlate with changes in the levels of progesterone, prolactin and somatostatin. The duration of the birth process correlates negatively with peripheral oxytocin levels. During lactation, the stimuli from the piglets affect the release of several hormones which not only regulate the let down of milk but also sow metabolism and mammary milk production. The sow's nursing behavior ensures an even distribution of milk to her piglets. The piglets suckling behavior, in turn, is mainly a way to communicate their individual nutritional needs.  相似文献

A study of the research on postpartum mother-infant bonding shows that results from poorly constructed research programs were published in major journals and became a part of hospital policy because the bonding concept was politically useful in the struggle between advocates of natural childbirth and managers of the medical model of birth. The concept was also uncritically accepted because it was consistent with a longstanding ideology of motherhood that sees women as the prime architects of their children’s personalities. Diane Eyer earned her Ph.D. in developmental psychology from the University of Pennsylvania. She is currently writing a book on the ways in which the concepts of bonding and attachment have affected our understanding of appropriate early childcare.  相似文献

Abstract

When the Human Genome Project (HGP) was launched, our genome was presented as our ‘blueprint’, a metaphor reflecting a genetic deterministic epistemology. Eventually, however, the HGP undermined rather than strengthened the understanding of genomes as blueprints and of genes as ultimate causal units. A symbolical turning point was the discovery that the human genome only contains~22,500 genes. Initially, this was seen as a narcissistic offence. Gradually, however, it strengthened the shift from traditional genetics and biotechnology (i.e., gene-oriented approaches) to genomics, i.e. genome-oriented or systems approaches, emphasizing complexity. The 20th century can be regarded as the century of biotechnology and of the gene. Its history demonstrated that the will to know (notably: to know ourselves) has never been a disinterested affair: it is driven by a will to improve (notably: to improve ourselves). In this article it is claimed that, as genomics takes us beyond a genetic deterministic understanding of life, this must have consequences for societal research and debate as well. Policies for self-improvement will increasingly rely on the use of complex interpretation. Therefore, the emphasis must shift from issues such as genetic manipulation and human enhancement to issues involved in governance of novel forms of information.  相似文献

Summary Pale eggs and partial cleavage are two mutations with a maternal effect that are found in the same family ofXenopus l. laevis. The pale eggs have animal hemispheres of a yellow to beige colour and give rise to normally pigmented tadpoles and frogs. The cells of pale embryos contain fewer melanosomes than those of controls. The partial cleavage eggs are characterized by an abnormality of cleavage visible from the eight-cell stage onwards, by abnormal yolk platelet distribution and abnormal cytological features. Cleaved, syncytial and uncleaved areas are observed in these eggs, which are lethal at the blastula stage.  相似文献
