The effect of waveguiding property (i.e., the intensity distribution) of the photoreceptor on the number of photons absorbed in a photoreceptor has been studied. It has been found that the effect is significant only for large values of the exposure and the maximum effect is less than 11% in the case of human rod photoreceptor. Aquaspirillum magnetotacticum MS-1 cells cultured microaerobically (dissolved O2 tension 1% of saturation), expressed proteins with superoxide dismutase (SOD) activity. The majority (roughly 95%) of total cell superoxide dismutase activity was located in the cell periplasm with little or no activity in the cell cytoplasm. Irontype SOD (FeSOD) contributed 88% of the total activity activity detected, although a manganese-type SOD (MnSOD) was present in the periplasm as well. Cells cultured at a higher dissolved O2 tension (10% of saturation) expressed increased activity of the MnSOD relative to that of the FeSOD. 相似文献

The dielectrophoretic (DEP) crossover method has been applied to the detection of cell responses to toxicants. Time and dose responses of the human cultured leukemia (HL-60) line were measured for paraquat, styrene oxide (SO), N-nitroso-N-methylurea (NMU) and puromycin. These toxicants were chosen because of their different predominant mechanisms of action, namely membrane free radical attack, simultaneous membrane and nucleic acid attack, nucleic acid alkylation, and protein synthesis inhibition, respectively. For all treatments, the specific membrane capacitance (Cmem) of the cells decreased while the specific membrane conductance (Gmem) increased in dose- and time-dependent manners. The DEP responses correlated sensitively with alterations in cell surface morphology, especially folds, microvilli, and blebs, observed by scanning electron microscopy. The DEP method was more sensitive to agents that had a direct action on the membrane than to agents for which membrane alterations were secondary. The responses to paraquat and SO, which directly damaged the cell membrane, could be detected 15 min after exposure, while those for puromycin and NMU, which acted on intracellular targets, could be detected after 30 min. The detection times and dose sensitivity results showed that the DEP method is much faster and more sensitive than conventional cell and higher organism viability testing techniques. The feasibility of producing small instruments for toxicity detection and screening based on cellular dielectric responses is discussed. 相似文献

This paper discusses policy responses to the potential loss of biodiversity in the Mara Area of Kenya from the conversion of essentially wild and undeveloped rangeland to developed agriculture. Property rights are central to the debate, and raise two fundamental issues. First, to what extent do the Maasai, the traditional users and owners of the land, have the right to benefit from the development potential of their land to further their economic, social and political standing, even if by so doing they create domestic and global externalities through the loss of biodiversity. Second, if the state alienates their development rights in the name of conservation, then to what extent should the state compensate the Maasai for their lost economic opportunities. To the Maasai, conservation as implemented through Government policy is a publicc bad: they are denied access to resources, their costs of production are significantly increased, and development is slowed down or actively discouraged. A cost:benefit analysis suggests that it is neither supportable nor sustainable to condemn the Maasai to a poverty trap on behalf of conservation, and that it is instead socially prolitable for the Kenyan Government to meet in full their opportunity costs of forgone economic benefits. 相似文献

New developments in magnetic labelling techniques for cells and microspheres have extended the useful range of magnetic separation, particularly high gradient magnetic separation, into biotechnical areas. The basic magnetic principles involved are reviewed and representative samples of labelling techniques and results drawn from the past three years are presented. Vibrational optical coherence elastography (OCE) is a promising tool for extracting the mechanical property of soft tissue. Purpose of this study is focusing on settling the optimal frequency range for vibrational OCE with evenly distributed stress filed. A finite element model of 2% agar phantom was built by ANSYS with a vibration stimulation frequency range from 200 to 3000 Hz. Practical experiments were carried out for cross‐validation with the same frequencies and sample. Lateral and horizontal stress filed distributions under different frequencies were mathematically evaluated by coefficient of variance and degree of linearity. Results from simulation and practical experiment cross‐validated each other and 1000 Hz was set as the maximum ideal frequency for vibrational OCE, while the minimum frequency is set by theoretical calculation with a result of 250 Hz. An ex vivo biological sample was utilised to testify performance of vibrational OCE with excitation frequencies in and out of concluded optimal range, which showed that stiffness was better mapped out in optimal frequency range. 相似文献

矿业废弃地生态系统自然恢复的植被演替过程与机理是生态恢复研究的重要内容之一.以空间代替时间的方法,选择立地条件基本一致的4个不同自然恢复年限铅锌矿区为对象,研究黔西北土法炼锌渣场废弃地植被自然演替与矿渣基质理化性质的交互效应.结果表明: 随着堆置时间的增加,矿渣基质的营养条件明显得到改善,全氮、全磷和全钾含量极显著增加, pH上升,电导率下降,容重降低,有效铅和镉显著降低. 同时,随着恢复时间的增长,植物群落的物种丰富度、多样性指数和均匀度也相应提高.植物群落组成以多年生草本植物为主,植物群落演替在前20年较为缓慢,30年后植被群落盖度可达到53%,超过40年盖度可达87%.矿渣理化性质与物种多样性显著相关,典型变量分别是全氮、全磷和全钾;物种多样性指数与有效铅和镉呈显著负相关.土法炼锌渣场废弃地植被自然演替过程在30年后速度加快,植被生长的限制因子是营养供给不足和重金属的有效性高. 相似文献

The environmental record from Lake Baikal, Russia, from 310 to 50 ky BP (MIS 9a to MIS 3) was interpreted using rock magnetic, UV–Vis spectral, mineralogical, and diatom analyses. The age model was based on a correlation of the diatom and chemical weathering records and the summer insolation curve at 55°N and checked against an age model based on the proxy of relative palaeointensity of the Earth's magnetic field. Peaks in chemical weathering within the watershed, inferred from maximum concentration of magnetic and coloured minerals and mica, the lowest mean Fe oxidation state in silicates and highs in expandable clay minerals correlated with the Northern Hemisphere summer insolation minima at 55°N. Reconstructed changes in weathering intensity are better correlated to insolation patterns than to global ice volume records. We propose a scheme of yet missing palaeoenvironmental interpretation of the diatom assemblage, including also some extinct species. Aulacoseira baicalensis and Aulacoseira skvortzowii were abundant in the early stages of lake flora recovery immediately after deglaciation and during MIS 7e and MIS 5e; periods of more pronounced continental climate and peak chemical weathering. Stephanodiscus formosus var. minor, Cyclotella minuta and Cyclotella ornata dominated in intervals of decreased seasonality and decreased humidity at the end of most interglacial/interstadial diatom zones. Stephanodiscus grandis, Stephanodiscus carconeiformis and Stephanodiscus formosus were ubiquitous between MIS 8 and MIS 5, an interval marked by high seasonality, i.e., large differences between winter and summer insolation, and low humidity revealed by a low hydrolysis of expandable clay minerals in the watershed. Diatom concentrations peaked in the climatic optima of MIS 7e and MIS 5e and in the short periods marked by shifts to warmer conditions in the upper sections of MIS 5: MIS 5c (103–99 ky BP), MIS 5b (90–88 ky BP), and MIS 5a (84–79 ky BP) in which increased humidity resulted in enhanced hydrolysis of clay minerals. No such short similar climatic optimums were found from MIS 9a to MIS 6. Sharp climate deteriorations recorded in the diatom and clay mineral records at 107, 94, and 87 ky BP, however, occurred within 1–2 ky of cold extremes in North Atlantic sea surface temperature emphasizing the strong teleconnections between the two localities. 相似文献

A binuclear copper complex, [Cu2(BPMP)(OAc)2][ClO4]·H2O, has been prepared using the binucleating ligand 2,6-bis[bis(pyridin-2-ylmethylamino)methyl]-4-methylphenol (H-BPMP). The
X-ray crystal structure reveals the copper centers to have a five-coordinate square pyramidal geometry, with the acetate ligands
bound terminally. The bridging phenolate occupies the apical position of the square-based pyramids and magnetic susceptibility,
electron paramagnetic resonance (EPR) and variable-temperature variable-field magnetic circular dichroism (MCD) measurements
indicate that the two centers are very weakly antiferromagnetically coupled (J = −0.6 cm−1). Simulation of the dipole–dipole-coupled EPR spectrum showed that in solution the Cu–O–Cu angle was increased from 126°
to 160° and that the internuclear distance was larger than that observed crystallographically. The high-resolution spectroscopic
information obtained has been correlated with a detailed ligand-field analysis to gain insight into the electronic structure
of the complex. Symmetry arguments have been used to demonstrate that the sign of the MCD is characteristic of the tetragonally
elongated environment. The complex also displays catecholase activity (k
cat = 15 ± 1.5 min−1, K
M = 6.4 ± 1.8 mM), which is compared with other dicopper catechol oxidase models.
