We have previously reported that lymphocytes from W/Fu rats immunized with syngeneic (C58NT)D tumor cells were cytotoxic against these cells in a 4-hr 51Cr release assay. We have investigated the feasibility of cryopreserving lymphocytes and target cells and have selected freezing conditions which provide good yields of viable cells and functional activity. Lymphocytes from different animals had a recovery of 60–80% viability which resulted in a corresponding 55–75% recovery of cytotoxic activity. Repeated testing of lymphocyte cytotoxicity from a pool of frozen spleen cells against either fresh or frozen (C58NT)D cells gave reproducible cytotoxicity. In addition, recovery of high levels of lymphocyte function was also demonstrated when cryopreserved cells were employed in long-term cytotoxic assays, i.e., 3H-proline and 125IUdR release assays, in the lymphoproliferative response to mitogens (PHA and Con A)3 or tumor cells (MLTI) as measured by 3H-thymidine incorporation, and in the in vitro generation of secondary cytotoxicity.By employing these cryoprotective techniques it is possible to have: 1) a population of lymphoid cells with known functional activity and 2) a pool of target cells with known susceptibility to lysis and antigenic content. Furthermore, the use of frozen cells as internal standards in each test also permits the analysis of assay variation as well as the study of variation in various cell types.  相似文献

An exo-β-glucan hydrolase, present in the digestive juice of the snail, Helix pomatia, has been purified to homogeneity by chromatography on Bio-Gel P-60, Sephadex G-200, DEAE-cellulose, and DEAE-Sephadex. The enzyme degrades β-(1 → 3)-linked oligosaccharides and polysaccharides, rapidly and to completion, or near completion, yielding glucose as the major product of enzyme action. Mixed linkage (1→3; 1→4)-β-glucans are also extensively degraded and β-(1→6)- and β-(1→4)-linked glucose polymers are slowly degraded by the enzyme. This enzyme differs from other exo-β-glucanases, reported previously, in the broadness of its substrate specificity. The Km values for action on laminarin and lichenin are respectively 1.22 and 2.22 mg/ml; the maximum velocity of action on laminarin is approximately twice that on lichenin. The enzyme has a molecular weight of 82,000 as determined by polyacrylamide gel electrophoresis. Maximum activity is exhibited at pH 4.3 and at temperatures of 50–55 °C.  相似文献

The essay reviews John Kendrew's pioneering work on the structure of myoglobin for which he shared the Nobel Prize for Chemistry in 1962. It reconstructs the status of protein X‐ray crystallography at the time Kendrew entered the field in 1945, after distinctive service in operational research during the war. It reflects on the choice of sperm whale myoglobin as research material. In particular, it highlights Kendrew's early use of digital electronic computers for crystallographic computations and the marshaling of other tools and approaches that made it possible to solve the structure at increasing resolution. The essay further discusses the role of models in structure resolution and their broader reception. It ends by briefly reviewing Kendrew's other contributions in the formation and institutionalization of molecular biology.  相似文献

We examined four species of sea pen (Anthoptilum grandiflorum, Halipteris finmarchica, Pennatula aculeata and Pennatula grandis) collected from the Gulf of St. Lawrence and mouth of the Laurentian Channel, eastern Canada. An exponential length–weight relationship was found for all four species, where growth in weight was progressively greater than growth in length with increasing colony size. Halipteris finmarchica, P. grandis and P. aculeata presented the better allometric fits, explaining over 80% of the variance. In addition, a count of growth increments visible in transverse sections in 86 A. grandiflorum and 80 P. aculeata samples was made. Presumed ages ranged between 5 and 28 years for A. grandiflorum and 2 and 21 years for P. aculeata. Radiocarbon assays were inconclusive and could not be used to confirm these ages; further age validation is required. Radial growth of the rod is slow during the first years, increasing at intermediate sizes of the colony and slowing down again for large colonies. Similar results were obtained from the relationship between colony length and number of growth increments where a logistic model was the best fit to the data. On average Spearman’s rank correlations showed 11% of shared variance between sea pen length or weight and environmental variables. Bottom temperature and salinity, depth and summer primary production were significantly correlated to sea pen size for most species.  相似文献

This historical minireview traces the development and application of methods for gene-targeted and site-directed mutagenesis of photosynthesis genes in cyanobacteria (mainly Synechocystis sp. PCC 6803). This approach allowed important data to be obtained on the structure and function of Photosystem I and Photosystem II complexes. I describe some of the major contributions of molecular genetics and subsequent mutant analysis in the 1980s and early 1990s that led to substantial advances in our knowledge of basic principles regarding the organization of the photosynthetic apparatus. This molecular-genetic research on cyanobacteria has initiated a fresh wave of photosynthesis research and created a solid foundation for rapid progress at the threshold of the twenty-first century. Photosynthetic activity and the composition of the photosynthetic apparatus are strongly regulated by environmental conditions. Some visually dramatic changes in pigmentation of cyanobacterial cells that occur during changing nutrient and light conditions reflect marked alterations in components of the major light-harvesting complex in these organisms, the phycobilisome. As noted well over 100 years ago, the pigment composition of some cyanobacteria is very sensitive to ambient wavelengths of light; this sensitivity reflects molecular changes in polypeptide constituents of the phycobilisome. The levels of different pigmented polypeptides or phycobiliproteins that become associated with the phycobilisome are adjusted to optimize absorption of excitation energy present in the environment. This process, called complementary chromatic adaptation, is controlled by a bilin-binding photoreceptor related to phytochrome of vascular plants; however, many other regulatory elements also play a role in chromatic adaptation. My perspectives and biases on the history and significance of this process are presented in this essay. A 67-year-old man underwent a third ablation procedure for a recurrent atrial tachycardia (AT) after an extensive pulmonary vein (PV) isolation, linear ablation along the left atrial (LA) roof and posterolateral mitral isthmus (MI), and defragmentation of persistent atrial fibrillation and an induced perimitral AT. High-resolution mapping during the clinical AT using the Rhythmia system (Boston Scientific) suggested that the AT was a ridge-related reentrant AT and exhibited a reconnection of the left PVs (LPVs). The residual electrograms in the posterior LPVs were surrounded by endocardial scar, which was like an island consisting of residual LPV electrograms. Retrograde venography of the vein of Marshall (VOM) demonstrated that the VOM reached the posterior left superior PV through the ridge between the LA appendage and left inferior PV and then the LPV carina. An ethanol infusion into the VOM resulted in a simultaneous AT termination and complete electrical isolation of the LPVs, that is, the disappearance of the residual LPV electrograms. The insular residual LPV electrograms in the present case did not appear to be endocardially connected to the LA, because the LPV electrograms were surrounded by endocardial scar and there was a large time gap between the earliest activation in the posterior LPVs and activation in the surrounding area. The VOM course on the venography and elimination of the residual LPV electrograms with an ethanol infusion into the VOM suggested that the insular residual LPV electrograms were electrically connected to the posterolateral LA via the VOM and its branches.  相似文献

Aim To address the biogeographical enigma of why guanacos (Lama guanicoe) are in the Falkland Islands we investigated the following questions: (1) What was the origin of the introduced guanacos? (2) What were the initial population sizes? (3) Why are they found only on one island? and (4) Who was John Hamilton and what role did he play? Location The Falkland Islands are located in the South Atlantic Ocean 600 km east of Patagonia at the southern end of South America. While dominated by East and West Falkland Islands, the archipelago is composed of some 750 islands. Sedge and Staats Islands, two small outlying islands of West Falkland, are the focus of this paper. Methods Historical information was collected from known relevant documents housed at the Falkland Islands Government Archives in Stanley, and personal interviews conducted with past and present residents of West Falklands. Research expeditions were made to Staats Island in 1999, 2002 and 2003 to assess the guanaco population size, distribution and social structure. Results Guanacos were unsuccessfully introduced in 1862 to East Falkland south of Mt Pleasant where Prince Alfred hunted them in 1871. John Hamilton, Scottish immigrant to the Falklands and Patagonia of southern Argentina and Chile, was the driving force in the introduction of guanacos from the region of Rio Gallegos, Argentina during the 1930s. The guanaco was one of several wildlife species he introduced, however, only the guanaco, Patagonia grey fox (Dusicyon griseus) and perhaps the sea otter (Lutra felina) survive. Hamilton's acting agent, Jimmy Miller, imported four shipments totalling 26 guanacos from 1934 to 1939. In 1934 the Falkland Government authorized Miller to introduce guanacos to Sedge Island, all 11 of which disappeared. Whether intentional or accidental, 15 guanacos were taken to Staats Island, an islet of 500 ha on the western edge of the archipelago. Historically, guanacos are unexpected on Staats Island because documentation authorizing their introduction is unknown. Guanaco numbers have fluctuated widely on Staats Island for 65 years primarily due to culling. In 1959 the population was dangerously close to extirpation, but today 400 thrive there. A severely reduced gene pool and genetic bottlenecking were suggested by recent field studies, revealing preliminary evidence of deleterious consequences of inbreeding. Main conclusions John Hamilton, spirited and visionary Scottish immigrant to the Falklands in the early 1880s, was responsible for the introduction of guanacos into the Falkland Islands. While there are some gaps in the historical events, the enigma of how and why guanacos were introduced to a single island in the South Atlantic Ocean is understood. Today, Staats Island, as a closed system, is a rare natural experiment in progress. It offers unique opportunities for addressing advanced questions in ungulate population, behavioural and genetic ecology. The population potentially also represents breeding stock for farming the guanaco's highly valuable wool on other islands. Thus, among his many efforts to practice land stewardship and promote economic diversity through the introduction of Patagonian wildlife, a remaining legacy of John Hamilton to the Falkland Islands is unmistakably the guanacos of Staats Island.  相似文献

This paper presents a brief introduction to some basic statistical tools and techniques with examples applied to biological data. Perhaps after becoming more familiar with statistics and more appreciative of their power in extracting information from data the previously fearful and skeptical researcher may be more inclined to make them an integral part of his research technique and also include them in his publications. For problems of greater difficulty, consultation at the early stages, preferably before undertaking the project, with a good biostatistician is suggested.  相似文献
