A natural lactic fermentation of mixtures of water and whole flour of either maize or high-tannin sorghum was obtained either before or after cooking to a weaning gruel: The preparations had a final pH of about 3.8 (range 3.67 to 4.00) and a ratio of lactic acid to acetic acid of 91 (w/w). The growth of added (about 107 c.f.u./g gruel) Gram-negative intestinal pathogenic bacteria, enterotoxigenicEscherichia coli, Campylobacter jejuni, Shigella flexneri andSalmonella typhimurium, was strongly inhibited in the sour gruels, and the effect could primarily be explained by the low pH caused by the formation of lactic and acetic acids during the fermentation process. Of the added Gram-positive bacteria,Bacillus cereus andStaphylococcus aureus showed similar inhibited growth up to 7h after inoculation in the sour gruels. The strain ofStaphylococcus, however, showed only a continued reduction in growth in the fermented gruel samples, which had a viable lactic bacteria culture indicating the presence of a bacteriocin. This implies that a low pH (< 4.0) alone is not sufficient to sustain the inhibition of the growth ofStaphylococcus aureus. 双歧杆菌的粘附特性及其对肠道致病菌的体外拮抗作用 总被引:2,自引:0,他引:2
本文采用体外细胞培养法, 从实验室现有的20株不同生境来源的双歧杆菌中筛选具有较强粘附能力的菌株, 并通过混合培养和牛津杯方法, 研究了具有粘附特性的双歧杆菌对肠道致病菌的体外拮抗作用。结果显示, 长寿老人源菌株A03和I06的粘附能力最强, 其菌液对金黄色葡萄球菌及大肠杆菌均具有显著的抑制性, 但是菌体及中和后的发酵液均没有抑菌性, 说明受试菌主要通过其代谢产物中的有机酸来发挥其抑菌性能; 此外, 通过分析比较受试菌和肠道致病菌分别与Caco-2细胞粘附后释放的乳酸脱氢酶量, 证实双歧杆菌与致病菌对细胞的作用具有本质上的区别, 双歧杆菌的粘附能减缓致病菌对细胞所造成的损害。 相似文献

In the present study, four Lactobacillus strains from the cheese were analyzed for its probiotic potential against enteropathogenic bacteria. The probiotic properties of the selected strains were also analyzed and the selected bacterial strains showed high tolerance in bile salts and organic acid. The strain L. plantarum LP049 showed maximum survival rate (92 ± 4.2% and 93.3 ± 2%) after 3 h of treatment at 0.25% (w/v) bile salts and 0.25% (w/v) organic acid concentrations. The ability of the Lactobacillus strains to adhere to human epithelial cells (HT-29 cell lines) was evaluated and L. plantarum LP049 showed maximum adhesion property (19.2 ± 1.1%) than other tested strains. The Lactobacillus strains produced lactic acid at various concentrations. Compared with other strains, maximum level of lactic acid (3.1 g/L), hydrogen peroxide (4.31 mM) and bacteriocin (31 AU/mg) was detected in LB049. The inhibitory activity of culture supernatant against various bacterial pathogens was observed. The zone of inhibition ranged between 6 ± 2 mm and 23 ± 2 mm. The cell free extract showed activity against, Escherichia coli (ATCC 10536), Salmonella enteritidis (ATCC 13076), Shigella flexneri (ATCC 29903), and Enterococcus faecium (ATCC 8459). Consequently, L. plantarum LP049 may be considered as a potential candidate for the production of novel bioactive metabolites for therapeutic and bio-protective applications. 相似文献

目的建立以多功能悬浮点阵技术为基础的临床常见肠道致病菌的快速检测方法。方法以细菌16S rDNA基因保守区序列设计1对通用引物,采用不对称PCR扩增7种临床常见肠道致病菌标准菌株,多功能悬浮点阵技术对不同菌株的PCR产物进行检测以验证相应菌种探针的特异性,最后对48份粪便标本进行肠道致病菌高通量快速检测。结果 7种临床常见肠道致病标准菌株的不对称PCR得到了大量单链产物,其产物用多功能悬浮点阵技术的检测特异性为100%,48份粪便标本不对称PCR产物可与相应探针发生特异性结合,且在多功能悬浮点阵技术的相应检测信号大于阴性对照3倍以上,5种细菌的多功能悬浮点阵技术检测结果与培养鉴定结果符合率100%,48份标本PCR产物均与志贺菌属探针发生杂交反应(阳性率100%)。结论 16S rDNA可以作为细菌快速鉴定的靶序列,不对称PCR产物可以显著提高与悬浮芯片杂交检测的灵敏度,多功能悬浮点阵技术在鉴定细菌方面具有简单快速、高通量、高检出率等特点,可以作为细菌快速鉴定的一种新方法,但无法鉴别志贺菌属和大肠埃希菌属。 相似文献

双歧杆菌QJ405、乳杆菌QJ405在试管内对肠道致病菌抑杀效果的初步报告 总被引:7,自引:2,他引:7
目的 观察双歧杆菌QJ405和乳杆菌QJ405在试管内对肠道致病的抑菌和杀菌效果。方法 抑菌实验:实验组,双歧杆菌QJ405和乳杆菌QJ405分别与各致病菌共同培养;对照组,实验组中的细菌各自单独培养。然后观察一定间隔时间(每2h或4h)细菌的数量变化,并绘制生长曲线。杀菌实验:将2种菌的培养上清液定量滴在已均匀涂布定量致病菌的平板表面,然后观察滴过上清液的区域内是否有细菌生长。结果:4种肠道致病菌和2株标准菌株(ATCC252922,ATCC25923)的生长明显受到抑制。乳杆菌QJ405的代谢产物有杀菌作用。结论 双歧杆菌QJ405和乳杆菌QJ405具有益生菌的性质。 相似文献
