Waterstriders on ponds and creeks compensate for displacement caused by wind or water current with periodical jumps against the direction of drift. This behavior is mediated by visual stimuli.
1.  Waterstriders are able to detect the direction and extent of their displacement on an artificial canal when visual cues are reduced to a single point-like light source above the upstream end of the canal. They adjust the frequency and direction of the compensatory jumps to keep their mean position on the canal constant. The mean amplitude of the jumps is constant and independent of the stream velocity.
2.  During drift compensation the light spot is kept in the frontal visual field at a fixed angle of elevation. This set angle differs among individuals and can be changed with time.
3.  The station-keeping ability can hardly be explained by a velocity servo system since there is no accumulating position error. The average compensatory movements are governed by a discontinuously working position servo.
4.  When the light spot is switched off, a few jumps are still performed. Jumps, therefore, are not triggered individually by visual cues. Waterstriders must possess an endogenous jump-generator.

A method has been developed for the rapid separation of cells in suspension from non-cell associated lipid vesicles in various assays for vesicle-cell interaction. Separation is achieved on a discontinuous Ficoll-Paque gradient. Cells and free vesicles are totally separated, as evidenced by both radiolabelled vesicles, and vesicles containing the fluorescent dye 6-carboxyfluorescein. The main advantages of this method are the rapidity, efficacy, and gentleness of the separation. Viability of the cells remains consistently high (greater than 96%) throughout the separation. Since this method involves a one-step centrifugation, it precludes the necessity for repeated washings of cells which have been incubated with lipid vesicles.  相似文献

Many flightless beetles like the large apterous dung beetle Circellium bacchus, possess a subelytral cavity (SEC) providing an extra air space below the elytra which connects to the tracheal system (TS) via metathoracic and abdominal spiracles. By measuring subelytral and intratracheal pressure as well as body movements and gas exchange simultaneously in a flow-through setup, we investigated the contribution of convection on Circellium respiratory gas exchange.No constriction phase was observed. TS and SEC pressures were always around atmospheric values. During interburst phase open abdominal spiracles and a leaky SEC led to small CO2-peaks on a continuous CO2 baseline, driven by intermittent positive tracheal pressure peaks in anti-phase with small negative subelytral pressure peaks caused by dorso-ventral tergite action.Spiracle opening was accompanied by two types of body movements. Higher frequency telescoping body movements at the beginning of opening resulted in high amplitude SEC and TS pressure peaks. High frequency tergite movements caused subelytral pressure peaks and led to a saw tooth like CO2 release pattern in a burst. We propose that during the burst open mesothoracic spiracles increase the compliance of the subelytral cavity allowing big volumes of tracheal air being pulled out by convection.  相似文献

Flightless, diurnal tenebrionid beetles are commonly found in deserts. They possess a curious morphological adaptation, the subelytral cavity (an air space beneath the fused elytra) the function of which is not completely understood. In the tenebrionid beetle Eleodes obscura, we measured abdominal movements within the subelytral cavity, and the activity of the pygidial cleft (which seals or unseals the subelytral cavity), simultaneously with total CO2 release rate and water loss rate. First, we found that E. obscura has the lowest cuticular permeability measured in flow-through respirometry in an insect (0.90 microg H2O cm(-2) Torr(-1) h(-1)). Second, it does not exhibit a discontinuous gas exchange cycle. Third, we describe the temporal coupling between gas exchange, water loss, subelytral space volume, and the capacity of the subelytral space to exchange gases with its surroundings as indicated by pygidial cleft state. Fourth, we suggest possible mechanisms that may reduce respiratory water loss rates in E. obscura. Finally, we suggest that E. obscura cannot exchange respiratory gases discontinuously because of a morphological constraint (small tracheal or spiracular conductance). This "conductance constraint hypothesis" may help to explain the otherwise puzzling phylogenetic patterns of continuous vs. discontinuous gas exchange observed in tracheate arthropods.  相似文献

In this research, the B3LYP density functional and Stevens effective core potentials are used to compare carbon-hydrogen and carbon-heteroatom bond activation by an iridium(I) complex. Of particular importance is to address the kinetic (transition state) and thermodynamic (ground state) selectivity. The complex Ir(PH3)2(H) with CH3-X (X=F, Cl, OH, SH, NH2, PH2) as the substrate has been used as a model. Good agreement in geometries is obtained between the target molecules and experimental models. The resultant products of C-H and C-X oxidative addition are Y-shaped minima (i.e., a distorted trigonal bipyramid with one acute and two obtuse angles among the equatorial ligands). Oxidative addition of the C-X bond to the substrate is exothermic for groups 16 and 17, but endothermic for group 15. A significant thermodynamic preference for C-X activation over C-H activation is observed for these Ir(I) complexes. However, analysis of the transition states for oxidative addition suggests that there is a kinetic preference for C-H activation.  相似文献

South American subtropical dry forests are highly threatened by fragmentation. Despite considerable research efforts aimed at predicting ecosystem alterations due to this driver of global change, we still need to deal with general principles to improve our ability to predict the impact of fragmentation. Our work is one of the few studies that analyse the relationship between forest fragmentation and decomposition. In 12 remnants of Chaco Serrano forest in Central Argentina we tested if decomposition rates of a common leaf-litter substrate varied with fragment size and between the forest edge and interior. Decomposition declined with fragment size, with no significant effects of location (edge/interior) or interaction between the two components of fragmentation. Our results suggest that in situ conditions for decomposition may change as a consequence of forest fragmentation, specifically as a result fragment size. This may lead to impaired nutrient recycling in smaller forest remnants.  相似文献

A simulation rumen model has been developed to function under non-steady state conditions in order to allow prediction of nutrient availability in dairy cows managed under discontinuous feeding systems. The model simulates availability of glycogenic, aminogenic and lipogenic nutrients to lactating dairy cows fed discontinuously. The model structure considers input of up to three different feeds fed independently at any time during the day. Feeds are described by their nitrogen (N), carbohydrate and fatty acid fractions. The N containing feed fractions include ruminally undegraded crude protein (CP), ruminally insoluble but potentially degradable CP, ruminally soluble CP and ammonia N. The feed carbohydrate fractions include ruminally undegradable neutral detergent fibre (NDF), ruminally degradable NDF, ruminally insoluble starch, ruminally soluble starch and sugars. The fatty acids in the feeds are divided between long chain fatty acids and volatile fatty acids (VFA). Additionally four pools were defined representing absorption of amino acids, glucose, long chain fatty acids and volatile fatty acids. The rumen microbial population is represented as a single pool. Besides a flexible structure, new features to the extant model include adoption of the concept of chewing efficiency (or chewing effectiveness) during eating, variable fractional ruminal absorption rates of VFA and variable fractional ruminal degradation rates of NDF as a function of rumen liquid pH, as well as a variable rumen volume which directly affects rumen concentrations of metabolites. The model continuously (i.e., by minute) predicts release of soluble components from the feeds in the rumen, concentration and absorption of fermentation end products in the rumen, rumen pools of nutrients and microbial biomass dynamics, as well as passage of microbial biomass and non-fermented nutrients from the rumen, in response to various feeding strategies. Model evaluation covered a wide range of feeding strategies that included pasture and housed feeding systems. Overall, the mean square prediction error (MSPE) as a percentage of the observed mean was relatively low (<10%) with a high amount of the total variation explained by random variation (>65%). Deviation from unity varied between 23% (rumen dry matter content) and 25% (NDF), indicating some consistent over and/or under prediction. A more detailed evaluation was done based on studies available that reported diurnal behaviour of key model outputs such as rumen pools, rumen pH, and rumen VFA. The predictions broadly simulated the observed values quantitatively, relative to general diurnal patterns, and relative to differences between treatments in the predicted diurnal patterns. Results show that the model provides a tool to assess potential outcomes of changing feeding strategies which may be particularly valuable in assessing selection of feeds, amounts and times of the day to offer the feeds. The continuous nature of the simulated output also allows determination of the time(s) of the day that ruminal (and/or post-ruminal) delivery of nutrients may limit ruminal output of nutrients (and/or availability of nutrients) to support milk nutrient synthesis.  相似文献

Starch is a reserve chemical source of the energy of the sun found in plants as a water-insoluble granule that differs in their chemical and physical properties, depending on the source. The granules can be solubilized by heating in water or by treatment with various reagents, such as 1M NaOH. alpha-Amylases are widely distributed enzymes that initiate the hydrolysis of starch into low molecular weight maltodextrins. We recently found that the activities of a single alpha-amylase on two different starches were significantly different. We then determined the activities of Bacillus amyloliquefaciens and porcine pancreas alpha-amylases, using eight different starches, solubilized by two methods: autoclaving at 121 degrees C and 1M NaOH at 20 degrees C. There were significant differences in the activities of both of the amylases on all eight of the starches. Previously, it had been found that polyethylene glycol (PEG) stabilized and activated the activities of both enzymes, using a soluble amylose as the substrate. Addition of PEG to the enzymes greatly increased the activities on the eight starches, but the activities still differed significantly. The different activities with the starches were hypothesized as differences in the amounts of secondary and tertiary structures that are partially retained when the different starches are solubilized; the activities on addition of PEG is hypothesized as the formation of highly active species from a series of less active forms.  相似文献

We perform a bifurcation analysis of the mathematical model of Jones and Kompala [K.D. Jones, D.S. Kompala, Cybernetic model of the growth dynamics of Saccharomyces cerevisiae in batch and continuous cultures, J. Biotechnol. 71 (1999) 105-131]. Stable oscillations arise via Andronov-Hopf bifurcations and exist for intermediate values of the dilution rate as has been noted from experiments previously. A variety of discontinuity induced bifurcations arise from a lack of global differentiability. We identify and classify discontinuous bifurcations including several codimension-two scenarios. Bifurcation diagrams are explained by a general unfolding of these singularities.  相似文献
