用胎球蛋白-Sepharose亲和层析和凝胶过滤层析从精子和精浆中分离纯化了猪精子凝集素(简称BSL)。BSL的血凝活性只被若干糖蛋白和聚糖所抑制。BSL的分子量为56kd,由分子量分别为13.6kd(β)和16.0kd(α)的两个不同的亚基以α1β3所组成。BSL为糖蛋白,含中性糖3.2%,不含唾液酸。用ELISA法测定猪精子中BSL的含量及分布,表明70%嵌入在精子膜中,25%结合在精子表面, 相似文献

Summary Cell suspensions of the fresh-watersponge Ephydatia fluviatilis have been fractionated by means ofFicoll gradient centrifugation. Three fractions were isolated. The densest contains archeocyte-like cells only; the intermediate fraction is very rich in choanocytes, and the lightest is a mixture of cell types. The distribution pattern of ten species of Collembola was studied during the four years period from July 1971 to May 1975 in a pine forest soil. The distribution patterns were analysed for two scales of distribution, i. e., the distribution over the plot of 10×10 m2 and the micro-distribution within a block sample consisting of 36 contigious units each 2×2 cm2 in area, by applying the -m regression method. The fundamental pattern which appeared was quite similar for the species examined and individuals were aggregated in response to the heterogeneity of habitat conditions. The causes of aggregations were discussed with regard to some environmental factors. The relative abundances of 10 species within the collembolan community was examined in relation to the habitat utilization and the relative abundance was not related to the degree of aggregation but rather to the area occupied by individuals. This suggests that the more numerically abundant species tend to occupy broader micro-habitat. Biological meaning of aggregation was discussed in connection with the population biology and community organization of collembola. 相似文献

The modified Blalock-Taussig shunt is a surgical procedure used as a palliation to treat complex congenital heart defects. It consists of an interposing prosthetic tube between the innominate/subclavian artery and the right pulmonary artery. Previous experience indicates that the pressure drop across the shunt is affected by the pulmonary pressure at the distal anastomosis combined with the distensibility of the anastomosis. In this study, a computational fluid-structure interaction approach is presented to investigate the haemodynamic behaviour. Steady-state fluid dynamics and structural analyses were carried out using commercial codes based on the finite element method (FIDAP and ABAQUS) coupled by means of a purposely-developed procedure to transfer boundary conditions. Both prosthetic tube and artery walls were characterised by non-linear material properties. Three different pulmonary pressures (2, 5 and 15 mmHg) and two volume flow rates (0.4 and 0.8 l/min) were investigated. Results indicate that the effects of distensibility at the distal anastomosis on the shunt pressure drop are relevant only when the distal anastomosis on the shunt pressure drop are relevant only when the distal anastomosis is not fully distended, which occurs when the pulmonary pressure is lower than 5 mmHg. 相似文献

猪骨骼肌磷酸葡萄糖异构酶的提纯及性质研究 总被引:3,自引:0,他引:3
本文报道猪骨骼肌磷酸葡萄糖异构酶(GPI.EC 5.3.1.9)的提纯及其性质。设计了四步提纯法(稀盐溶液抽提、有机溶媒沉淀、透析、葡聚糖G-100柱层析),对猪的五种不同解剖部位的骨骼风进行了GPI的提纯。结果均得到聚丙烯酰胺凝胶电泳(PAGE)图谱为一条带的产品。提纯效果以猪股二头肌为例:提纯10.68倍、活力回收32.37%,比活力1.6×105单位。对提纯酶性质的研究表明;猪肌提纯的GPI由三个亚基组成。分子量约为120Kd,等电点为 6. 6,最适pH8.5,最适温度 35℃,米氏常数:6.02mmol/L,活化能为32378.4焦耳/K,mol。免疫电泳实验证实猪肌提纯酶具抗原性。其免疫血清与自牛、兔、鸡、鱼的骨骼肌用同法提纯的GPI有交叉免疫反应。免疫血清且对原酶液有抑制作用。猪肌GPI与其它数种动物肌肉之GPI的PAGE行为、混合电泳行为,等电聚焦行为、氨基酸组成等各方面基本相同,可以认为这是猪肌GPI与其它数种动物肌肉的GPI为同源蛋白质的佐证。 相似文献

We investigated the effect of aluminium (Al 3+ ) on lipid peroxidation and physico-chemical properties of high density lipoproteins (HDL) isolated from human plasma. Our results demonstrated that Al 3+ enhances lipid peroxidation of human HDL as shown by the significant increase in lipid hydroperoxides in Al-treated HDL with respect to control HDL. The oxidative effect was higher at acid pH (pH 5.5) with respect to pH 7.4. Moreover, a stimulating effect of Al 3+ on iron-induced lipid peroxidation of HDL was demonstrated. The study of the effect of Al 3+ on the physico-chemical properties of HDL, using the fluorescence polarization (Pf) of the probes TMA-DPH (1-(4-trimethylammoniumphenyl)-6-phenyl-1,3,5-hexatriene iodide) and DPH (1,6-diphenyl-1,3,5-hexatriene), showed a significant decrease of Pf in Al-treated HDL with respect to control. These results suggest that Al 3+ induces a decrease of molecular order at the lipoprotein surface. Moreover, the study of tryptophan (Trp) fluorescence demonstrated that aluminium induces structural modifications of HDL apoproteins and on HDL physico-chemical properties. The effect of Al 3+ on lipid peroxidation of HDL was observed at aluminium concentrations similar to those observed in the brain of patients affected by neurological diseases. An extracellular α-amylase produced by the thermophilic bacterium Thermus filiformis Ork A2 was purified from cell-free culture supernatant by ion exchange chromatography. The molecular mass was estimated to
be 60 000 Da by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The enzyme was rich in both basic and hydrophobic
amino acids, presenting the following NH2-terminal amino acid sequence: Thr-Ala-Asp-Leu-Ile-Val-Lys-Ile-Asn-Phe. Amylolytic activity on soluble starch was optimal
at pH 5.5–6.0 and 95°C, and the enzyme was stable in the pH range of 4.0–8.0. Calcium enhanced thermostability at temperatures
above 80°C, increasing the half-life of activity to more than 8 h at 85°C, 80 min at 90°C, and 19 min at 95°C. Ethylenediaminetetraacetic
acid (EDTA) inhibited amylase activity, the inhibition being reversed by the addition of calcium or strontium ions. The α-amylase
was also inhibited by copper and mercuric ions, and p-chloromercuribenzoic acid, the latter being reversed in the presence of dithiothreitol. Dithiothreitol and β-mercaptoethanol
activated the enzyme. The α-amylase exhibited Michaelis-Menten kinetics for starch, with a K
m of 5.0 mg·ml−1 and k
cat/K
m of 5.2 × 105 ml·mg−1 s−1. Similar values were obtained for amylose, amylopectin, and glycogen. The hydrolysis pattern was similar for maltooligosaccharides
and polysaccharides, with maltose being the major hydrolysis product. Glucose and maltotriose were generated as secondary
products, although glucose was produced in high levels after a 6-h digestion. To our knowledge this is the first report of
the characterization of an α-amylase from a strain of the genus Thermus.

A large family of cysteine-rich secretory proteins (CRISPs) includes proteins of different origin, the function of the majority of CRISPs being unknown. For CRISPs isolated from snake venom, two types of activities were found: two proteins blocked cyclic nucleotide-gated ion channels, several others blocked potassium-stimulated smooth muscle contraction. Thus, snake CRISPs represent potentially valuable tools for studies of ion channels, which makes promising a search for new CRISPs. Here we report on the isolation of several novel CRISPs from the venoms of Asian cobra Naja kaouthia and African cobra Naja haje using a combination of different types of liquid chromatography. Four CRISP variants were identified in N. kaouthia venom and three proteins, one of them acidic, were found in N. haje venom. Acidic CRISP was found in a reptilian venom for the first time. Our data suggest that each cobra venom contains a pool of different CRISPs. 相似文献

Effects of extrusion and glycerol content on properties of oxidized and acetylated corn starch-based films 总被引:2,自引:0,他引:2
Oxidized and acetylated corn starch-based films were prepared by casting with glycerol as a plasticizer. The present study investigated the effects of extrusion prior to film-making and glycerol content on the properties of starch films. The films with extrusion exhibited lower tensile strength, higher elongation at break, higher water vapor permeability and higher oil permeability than those without extrusion. Extrusion reduced heat sealability of the films. With the increase of glycerol content, the films became more flexible with higher elongation at break and lower tensile strength. Water vapor permeability, oil permeability and the range between the onset temperature and the melt peak temperature rose as glycerol content increased. The thermograms indicated that plasticizers and biopolymers were compatible. These results suggested that extrusion did no good to starch films while glycerol content had apparent effect on the mechanical and barrier properties of the films. 相似文献
