通过优化高产奶牛体细胞克隆胚胎体外生产技术条件,制备高质量的奶牛克隆胚胎,旨在提高奶牛体细胞核移植产业化应用效率。就受体卵母细胞去核方法、不同年龄供体牛细胞来源、血清饥饿与否以及不同气相组成培养等条件对奶牛体细胞克隆胚胎生产效率的影响进行了研究和探讨。结果表明,虽然荧光染色辅助去核和盲吸法的去核率、囊胚发育率分别为100%、24.83%和92.44%、28.26%,两者之间无显著差异(P>0.05),但盲吸法操作简单、效率高;不同年龄来源供体牛的细胞系构建的克隆胚胎的囊胚发育率分别为31.43%、25.68%,两者之间没有显著差异(P>0.05);经血清饥饿和未饥饿供体细胞重构的克隆胚胎囊胚发育率分别为24%、29.9%,两者之间没有显著差(P>0.05);富氧和低氧气相培养的克隆胚胎的囊胚发育率分别为28.26%、31.55%,两者之间差异不显著(P>0.05),低氧气相组成更有利于囊胚的发育。根据上述结果,奶牛体细胞核移植胚胎(克隆胚胎)的产业化生产条件为:供体细胞无需进行同期饥饿处理,直接注入到盲吸去核后的受体卵子透明带下构建克隆胚胎,融合后的克隆胚胎在密封的混合三气(5%CO2-5%O2-90%N2)的气相组成下进行体外培养,能保持稳定的囊胚发育率。 相似文献

Myofibrillogenesis, the process of sarcomere formation, requires close interactions of sarcomeric proteins and various components of sarcomere structures. The myosin thick filaments and M-lines are two key components of the sarcomere. It has been suggested that myomesin proteins of M-lines interact with myosin and titin proteins and keep the thick and titin filaments in order. However, the function of myomesin in myofibrillogenesis and sarcomere organization remained largely enigmatic. No knockout or knockdown animal models have been reported to elucidate the role of myomesin in sarcomere organization in vivo. In this study, by using the gene-specific knockdown approach in zebrafish embryos, we carried out a loss-of-function analysis of myomesin-3 and slow myosin heavy chain 1 (smyhc1) expressed specifically in slow muscles. We demonstrated that knockdown of smyhc1 abolished the sarcomeric localization of myomesin-3 in slow muscles. In contrast, loss of myomesin-3 had no effect on the sarcomeric organization of thick and thin filaments as well as M- and Z-line structures. Together, these studies indicate that myosin thick filaments are required for M-line organization and M-line localization of myomesin-3. Pluripotent stem cells derived from neonatal or adult testes are a useful tool to examine the mechanisms of pluripotency and a resource for cell-based therapies. However, therapies usingthese cells will only benefit males but not females. Recently, female germline stem cells (FGSCs) were discovered in ovaries. Whether FGSCs can be converted into pluripotent stem cells, similar to spermatogonial stem cells, is unknown. Here, we demonstrate that female embryonic stem-like cells (fESLCs) can be generated within 1 month from the stably proliferating FGSCs cultured in embryonic stem cell (ESC) medium, fESLCs exhibit properties similar to those of ESCs in terms of marker expression and differentiation potential. Thus, our findings suggest that generation of patient-specific fESLCs is feasible and provides a foundation for personalized regenerative applications. 相似文献

The present experiments were designed to study the effects of glucose, EDTA, glutamine on the in vitro development of single blastomeres from 2-cell embryos in mouse, and the efficiency of cryopreservation of blastocysts from single blastomers with different vitrification. Single blastomeres derived from female ICR x male BDF1 2-cell embryos were cultured in mKRB with or without glucose, EDTA and glutamine, respectively. The expanded blastocyst rates were significantly different between in mKRB with glucose and without glucose (34% vs 65%); The blastomeres were cultured in mKRB with EDTA and glutamine but glucose, the expanded blastocyst rate (90%) was significantly higher than other groups. The blastocysts derived from single blastomeres were vitrified in liquid nitrogen after equilibration in GFS40 for 0.5-2 min, the survival rate 24%-51%. The blastocysts were pretreated in mPBS with 10% glycerol for 5 min, followed by exposure to GFS40 at 25 degrees C for 0.5 min, then vitrified in liquid nitrogen(two-step method), the survival rate was 61%. However, the survival rates increased to 64% and 70% when the blastocysts were vitrified(one-step method) ater equilibration in EFS40 at 25 degrees C for 0.5-1 min. 相似文献
