The garlic stalk is a byproduct of garlic production and normally abandoned or burned, both of which cause environmental pollution. It is therefore appropriate to determine the conditions of efficient decomposition, and equally appropriate to determine the impact of this decomposition on soil properties. In this study, the soil properties, enzyme activities and nutrient dynamics associated with the decomposition of garlic stalk at different temperatures, concentrations and durations were investigated. Stalk decomposition significantly increased the values of soil pH and electrical conductivity. In addition, total nitrogen and organic carbon concentration were significantly increased by decomposing stalks at 40°C, with a 5∶100 ratio and for 10 or 60 days. The highest activities of sucrase, urease and alkaline phosphatase in soil were detected when stalk decomposition was performed at the lowest temperature (10°C), highest concentration (5∶100), and shortest duration (10 or 20 days). The evidence presented here suggests that garlic stalk decomposition improves the quality of soil by altering the value of soil pH and electrical conductivity and by changing nutrient dynamics and soil enzyme activity, compared to the soil decomposition without garlic stalks.  相似文献

Swertia tetraptera Maxim is an annual alpine herb endemic to the Qinghai-Tibetan Plateau (QTP). Its populations are locally scattered as isolated patches throughout this region. Genetic variation within and among thirty-four populations of this species was assessed using ISSR fingerprinting with 10 primers. High levels of genetic diversity exist within species (P = 98.9%, I = 0.3475; He = 0.2227), while the within-population diversity is low (P = 32.7%, I = 0.177; He = 0.12). High levels of genetic differentiation were detected among populations based on various statistics, including Nei’s genetic diversity analysis (GST = 0.4608), Bayesian analysis (θB = 0.476) and AMOVA (FST = 0.57). That is, populations shared low levels of genetic identity (I = 0.2622–0.0966). This genetic structure was probably due to severe genetic drift, breeding system and limited gene flow. The observed genetic structure of the populations implies that different populations across the distribution range of the species should be sampled to maintain high genetic diversity when a conservation strategy is implemented.  相似文献

Barrier islands shrub thickets, the dominant woody community of many Atlantic coast barrier islands, are very sensitive to changes in the freshwater lens and thus, constitute a strong indicator of summer drought. NDVI was computed from airborne images and multispectral images on Hog Island (VA, USA) to evaluate summer growing season changes in woody communities for better predictions of climate change effects. Patterns of NDVI were compared year to year and monthly relative to precipitation and water table depth at the appropriate temporal scale. The highest absolute values of NDVI as well as the larger surface covered by woody vegetation (NDVI > 0.5) occurred in the wet year (2004) with a bimodal distribution of NDVI values (around 0.65 and 0.9) while both dry years (2007 and 2008) showed similar values in maximum, mean and standard deviation and unimodal distributions (around 0.75) of NDVI values. Positive linear adjustments were obtained between maximum (r2 > 0.9) and mean NDVI (r2 > 0.87) and the accumulated rainfall in the hydrological year and the mean water table depth from the last rainfall event till the date of the image acquisition. The spatial variations revealed that water table depth behaved different in wet and dry years. In dry years there was a remarkable increase in mean and maximum values linearly related to water table depth. The highest slope of the adjustment in 2007 indicated a sharp response of vegetation in the driest year. Monthly series of NDVI showed the major role of lack of precipitation through July and August in 2007 with missing classes of NDVI above 0.8 and unimodal distributions in mid-late summer. Best linear fits (r2 close to 1) were obtained with rainfall at different temporal scales: accumulated rainfall in the hydrological year 2004 and accumulated rainfall in the last month previous to the date of 2007 image. Thus, in dry years productivity is closely related to water available from recent past as opposed to over the year for wet years. Good fits (r2 values higher than 0.88) were obtained between monthly decrease in water table depth and NDVI variables just in the dry year. These results demonstrate the important feedback between woody vegetation response to changes in the freshwater lens using empirical data and could apply to other systems with strong directional gradients in resources.  相似文献

Nitric oxide synthase (NOS) may be uncoupled to produce superoxide rather than nitric oxide (NO) under pathological conditions such as diabetes mellitus and insulin resistance, leading to cardiac contractile anomalies. Nonetheless, the role of NOS uncoupling in insulin resistance-induced cardiac dysfunction remains elusive. Given that folic acid may produce beneficial effects for cardiac insufficiency partially through its NOS recoupling capacity, this study was designed to evaluate the effect of folic acid on insulin resistance-induced cardiac contractile dysfunction in a sucrose-induced insulin resistance model. Mice were fed a sucrose or starch diet for 8 weeks before administration of folic acid in drinking water for an additional 4 weeks. Cardiomyocyte contractile and Ca2+ transient properties were evaluated and myocardial function was assessed using echocardiography. Our results revealed whole body insulin resistance after sucrose feeding associated with diminished NO production, elevated peroxynitrite (ONOO) levels, and impaired echocardiographic and cardiomyocyte function along with a leaky ryanodine receptor (RYR) and intracellular Ca2+ handling derangement. Western blot analysis showed that insulin resistance significantly promoted Ca2+/calmodulin-dependent protein kinase II (CaMKII) phosphorylation, which might be responsible for the leaky RYR and cardiac mechanical dysfunction. NOS recoupling using folic acid reversed insulin resistance-induced changes in NO and ONOO, CaMKII phosphorylation, and cardiac mechanical abnormalities. Taken together, these data demonstrated that treatment with folic acid may reverse cardiac contractile and intracellular Ca2+ anomalies through ablation of CaMKII phosphorylation and RYR Ca2+ leak.  相似文献

Recent studies have suggested that the long distance movements of some terrestrial mammals are not migratory, but rather nomadic. Moreover, the spatial heterogeneity and temporal predictability of resources were proposed as factors contributing to alternative movement strategies, such as sedentarism (i.e., range residency), migration, and nomadism. Here, we propose that, at the individual level, a dependence on spatial memory is another important parameter for distinguishing among population-level patterns of spatial distribution. For instance, migratory animals would have a long memory of the areas they prefer to revisit, whereas nomadic animals would remember recently visited areas as places to avoid as they search for resources. We develop a computational model in which individuals’ movement decisions are based on the animals’ spatial memory of previously visited areas. Through this approach, we delineate how the interplay between landscape persistence and spatial memory leads to sedentarism, migration, and nomadism.  相似文献

Biofilm development in urinary tract catheters is an often underestimated problem. However, this form of infection leads to high mortality rates and causes significant costs in health care. Therefore, it is important to analyze these biofilms and establish avoiding strategies. In this study a continuous flow-through system for the cultivation of biofilms under catheter-associated urinary tract infection conditions was established and validated. The in vitro urinary tract catheter system implies the composition of urine (artificial urine medium), the mean volume of urine of adults (1 mL min–1), the frequently used silicone catheter (foley silicon catheter) as well as the infection with uropathogenic microorganisms like Pseudomonas aeruginosa. Three clinical isolates from urine of catheterized patients were chosen due to their ability to form biofilms, their mobility and their cell surface hydrophobicity. As reference strain P. aeruginosa PA14 has been used. Characteristic parameters as biofilm thickness, specific biofilm growth rate and substrate consumption were observed. Biofilm thicknesses varied from 105 ± 16 μm up to 246 ± 67 μm for the different isolates. The specific biofilm growth rate could be determined with a non invasive optical biomass sensor. This sensor allows online monitoring of the biofilm growth in the progress of the cultivation.  相似文献

Compared to immobilized lipase, soluble lipase has the merits of lower cost and faster reaction rate, thus much attention has been paid to soluble lipase-mediated methanolysis for biodiesel (fatty acid methyl ester, FAME) production in recent years. Our previous study showed that soluble lipase NS81006 could effectively catalyze the methanolysis of soybean oil (triglyceride, TG) for FAME preparation in oil/water biphasic system. Study on the related mechanism of soluble lipase NS81006-mediated methanolysis of TG was carried out in this paper. Based on the analysis of substances change in the reaction process, mechanism model was hypothesized and the model parameters were simulated by Matlab. The simulated model was validated further. The results showed that in the reaction process of soluble lipase NS81006-mediated methanolysis of TG in oil/water biphasic system, TG proceeded three-step hydrolysis to generate FFA (free fatty acid), and then FFA transformed into FAME by esterification with methanol. During the whole process, FFA is mainly generated through the hydrolysis of TG and intermediate DG (diglyceride), while the hydrolysis of FAME could be ignored.  相似文献

An iso-random Bi Bi mechanism has been proposed for adenylate kinase. In this mechanism, one of the enzyme forms can bind the substrates MgATP and AMP, whereas the other form can bind the products MgADP and ADP. In a catalytic cycle, the conformational changes of the free enzyme and the ternary complexes are the rate-limiting steps. The AP(5)A inhibition equations derived from this mechanism show theoretically that AP(5)A acts as a competitive inhibitor for the forward reaction and a mixed noncompetitive inhibitor for the backward reaction.  相似文献
