Highly viscous polysaccharide (250–350 kDa) of an alginate nature with a predominance of α-L-guluronic acid (M/G = 0.22) was obtained from Azotobacter vinelandi. The yield of polysaccharide was 20.5 ± 0.5 g/l when cultured in a medium containing molasses at a viscosity of the cultural liquid of over 30000 cSt. The biopolymer is stable at pH 4.0–9.0 in a wide temperature range and well soluble in highly mineralized water; it retains a high viscosity level and can be used in the petroleum industry for enhanced oil recovery.  相似文献

本文报导了胰腺提取物中两种可被钙/钙调素依赖性蛋白激酶磷酸化的热稳定蛋白。SDS-PAGE测定其表观分子量分别为17.7kD和6kD。经钙/钙调素依赖性蛋白激酶磷酸化后,其最大磷酸参入量为8.8μmol/g蛋白。同时磷酸化作用导致17.7kD蛋白在SDS-PAGE中迁移率发生变化。本文还进一步分析了各种阳离子对磷酸化的影响,并对此两种蛋白可能具有的生理功能进行了初步探讨。  相似文献

本文介绍用二相分配法制备蚕豆叶片原生质膜上的Ca~(2+)·Mg~(2+)-ATPase,用以研究镧系,稀土离子对此酶活性的影响。初步证实Pr~(3+)、Nd~(3+)对依赖于CaM的以及不依赖于CaM的蚕豆叶片原生质膜上Ca~(2+)·Mg~(2+)-ATPase活性的抑制不是CaM专一的。  相似文献

Adventitious root formation in excised cucumber (Cucumis sativus L.) cotyledons was significantly promoted by (±)-cis-chrysanthemic acid at 0.006–1.8 mM. The effect of (±)-cis-chrysanthemic acid on indole-3-acetic acid (IAA)-induced rooting was additive. Rooting in excised cucumber cotyledons was significantly promoted by several isomers of chrysanthemic acid and sodium (±)-cis-chrysanthemate at 0.18 mM. Rooting in mung bean (Phaseolus radiatus L.) hypocotyls was also stimulated by the sodium salt at 0.06–0.6 mM. Rooting of kidney bean (Phaseolus vulgaris L.) hypocotyls was also clearly enhanced by sodium (±)-cis-chrysanthemate at 0.18–6 mM.  相似文献

The specificities of extracellular and ribosomal serine proteinase from Bacillus natto, a food microorganism, were investigated. Both proteins have highly restricted and characteristic specificities. With the extracellular serine proteinase, initial cleavage site was observed at Leu15-Tyr16, secondary site at Ser9-His10 and additional cleavage sites at Gln4-His5 and His5-Leu6 in the oxidized insulin B-chain. Hydrolysis of proangiotensin with the extracellular serine proteinase was observed primarily at Phe8-His9 and secondary at Tyr4-Ile5. The extracellular serine proteinase has a Km of 0.08 mM and kcat of 3 s−1 for angiotensin hydrolysis. With the ribosomal proteinase, initial cleavage site of the oxidized insulin B-chain was observed at Leu15-Tyr16 and additional cleavage site at Phe24-Phe25. Hydrolysis of proangiotensin was observed at Tyr4-Ile5 bond with the ribosomal proteinase.  相似文献

生态能质(eco-exergy)是指系统从给定状态达到热力学平衡状态所做的功,可以度量生态系统的复杂生物化学组分及生态结构。系统具有的能质越大,有序化程度越高,稳定性也越强。生态能质和比生态能质(specific eco-exergy)指标能够评价水生生态系统健康状况,对水生生态系统演替阶段具有指示作用。本文阐述了生态能质的定义、生态学意义以及生态能质值和权重因子fi值的计算方法。对生态能质作为功能函数在水生生态系统结构动力学模型(structural dynamic models,SDMs)中以及作为生态指标在生态系统健康评价、生态恢复研究中的实际应用进行了总结。最后探讨了生态能质指标的局限性以及未来的发展。此外,文中建议将eco-exergy和specific eco-exergy统一翻译为生态能质和比生态能质。  相似文献

益生菌生物药物是指通过口服表达药用多肽(蛋白)的重组益生菌活细胞达到治疗疾病的新型口服给药系统。为了构建一种能有效防治2型糖尿病的酵母生物药物,文章首先构建了酿酒酵母(S.cerevisiae)整合型表达载体pNK1-PGK,并且通过绿色荧光蛋白(GFP)证明其表达功能正常,利用该载体将10×GLP-1 (Glucagon-like peptide-1)基因转化到酿酒酵母INVSc1中,通过营养缺陷型和Western blotting成功筛选出表达10×GLP-1的长效促胰岛素降糖酵母(Long-acting GLP-1 hypoglycemic yeast, LHY)。该酵母生长迅速,外源基因10×GLP-1表达稳定,表达量达到1.56 mg/g细胞湿重。通过链脲佐菌素和高脂高糖饮食联合诱导的方法构建了2型糖尿病小鼠模型,用LHY对其进行口服灌胃治疗,证明LHY具有较好疗效,明显降低血糖水平。  相似文献

The major flavonoids responsible for flower colours of Eustoma grandiflorum were characterized by TLC, HPLC, spectral and chemical analyses. Anthocyanins were delphinidin 3-rhamnosylgalactoside-5-glucoside and delphinidin 3-galactoside-5-glucoside, each acylated with p-coumaric acid, from the purple cultivar ‘Murasaki no Homare’ and the pelargonidin analogues, each acylated with either p-coumaric or ferulic acids, from the pink cultivar ‘Momo no Mine’. The major flavonol copigments were the 3-rhamnosylgalactoside-7-rhamnoside of myricetin, kaempferol and isorhamnetin and the 3-rhamnosylglucoside-7-rhamnoside of kaempferol and isorhamnetin. Flavonols present acylated with p-coumaric acid were myricetin 3-rhamnosylgalactoside-7-rhamnoside and robinin in both cis and trans forms, and isorhamnetin 3-rhamnosylgalactoside-7-rhamnoside. Robinin also was present acylated with caffeic or ferulic acids. Simulated in vitro colours obtained from the flavonoids present in this germplasm indicated that good blue colours were not attainable. Good blue colours were formed with delphinidin 3-p-coumaroyl-rhamnosylgalactoside-5-glucoside and C-glycosylflavone copigments such as swertisin and isoorientin. These copigments are readily available in other members of the Gentianaceae and this suggests the possibility of genetical engineering endeavours for increasing the colour range of this important new ornamental plant.  相似文献
