The cylindrical chaperonin GroEL of E. coli and its ring-shaped cofactor GroES cooperate in mediating the ATP-dependent folding of a wide range of polypeptides in vivo and in vitro. By binding to the ends of the GroEL cylinder, GroES displaces GroEL-bound polypeptide into an enclosed folding cage, thereby preventing protein aggregation during folding. The dynamic interaction of GroEL and GroES is regulated by the GroEL ATPase and involves the formation of asymmetrical GroEL:GroES1 and symmetrical GroEL: GroES2 complexes. The proposed role of the symmetrical complex as a catalytic intermediate of the chaperonin mechanism has been controversial. It has also been suggested that the formation of GroEL:GroES2 complexes allows the folding of two polypeptide molecules per GroEL reaction cycle, one in each ring of GroEL. By making use of a procedure to stabilize chaperonin complexes by rapid crosslinking for subsequent analysis by native PAGE, we have quantified the occurrence of GroEL:GroES1 and GroEL:GroES2 complexes in active refolding reactions under a variety of conditions using mitochondrial malate dehydrogenase (mMDH) as a substrate. Our results show that the symmetrical complexes are neither required for chaperonin function nor does their presence significantly increase the rate of mMDH refolding. In contrast, chaperonin-assisted folding is strictly dependent on the formation of asymmetrical GroEL:GroES1 complexes. Populations of scavenging seabird species in the North Sea may fluctuate with an artificial food source: the availability of fishery waste. To document this impact, it is necessary to assess the birds' nutritional status during periods with decreased fishing activity. Reference data for this purpose was collected from 22 herring gulls investigated during laboratory fasting. After 6 d of food deprivation and body mass losses exceeding 15%, the first birds entered starvation phase 3. Comparatively, this is a rather weak fasting capacity. Plasma levels of total protein and thyroid hormones decreased and beta-hydroxybutyrate increased with fasting duration. The leucocyte proportions were shifted from lymphocytes to heterophils. After 3 d of refeeding, most of the fasting changes were reversed. Plasma enzyme activities increased and hematocrit, hemoglobin, and erythrocyte numbers decreased in both fasting and control birds, most likely as a result of experimental stress and repeated blood sampling. Glucose, cholesterol, monocytes, basophils, and glycosylated hemoglobin remained fairly constant. Triglycerides, free fatty acids, uric acid, and urea varied significantly, but changes were not as clearly a result of fasting. Therefore, total protein, beta-hydroxybutyrate, triiodothyronine, thyroxine, and lymphocyte and heterophil percentages may be the most reliable indicators of the nutritional status and the condition of free-living herring gulls. 相似文献

Endoglin is a component of the transforming growth factor beta (TGF-beta) receptor complex, highly expressed by endothelial cells. Mutations in the endoglin gene are responsible for hereditary hemorrhagic telangiectasia type 1 (HHT1), an autosomal dominant vascular disorder caused by a haploinsufficiency mechanism. Vascular lesions (telangiectasia and arteriovenous malformations) in HHT1 are associated with loss of the capillary network, suggesting the involvement of endoglin in vascular repair processes. Using the chick chorioallantoic membrane (CAM) as an angiogenic model, we have analyzed the expression and function of chicken endoglin. A pan-specific polyclonal antibody (pAb) recognized chicken endoglin as demonstrated by immunostaining and Western blot analysis. In ovo treatment of chicken embryos with this pAb resulted in a significantly increased area of CAM. This effect was likely mediated by modulation of the ligand binding to endoglin as this pAb was able to inhibit TGF-beta1 binding. In a chromosome study on leucocyte cultures made in 13 patients treated with chlorpromazine, 15 treated with perphenazine, and nine treated with lysergide, a significantly higher frequency of gaps, breaks, and hypodiploid cells in the patients treated with perphenazine and lysergide occurred compared with the 41 controls studied. We investigate a non-linear network with two processing stages optimized to reduce the statistical dependencies in natural images. This network serves as a model for the neural information processing in the higher visual areas of primates (visual cortices V2-V4). The resulting population is analyzed with regard to non-linear selectivity and invariance properties. We find units that are very selective with respect to the space spanned by all possible input signals and units that are invariant with respect to certain stimulus classes. In comparison to the measured distribution of selectivity in V2 neurons, the selectivity histogram of the network units shows an even more pronounced tendency towards higher selectivities. A special property of the system is the emergence of non-linear interactions between coefficients from different scales and orientations, which are necessary for the exploitation of higher-order statistical redundancies of natural images. Forces acting on toroidal vortices in an unbounded medium (plasma vortices in air and vortex rings in air and water) are investigated. A solution to the equations describing such votrices is obtained. It is shown that this solution satisfactorily agrees with experiment. Based on the experimental results and the solution obtained, the drag coefficient C x of such vortices is found. For the same Reynolds numbers, the value of C x may be much less than the drag coefficient of a drop-shaped axisymmetric body (0.045), which is known to be the best streamlined object. 相似文献

The foraging behaviour (grazing time and diet composition), live-weight (LW) changes and parasitic infection (faecal egg counts of gastrointestinal nematodes) of 12 beef cows, 84 ewes and 84 goats suckling their offspring, managed in mixed grazing on heathlands with 24% of improved pasture of perennial ryegrass-white clover, were studied during the years 2003 and 2006. The results showed that goats tended to graze for a longer time, and utilised significantly more heathland vegetation than cattle and sheep, including in their diet in an average of 0.39 herbaceous plants, 0.11 gorse and 0.51 heather over the grazing season, comparing with respective mean values of 0.85, 0.02 and 0.13 in cattle and sheep. Dietary overlap was higher between cattle and sheep (0.76) than between sheep and goats (0.53) or between cattle and goats (0.47). Despite the high performance of autumn-calving cows in spring, sheep had the best LW changes per livestock unit (LU) during the whole grazing season. Goats' performance per LU was lower than in sheep but higher than in spring-calving cows. Regarding offspring production, lambs had higher LW gains per LU than calves and kids. Mean nematode eggs/g fresh faeces (epg) during the grazing season were higher in goats (91 epg) compared with sheep (34 epg) and cattle (14 epg), particularly from September to December. In conclusion, sheep performed best on these heathlands with improved pasture areas if the entire grazing season was considered, despite the good level of production in spring from autumn-calving cows. The results suggested the complementary use of goats in order to increase the effective utilisation of the available vegetation, achieving production levels similar to those from spring-calving cows. Therefore, mixed flocks of sheep and goats would be the most appropriate sustainable systems from the animal production and vegetation use points of view. 相似文献
