用含１．０ｍｍｏｌ／Ｌ ＳＡ的不同渗透势ＰＥＧ溶液漂浮处理小麦幼苗叶片,研究ＳＡ在水分逆境下脂质过氧化中的作用。结果表明,ＳＡ降低了叶片ＣＡＴ活性,轻度渗透胁迫下ＳＡ对稳定膜结构和功能有一定作用。较严重的渗透胁迫加ＳＡ处理使叶片失水量、膜相对透性和ＭＤＡ含量有所增加,Ｈ２Ｏ２和Ｏ２＾－积累较快,但与不加ＳＡ处理比较,ＳＯＤ和ＰＰＯＤ活性仍较高,脂质过氧化程度稍有加重。ＳＡ在诱导植物提高抗逆力中的作     

The newly synthesized plant growth regulator <Emphasis Type="Italic">S</Emphasis>-methylmethionine salicylate may provide protection against high salinity in wheat

High salinity is one of the major environmental factors limiting the productivity of crop species worldwide. Improving the stress tolerance of cultivated plants and thus increasing crop yields in an environmentally friendly way is a crucial task in agriculture. In the present work the ability of a new derivative, S-methylmethionine-salicylate (MMS), to improve the salt tolerance of wheat plants was tested parallel with its related compounds salicylic acid and S-methylmethionine. The results show that while these compounds are harmful at relatively high concentration (0.5 mM), they may provide protection against high salinity at lower (0.1 mM) concentration. This was confirmed by gas exchange, chlorophyll content and chlorophyll-a fluorescence induction measurements. While osmotic adjustment probably plays a critical role in the improved salt tolerance, neither Na or K transport from the roots to the shoots nor proline synthesis are the main factors in the tolerance induced by the compounds tested. MMS, S-methylmethionine and Na-salicylate had different effects on flavonol biosynthesis. It was also shown that salt treatment had a substantial influence on the SA metabolism in wheat roots and leaves. Present results suggest that the investigated compounds can be used to improve salt tolerance in plants.     

Application of an angiosperm-based classification system (BiPo) to Mediterranean coastal waters: using spatial analysis and data on metal contamination of plants in identifying sources of pressure

The Biotic Index based on Posidonia oceanica (BiPo) is a classification system for evaluation of the ecological status in Mediterranean coastal waters, developed in accordance with the EU Water Framework requirements. The aim of this study is to verify the applicability and reliability of the BiPo index to different geographical areas of the north-western Mediterranean (France, Spain and Italy), to understand whether such a classification system may be applied more extensively, as so far it has only been applied to coastal waters in Corsica. The ecological status determined for sites is verified against pressures revealed from satellite imagery and from trace metal contamination of plants, to identify the sources of pressure that may be responsible for a low ecological status. The results of this study indicate that: (i) the BiPo index responds reliably to pressures, in different areas of the Mediterranean; (ii) sites with an ecological quality ratio (EQR) close to the good/moderate boundary require particular attention to identify and reduce causes of deterioration; (iii) the support of chemical indicators, in this case metal contamination, is relevant to identify potential sources of pressure.     

Mortality of lemmings, Lemmus lemmus, at peak density in a mountainous area of Norway

We estimated carcass density and determined cause of death in a peak-density lemming, Lemmus lemmus , population at Finse, Norway. Of 74 recovered carcasses, 80% had subcutaneous haematoma (blood underneath the skin) in the head region, though the skin on the skull most often appeared unharmed. Thirty percent were apparently killed by a mustelid predator, while 51% were probably killed by corvids. Carcass density was 26.6 and 9.4ha^-1 in good and poor lemming habitats, respectively.     

Deconvolution can be used in electrooptic studies to correct for non-ideal electric excitation pulses only when the electric dipole moment of the studied molecules is predominantly induced

The electric field pulses used for most measurements of transient electrooptic properties such as birefringence and dichroism, are rectangular and assumed to be ideal, but in practice do all such pulses have non-zero rise and fall times. Claims have been made that this non-ideality may be taken into account by employing standard deconvolution techniques. We find that this approach yields exact results in the zero electric field limit when the electric dipole moment of the studied macromolecules is predominantly induced. However, for finite electric field strengths and/or macromolecules with partly or fully permanent electric dipole moments, we find that the deconvolution method yields erroneous estimates of the electrooptic relaxation times. When the decay time of the electric pulse and the electrooptic decay time are equal, and the system is operated in the Kerr domain, this systematic error for macromolecules with purely permanent electric dipole moment equals 37%. In a research field where the uncertainty of the reported relaxation times normally is assumed to be only a few percent this is an error that may seriously mislead unsuspecting users. We find that this systematic error can readily be avoided by employing standard numerical integration of a set of coupled first-order differential equations instead of the standard deconvolution techniques.     

Mycobacterium tuberculosis Multidrug Resistant Strain M Induces an Altered Activation of Cytotoxic CD8+ T Cells

In human tuberculosis (TB), CD8⁺ T cells contribute to host defense by the release of Th1 cytokines and the direct killing of Mycobacterium tuberculosis (Mtb)-infected macrophages via granule exocytosis pathway or the engagement of receptors on target cells. Previously we demonstrated that strain M, the most prevalent multidrug-resistant (MDR) Mtb strain in Argentine, is a weak inducer of IFN-γ and elicits a remarkably low CD8-dependent cytotoxic T cell activity (CTL). In contrast, the closely related strain 410, which caused a unique case of MDR-TB, elicits a CTL response similar to H37Rv. In this work we extend our previous study investigating some parameters that can account for this discrepancy. We evaluated the expressions of the lytic molecules perforin, granzyme B and granulysin and the chemokine CCL5 in CD8⁺ T cells as well as activation markers CD69 and CD25 and IL-2 expression in CD4⁺ and CD8⁺ T cells stimulated with strains H37Rv, M and 410. Our results demonstrate that M-stimulated CD8⁺ T cells from purified protein derivative positive healthy donors show low intracellular expression of perforin, granzyme B, granulysin and CCL5 together with an impaired ability to form conjugates with autologous M-pulsed macrophages. Besides, M induces low CD69 and IL-2 expression in CD4⁺ and CD8⁺ T cells, being CD69 and IL-2 expression closely associated. Furthermore, IL-2 addition enhanced perforin and granulysin expression as well as the degranulation marker CD107 in M-stimulated CD8⁺ T cells, making no differences with cells stimulated with strains H37Rv or 410. Thus, our results highlight the role of IL-2 in M-induced CTL activity that drives the proper activation of CD8⁺ T cells as well as CD4⁺ T cells collaboration.     

Cryopreservation of Sheep Red Blood Cells

The protection of sheep erythrocytes at freezing temperatures was investigated using glycerol, dimethylsulfoxide (DMSO), glucose and four different types of polyvinylpyrrolidone (PVP) as cryoprotective agents. Depending on type (molecular weight) and concentration good protection was obtained with PVP, whereas glycerol, DMSO and glucose were unsatisfactory. Recovery of cells after thawing was most successful when the cells had been frozen at a concentration of 1–2 × 109 cells/ml. No cells tolerated freezing at −20 °G. Best results were obtained when the cells were frozen directly in liquid nitrogen (−196°G).
