Blind goldfish were subjected to linear accelerations on a motor car and on a parallel swing. Moyements of the fish in a tank during the accelerations were recorded with a movie camera. During the horizontal acceleration, the fish aligns his longitudinal axis in a plane perpendicular to the direction of an apparent gravity with the fish's back pointing away from the direction of this apparent gravity vector. This is similar to the manner in which the fish usually aligns himself horizontally in response to the vertically downward terrestrial gravity and can therefore be termed gravity reference response. It is concluded that blind goldfish cannot distinguish between otolith displacements caused by passive tilts and equivalent otolith displacements caused by moderate inertial forces during rectilinear acceleration. With a horizontal jerk of higher magnitude, two additional responses can occur: horizontal 180° turns following tailward jerks and straight forward darting following noseward jerks.This work was supported by NASA Grant No. NGR 23-005-201.  相似文献

A flow-through system for rapid automated analysis of intracellular fluorescence parameters is described. In combination with commercially available instrumentation for fluorescence microscopy, fluorometry and pulse height analysis, the hydrodynamic focusing flow-through principle can be successfully applied. 2′-Deoxyadenosine and 3′-deoxyadenosine (cordycepin) can be incorporated into the 3′-terminal position of tRNAPhe by tRNA nucleotidyl transferase. tRNAPhe-C-C-2′dA and tRNAPhe-C-C-3′dA, missing the cis-diol group at the 3′-terminal end are resistant to periodate oxidation and are not able to form borate complexes. In aminoacylation experiments only the tRNAPhe-C-C-3′dA proved to be chargeable.  相似文献

The increase in the incorporation of amino acids into protein in vitro by preparations obtained from protein-fed rats as compared with preparations obtained from carbohydrate-fed rats has been described previously. After molecular sieving through Sephadex G-25 of cell-free preparations, the difference in incorporating activity between the two types of rats was diminished in systems containing ATP, phosphoenolpyruvate, pyruvate kinase, GTP, and a mixture of amino acids. When, after molecular sieving, a mitochondrial (15,000 g) supernatant was incubated for 4 min at 35°C the polysomal pattern of the preparations was unchanged. In the presence of ATP, phosphoenolpyruvate, and pyruvate kinase the polysomal incorporating activity was low and the polysomal pattern was only slightly changed. Addition of GTP increased the activity markedly, and a more pronounced activity was observed when a mixture of amino acids was added as well. As the amino acid incorporation ability increased, monosomes were formed from the polyribosomes. The activity of the polyribosomes was severalfold higher than that of non-Sephadex-treated preparations, indicating an activation of polysomal aggregates which under the usually applied conditions of incubation and prior to molecular sieving show little or insignificant activity. It was possible to activate polyribosomes from carbohydrate-fed and protein-fed rats to almost the same extent.  相似文献

Zusammenfassung Beidseitige Adrenalektomie und Hypophysektomie führen bei der Ratte zu gleichartigen histologischen Veränderungen in der Zona externa infundibuli. In beiden Fällen treten in der normalerweise weitgehend goniorinegativen Zona externa infundibuli große Mengen gomoripositiver Granula auf. Sie scheinen Fasern anzugehören, die senkrecht zur Längsachse des Infundibulum verlaufen.Die Befunde werden als weiterer Hinweis dafür betrachtet, daß die gomoripositiven Substanzen der Zona externa infundibuli eine Bedeutung für die Steuerung der Nebennierenrindenfunktion haben.
Studies on the functional morphology of the neurohypophysisII. Comparison of histological changes in the median eminence of the rat after bilateral adrenalectomy and hypophysectomy
Summary Bilateral adrenalectomy and hypophysectomy in the rat produce similar histological changes in the outer layer of the median eminence. In both cases, abundant gomoripositive granules are observed in the outer layer, which normally reacts gomorinegative. The gomoripositive granules seem to belong to fibres running vertically through the infundibulum.These findings are regarded as a further indication, that the gomoripositive substances of the outer layer of the median eminence play a significant role in controlling adrenocortical function.

Summary A method for the estimation of the yield on energy (Y ATP) and of the efficiency of oxidative phosphorylation, in vivo (P/O ratio) is described, which is based on the measurement of effective gas exchange values ( and ) and of the yield coefficient Y of continuously growing populations of baker's yeast which vary in the degree of fermentation and respiration. For Y ATP a value of 12.0±0.5 and for P/O ratio one of 1.1±0.05 was found and seems to be independent of the type of glucose catabolism (under glucose limitation).The gas exchange of populations of Saccharomyces cerevisiae synchronized at different growth rates was determined. The specific oxygen uptake and carbon dioxide formation rate, Q O 2, and Q CO 2, are shown to depend on the state of the cells in the budding cycle. Increase in gas metabolism and therefore increased energy generation coincides with the initiation of budding. The longer the generation time g the more expressed are these oscillations of energy formation over the budding cycle. The relationship between the course of energy generation and energy storage and the sequence of budding and single cell phase over the division cycle is discussed.  相似文献

Zusammenfassung Das Axialorgan vonAsterias rubens und sein Terminalfortsatz im Dorsalsack wurden licht- und elektronenmikroskopisch untersucht.Die Hämalkanäle des Axialorgans bilden ein labyrinthartiges Konvolut, das in den Axialsinus hineinragt. Die Wandung der Kanäle besteht aus einer Basalmembran, deren Außenfläche von Coelomzellen (Deckzellen, Epizyten) und Muskelzellen überzogen wird. Beide Zellarten enthalten zahlreiche Einschlüsse verschiedener Form und Struktur. Die Deckzellen breiten sich mit vielen zarten Fortsätzen auf der Oberfläche des Organs zu einem zytoplasmatischen Gitterwerk aus, das morphologisch an den Zellüberzug der Glomeruluskapillaren der Vertebratenniere erinnert. Sowohl die Deckzellen als auch die von ihnen teilweise umschlossenen Muskelzellen besitzen Geißeln, die sich in das Lumen des Axialsinus erheben. Auch die Muskelzellen sind mit Ausläufern versehen. Ein Teil dieser Fortsätze enthält Bündel von dicken und dünnen Myofilamenten. Wahrscheinlich handelt es sich um Muskelzellen vom Paramyosintypus.Die Basalmembran wird hie und da durch lockere Bündel von Kollagenfasern mit periodischer Gliederung ihrer Fibrillen bzw. Filamente verstärkt. Diese Bündel sind in die aufgelockerte Innenschicht der Basalmembran eingelagert.Die Hämalkanäle werden nicht von einer endothelartigen Zellschicht ausgekleidet, verhalten sich also ebenso wie die Blutgefäße der Anneliden. Der Innenfläche der Basalmembran schmiegen sich gelegentlich große freie, an Einschlüssen reiche Zellen vom Typ der Coelomozyten an bzw. diese Zellen lösen sich von ihr ab. Das Lumen der Hämalkanäle ist vielfach von freien Zellen ausgefüllt.Die Wandung der Gefäße des Terminalfortsatzes besitzt grundsätzlich den gleichen Aufbau wie die der Hämalkanäle im Axialorgan. Im Gegensatz zu diesen enthält sie jedoch viele nackte Axone, die unter und zwischen den Deckzellen und Muskelzellen verlaufen und mit letzteren engen Kontakt aufnehmen. Regelrechte Synapsen wurden jedoch nicht festgestellt. In den Axonen und ihren Endigungen finden sich Vesikel und massendichte Granula, unter ihnen solche, die von einer Membran umschlossen werden und damit an neurosekretorische Elementargranula erinnern.Die mitgeteilten Befunde stehen mit älteren Lebendbeobachtungen in Einklang, wonach das Axialorgan und sein Terminalfortsatz pulsatorische Bewegungen nach Art eines Herzens ausführten. Die strukturelleÄhnlichkeit des Axialorgans, in demRemane (1959) ein Homologen des Glomerulus von Hemichordaten erblickt, mit den Glomeruli der Vertebratenniere läßt darüber hinaus an eine exkretorische Funktion denken. Auf eine vergleichbare Analogie zwischen der sog. Endblase im Maxillarorgan von Crustaceen und den Nierenglomeruli der Wirbeltiere (Tyson, 1968) wird hingewiesen.
The axial organ of the starfish, Asterias rubens L.
Summary The axial organ and its terminal process (head piece) in the dorsal sac ofAsterias rubens were investigated with the light-and the electronmicroscope.The hemal channels of the axial organ form a labyrinthine system (spongy body) which is situated within the axial sinus. The walls of the channels consist of a basement membrane, the outer surface of which is covered by coelomic epithelial elements (epicytes) and muscle cells. Either cell type contains numerous inclusions of different shape and structure (phagocytosis, lysosomes). The epicytes spread with many delicate processes on the surface of the organ, thus moulding a cytoplasmic meshwork which is similar to the visceral layer of the glomerular capillaries of the vertebrate kidney. The muscle cells which are also provided with cytoplasmic processes, part of which contains bundles of thick and thin filaments (presumably constituting fibres of the paramyosin-type), are to some extent enclosed by the covering cells. Eithercell type bears cilia projecting into the lumen of the axial sinus.The basement membrane contains occasionally bundles of collagen fibres showing a periodical structure of their individual fibrils. These bundles are embedded in the loose interior layer of the basement membrane.The hemal channels are not lined by an endothelial cellular layer, resembling in this respect the vessels of the annelides, Occasionally, big free coelomocytes rich of inclusion bodies, cling to the inner surface of the basement membrane or become detached of it. The lumen of the hemal channels is frequently crowded with free cells.The structure of the vascular wall in the terminal process is principally the same as in the axial organ proper. In contrast to this it contains, however, many naked axons which run under or between the epicytes and muscle cells and which come into close contact with the latter. Typical synapses, however, have not been detected. Inside the axons and their terminals, vesicles and electron dense granules are to be found. The dense granules are often surrounded by a distinct membrane, thus bearing a close similarity to neurosecretory elementary granules.Our findings are in accordance with older in vivo observations after which the axial organ and its terminal process undergo contractions comparable to the pulsations of a heart. In addition to this, the structural similarity of the axial organ with the glomeruli of the vertebrate kidney suggests an excretory function. As already pointed out byRemane (1959) the axial organ of the Echinoderms might be the homologue to the glomerulus of Hemichordata. A comparable analogy between the terminal sac in the maxillary gland of the crustacea and the glomeruli of the vertebrate kidney (Tyson, 1968) is referred to.
