Cysts and cyst stages (merozoites) of Sarcocystis ovifelis were studied using methods of light microscope cytochemistry for detecting nucleic acids, proteins, polysaccharids, lipids and phosphatases, the latter substances being examined with negative results. DNA has been localized in the zoites whereas high contents of RNA and amylopectin were seen in cytoplasm. Tannofilic protein was detected evenly distributed both in the nucleus and cytoplasm. 相似文献

Development of immunohistochemical characteristics of intrafusal fibres in normal and de-efferented rat muscle spindles 总被引:1,自引:0,他引:1
Summary Intrafusal muscle fibres in adult muscle spindles differ in their myosin composition. After selective motor denervation intrafusal muscle fibres develop mature ultrastructural characteristics. In order to evaluate the role of fusimotor innervation on the maturation of the myosin composition of intrafusal muscle fibres we have examined with immunohistochemical techniques i) the postnatal development of muscle spindles in new-born rats and in 7–21 day old rats; ii) muscle spindles in the EDL of 21-day-old rats de-efferented at birth. For the characterization of myosins in intrafusal fibres we used three myosin antisera: antipectoral myosin, antiheart myosin and antiheart myosin adsorbed with muscle powder from the soleus muscle of guinea pig. We show in this study that during development intrafusal fibres change immunoreactivity and that in the absence of motor innervation bag fibres do not fully develop the myosin characteristics of control spindles. We conclude that the maturation of bag1 and bag2 fibres apparently requires next to the inductive influence of sensory axon terminals the presence and activity of fusimotor axons. 相似文献

The effect of platelet tropomyosin on the ATPase activity of a muscle actin-myosin subfragment 1 system has been examined in 30 mM KCl, 5 mM MgCl2, 2 mM ATP, 0.1 mM EGTA, 2 mM Tris, pH 7.8. Whereas muscle tropomyosin inhibits the activity by 60%, the platelet protein had no effect. Addition of muscle troponin in the absence of Ca2+ to the system inhibited the activity by up to 80% irrespective of whether muscle or platelet tropomyosin was used. The release of this inhibition by the addition of Ca2+ was much less in the case of platelet tropomyosin. This may result from the fact that platelet tropomyosin aggregates poorly in a head-to-tail manner and interacts only weakly with muscle troponin-T. In the presence of troponin-I and platelet tropomyosin, inhibition of the ATPase activity was 80%. This inhibition was largely released by the addition of troponin-C irrespective of the presence of Ca2+. The addition of brain calmodulin, however, released the inhibition in the presence of calcium but not in its absence. These effects can be correlated with the binding or lack of binding of the platelet tropomyosin to the actin filament. 相似文献

The surface properties of spinach chloroplasts, both of intact chloroplasts with surrounding envelope and broken chloroplasts consisting of the inner lamellar system, have been studied by partitioning them between two aqueous phases, especially using counter-current distribution technique. The two-phase system consists of poly(ethyleneglycol), dextran and water. The two polymers are enriched in opposite phases and by binding deoxycholate or palmitate to one of the polymers the affinity of chloroplasts for the corresponding phase is strongly enhanced. The partition of the two classes of chloroplasts, however, is not affected to the same degree and the affinity of the chloroplast envelope for deoxycholate and palmitate is stronger than that of the lamellar system. This has been correlated to the chemical composition of the two types of membranes. By studying the effect of salts on the partition it has been found that the lamellar system bears a larger number of negative charges as compared to the envelope of the intact chloroplast. 相似文献

Attempts were undertaken to study the character of the distribution of I. ricinus over a large territory. Methodic principles of the land survey of arthropods distribution and laboratory processing of its results, that were used before for I. persulcatus, were found to be quite suitable for I. ricinus. It was established that hungry adults of I. ricinus have at least three types of the distribution throughout the forests of Lithuania. The effects of eleven quassinoids on Rous sarcoma virus induced cell transformation and on growth of normal cells were examined. At concentrations of 0.15-1 μg/ml they inhibited foci formation (76–99 %) without toxic effects on normal cells. The most active compounds also affected virus production by transformed cells. In intact normal and transformed cells, protein and DNA synthesis was equally affected after 3 hours of exposure to quassinoids of both cell types. RNA synthesis was not inhibited. This study has shown that the structural requirement of a C-15 ester in the quassinoids for antileukemic activity and is not essential for their antitransforming activity. 相似文献

The use of Process Analytical Technology tools coupled with chemometrics has been shown great potential for better understanding and control of mammalian cell cultivations through real-time process monitoring. In-line Raman spectroscopy was utilized to determine the glucose concentration of the complex bioreactor culture medium ensuring real-time information for our process control system. This work demonstrates a simple and fast method to achieve a robust partial least squares calibration model under laboratory conditions in an early phase of the development utilizing shake flask and bioreactor cultures. Two types of dynamic feeding strategies were accomplished where the multi-component feed medium additions were controlled manually and automatically based on the Raman monitored glucose concentration. The impact of these dynamic feedings was also investigated and compared to the traditional bolus feeding strategy on cellular metabolism, cell growth, productivity, and binding activity of the antibody product. Both manual and automated dynamic feeding strategies were successfully applied to maintain the glucose concentration within a narrower and lower concentration range. Thus, besides glucose, the glutamate was also limited at low level leading to reduced production of inhibitory metabolites, such as lactate and ammonia. Consequently, these feeding control strategies enabled to provide beneficial cultivation environment for the cells. In both experiments, higher cell growth and prolonged viable cell cultivation were achieved which in turn led to increased antibody product concentration compared to the reference bolus feeding cultivation. 相似文献
