Background

E. coli belonging to the phylogenetic group B2 are linked to Inflammatory Bowel Disease (IBD). Studies have shown that antimicrobials have some effect in the treatment of IBD, and it has been demonstrated that E. coli Nissle has prophylactic abilities comparable to 5-aminosalicylic acid (5-ASA) therapy in ulcerative colitis. The objective of this study was to test if ciprofloxacin and/or E. coli Nissle could eradicate IBD associated E. coli in the streptomycin-treated mouse intestine.

Results

After successful colonization with the IBD associated E. coli strains in mice the introduction of E. coli Nissle did not result in eradication of either IBD associated strains or an E. coli from a healthy control, instead, co-colonization at high levels were obtained. Treatment of mice, precolonized with IBD associated E. coli, with ciprofloxacin for three days alone apparently resulted in effective eradication of tested E. coli. However, treatment of precolonized mice with a combination of ciprofloxacin for 3 days followed by E. coli Nissle surprisingly allowed one IBD associated E. coli to re-colonize the mouse intestine, but at a level 3 logs under E. coli Nissle. A prolonged treatment with ciprofloxacin for 7 days did not change this outcome.

Conclusions

In the mouse model E. coli Nissle can not be used alone to eradicate IBD associated E. coli; rather, 3 days of ciprofloxacin are apparently efficient in eradicating these strains, but surprisingly, after ciprofloxacin treatment (3 or 7 days), the introduction of E. coli Nissle may support re-colonization with IBD associated E. coli.  相似文献

Inbreeding depression and its consequences for mate choice have been extensively studied in free-living animals. However, very little is known about its significance for parasites, although it is well recognized that the mating systems of parasites can have important implications for their epidemiology and evolution. In this article, we show that the cestode Schistocephalus solidus shows incestuous mate preference despite evidence for very strong inbreeding depression. When given the simultaneous choice between mating with a sibling and an unrelated partner, on average, the cestode preferred its sibling. To explain this surprising result, we present three hypotheses that suggest different benefits to fitness of incestuous mating, which could, alone or in concert, outweigh the cost of inbreeding depression.  相似文献

Blue mussels (Mytilus edulis L.) are a valuable resource for commercial shellfish production and may also have uses as a tool in habitat improvement, because mussel beds can increase habitat diversity and complexity. A prerequisite for both commercial mussel production and habitat improvement is the availability of seed mussels collected with minimum impact on the benthic ecosystem. To examine whether mussels collected in suspended cultures can be used for bottom culture production and as tool in habitat improvement, the differences in predatory defence responses between suspended and bottom mussels exposed to the predatory shore crab (Carcinus maenas L.) were tested in laboratory experiments and in the field. Predatory defence responses (byssal attachment and aggregation) and morphological traits were tested in laboratory, while growth and mortality were examined in field experiments. Suspended mussels had an active response in relation to the predator by developing a significantly firmer attachment to the substrate and a closer aggregated structure. Bottom mussels had a passive strategy by having a thicker shell and larger relative size of the adductor muscle. In a field experiment mussels originated from suspended cultures had a higher length increment and lower mortality when compared to bottom mussels. It is concluded that suspended mussels potentially are an alternative resource to bottom culture and can be used in habitat improvement of mussel beds, but that the use of suspended mussels has to be tested further in large-scale field experiments.  相似文献

Abstract A hermaphroditic individual that prefers to outbreed but that has the potential of selfing faces a dilemma: in the absence of a partner, should it wait for one to arrive or should it produce offspring by selfing? Recent theory on this question suggests that the evolutionary solution is to find an optimal delay of reproduction that balances the potential benefit of outcrossing and the cost of delaying the onset of reproduction. Assuming that resources retained from breeding can be reallocated to future reproduction, isolated individuals, compared with individuals with available mates, are predicted to delay their age at first reproduction to wait for future outcrossing. Here, I present empirical support for this idea with experimental data from the hermaphroditic cestode Schistocephalus solidus. I show that individuals breeding alone delay their reproduction and initially produce their eggs at a slow rate relative to cestodes breeding in pairs. This delay is partly compensated for by a later higher egg production, although singly breeding cestodes still pay a cost of overall lower egg production.  相似文献

Seedlings of barley (Hordeum vulgare L. cvs Salka and Zita), buckwheat (Fagopyrum esculentum Moench) and rape (Brassica napus L. ssp. napus cv. Line) were grown in complete nutrient solutions with 8 or 10 different P concentrations in the range of 0–2 mM. Phosphate export from roots to shoots was determined from the amount of 32P (or 33P) absorbed and exported to shoots in 1 h from a nutrient solution containing 0.1 mM radiolabelled phosphate. P export was also determined in the presence of a metabolic uncoupler (DNP, 2.4-dinitrophenol) and a protein synthesis inhibitor (CH, cycloheximide). Phosphorus export from roots to shoots reached a maximum at a certain optimum level of phosphorus in shoots and roots, and decreased at both higher and lower P levels. Maxinmm P export was 1.7 ± 0.2 and 4.5 ± 0.5 (mean ±se of the three species) times higher than the P export at the lowest and highest [P]root, respectively. Hill plots as well as plots of the untransformed decreasing P export vs root or shoot P concentrations above the optimum were linear and had high correlation coefficients. The Hill coefficient (nH) based on [P]root, was —7.7 for barley cv. Salka and varied between -3.8 and -4.5 for the other species. Based on [P]shootot nH was—16.1 for barley cv. Salka, -3.7 for barley cv. Zita and -6.4 for the two dicotyledonous species. Relative to the amount of P simultaneously absorbed by the root system, the import of P per unit shoot weight decreased linearly over the whole range of shoot P concentrations in the dicotyledonous species. In contrast, the relative import of P per unit shoot weight of the two barley cultivars increased at low levels of [P]shoot and decreased at higher levels. DNP and CH almost eliminated P export from roots to shoots of seedlings with low or high P status. In seedlings with medium P status only 60 to 75% of the P export was affected.  相似文献

We demonstrate here a rapid and cost-effective technique for nanoscale patterning of functional molecules on the surface of a DNA origami. The pattern is created enzymatically by transferring a functionalized dideoxynucleotide to the 3'-end of an arbitrary selected set of synthetic DNA oligonucleotides positioned approximately 6 nm apart in a 70 × 100 nm(2) rectangular DNA origami. The modifications, which are performed in a single-tube reaction, provide an origami surface modified with a variety of functional groups including chemical handles, fluorescent dyes, or ligands for subsequent binding of proteins. Efficient labeling and patterning was demonstrated by gel electrophoresis shift assays, reverse-phase HPLC, mass spectrometry, atomic force microscopy (AFM) analysis, and fluorescence measurements. The results show a very high yield of oligonucleotide labeling and incorporation in the DNA origami. This method expands the toolbox for constructing several different modified DNA origami from the same set of staple strands.  相似文献

Alternative splicing enhances proteome diversity and modulates cancer-associated proteins. To identify tissue- and tumor-specific alternative splicing, we used the GeneChip Human Exon 1.0 ST Array to measure whole-genome exon expression in 102 normal and cancer tissue samples of different stages from colon, urinary bladder, and prostate. We identified 2069 candidate alternative splicing events between normal tissue samples from colon, bladder, and prostate and selected 15 splicing events for RT-PCR validation, 10 of which were successfully validated by RT-PCR and sequencing. Furthermore 23, 19, and 18 candidate tumor-specific splicing alterations in colon, bladder, and prostate, respectively, were selected for RT-PCR validation on an independent set of 81 normal and tumor tissue samples. In total, seven genes with tumor-specific splice variants were identified (ACTN1, CALD1, COL6A3, LRRFIP2, PIK4CB, TPM1, and VCL). The validated tumor-specific splicing alterations were highly consistent, enabling clear separation of normal and cancer samples and in some cases even of different tumor stages. A subset of the tumor-specific splicing alterations (ACTN1, CALD1, and VCL) was found in all three organs and may represent general cancer-related splicing events. In silico protein predictions suggest that the identified cancer-specific splice variants encode proteins with potentially altered functions, indicating that they may be involved in pathogenesis and hence represent novel therapeutic targets. In conclusion, we identified and validated alternative splicing between normal tissue samples from colon, bladder, and prostate in addition to cancer-specific splicing events in colon, bladder, and prostate cancer that may have diagnostic and prognostic implications.  相似文献

We tested the hypothesis that the cestode Schistocephalus solidus is capable of premature gamete exchange as a plerocercoid in the last intermediate stickleback host. The existence of such a reproductive mode is suggested by the highly advanced gonadal development in the plerocercoid and the large fitness gain of outcrossing. In addition, eggs from selfing cestodes have a higher hatching rate when the cestode originated from a doubly infected stickleback than when it came from a singly infected fish. We hatched eggs from 10 singly breeding cestodes that originated from doubly infected sticklebacks with the prediction that some should be outcrossed and share alleles with both the breeding cestode and the second cestode in the coinfection if the hypothesis is correct. However, all of the 430 tested larvae matched only the alleles of the breeding cestode. It is therefore very unlikely that S. solidus engaged in gamete exchange in its fish host. We suggest an alternative hypothesis to explain the higher hatching rate of eggs produced by cestodes from doubly infected fish as compared to those from single infections.  相似文献

According to current evolutionary dogma, multiple infections generally increase a parasite's virulence (i.e. reduce the host's reproductive success). The basic idea is that the competitive interactions among strains of parasites developing within a single host select individual parasites to exploit their host more rapidly than their competitors (thereby causing an increase in virulence) to ensure their transmission. Although experimental evidence is scarce, it often contradicts the theoretical expectation by suggesting that multiple infections lead to decreased virulence. Here, we present a theoretical model to explain this contradiction and show that the evolutionary outcome of multiple infections depends on the characteristics of the interaction between the host and its parasite. If we assume, as current models do, that parasites have only lethal effects on their host, multiple infections indeed increase virulence. By contrast, if parasites have sub-lethal effects on their host (such as reduced growth) and, in particular, if these effects feed back onto the parasites to reduce their rate of development, then multiplicity of infection generally leads to lower virulence.  相似文献
