A new disease resistance locus in Arabidopsis, RPS3 , was identified using a previously cloned avirulence gene from a non- Arabidopsis pathogen. The avrB avirulence gene from the soybean pathogen Pseudomonas syringae pv. glycinea was transferred into a P. syringae pv. tomato strain that is virulent on Arabidopsis , and conversion to avirulence was assayed on Arabidopsis plants. The avrB gene had avirulence activity on most, but not all, Arabidopsis ecotypes. Of 53 ecotypes examined, 45 were resistant to a P. syringae pv. tomato strain carrying avrB , and eight were susceptible. The inheritance of this resistance was examined using crosses between the resistant ecotype Col-0 and the susceptible ecotype Bla-2. In F2 plants from this cross, the ratio of resistant:susceptible plants was approximately 3:1, indicating that resistance to P. syringae expressing avrB is determined by a single dominant locus in ecotype Col-0, which we have designated RPS3 . Using RFLP analysis, RPS3 was mapped to chromosome 3, adjacent to markers M583 and G4523, and ≤ 1 cM from another disease resistance locus, RPM1 . In soybean, resistance to P. syringae strains that carry avrB is controlled by the locus RPG1 . Thus, RPG1 and RPS3 both confer avrB -specific disease resistance, suggesting that these genes may be homologs. 相似文献

To test the possible interaction of polyamines in plant growth responses, transgenic tobacco plants containing the Avena sativa L. (oat) arginine decarboxylase (ADC) gene under the control of a tetracycline-inducible promoter were generated. Inducible overexpression of oat ADC in transgenic tobacco led to an accumulation of ADC mRNA, increased ADC activity and changes in polyamine levels. Transgenic lines, induced during vegetative stage, displayed different degrees of an altered phenotype, the severity of which was correlated with putrescine content. These phenotypic changes were characterized by short internodes, thin stems and leaves, leaf chlorosis and necrosis, as well as reduced root growth. This is the first report to show altered phenotypes as a consequence of polyamine changes under tetracycline-induction in in vivo conditions. Interestingly, overexpression of oat ADC in tobacco resulted in similar detrimental effects to those observed by ADC activation induced by osmotic stress in the homologous oat leaf system. In the context of the role of specific polyamines in plant growth and development, the present results indicate that activation of the ADC pathway leading to high levels of endogenous putrescine (or its catabolytes) is toxic for the vegetative growth of the plant. In contrast, no visible phenotypic effects were observed in flowering plants following tetracycline induction. Further characterization of the different transgenic lines may shed light on the action of specific polyamines in different plant developmental processes. 相似文献

Dung‐heap counts were used to estimate density of grey duiker (Sylvicapra grimmia Linnaeus 1758) in the Matobo National Park, Zimbabwe. To test assumptions of this method, defecation rate and defecation site selection were investigated under captive and field conditions, and densities were compared with independent estimates derived from territory mapping. Many assumptions were violated: males defecated more frequently than females with mean dry mass per deposit greater in females, but total daily faecal production was similar between sexes. Spatial distribution of faeces was clumped, and 52.8% of locations contained multiple deposits. Duikers exhibited habitat type preferences (i.e. low‐ to medium‐density woodland) with herbaceous layer heights 40–100 cm and visibility >20 m. Calculated grey duiker density from dung‐heap counts in cleared plots was 9.7 ± 1.3 animals km−2, approximately double the territory‐mapping estimate based on Minimum Convex Polygons (5.13 animals km−2) but similar to the 75% Fixed Kernel estimate (10.95 animals km−2). Provided that sex ratios approach parity and sampled area is representative of all utilized habitats, violation of basic assumptions of the dung‐heap count method has a minor effect on density estimate accuracy. 相似文献

1. We studied seasonal changes in the carbon and nitrogen stable isotope ratios of larval Chironomus anthracinus and C. plumosus from the profundal sediments of four contrasting lakes. 2. Pronounced seasonal changes in both δ13C and δ15N values were evident in chironomid larvae of both species from two summer‐stratified, eutrophic lakes: Esthwaite Water and Wyresdale Park. Changes were most marked in the larvae of C. plumosus and in larvae from greater depths. In contrast, neither C. anthracinus in summer‐stratified but mesotrophic Schöhsee, nor C. plumosus in polymictic Großer Binnensee, showed marked seasonality in larval stable isotope ratios. 3. The particularly strong 13C‐depletion of larvae from the stratified, eutrophic lakes is attributed to a significant contribution of methane‐derived carbon to their diets. Feeding by larvae on isotopically light methanotrophic bacteria appears to occur mainly when autumn overturn of the water column restores oxygenated conditions to the sediment surface. At this time both δ13C and δ15N values of larvae decreased sharply. 4. Changes in the mean stable isotope ratio of the larval populations can also occur when larger, more isotopically light, larvae pupate and emigrate from the population to hatch as imagos. This effect can induce seasonal changes in larval isotope values even in lakes in which there is no evidence of a significant involvement of methane‐derived carbon in their diets. Variations in emergence patterns between species and between lakes may generate differences in the seasonal pattern of change in stable isotope ratios in larval populations. 5. Our results emphasise the importance of adequate seasonal sampling if stable isotope ratios are to be used as biomarkers to study the role of key groups, such as chironomid larvae, in the trophic structure of lakes. 相似文献

Aim To explore rates of rain forest expansion and associated ecological correlates in Eucalyptus‐dominated woodland savanna vegetation in north‐eastern Australia, over the period 1943–91. Location Iron Range National Park and environs, north‐east Queensland, Australia. This remote region supports probably the largest extent of lowland (< 300 m) rain forest extant in Australia. Rainfall (c. 1700 mm p.a.) occurs mostly between November and June, with some rain typically occurring even in the driest months July–October. Methods Interpretation of change in lowland rain forest vegetation cover was undertaken for a 140 km2 area comprising complex vegetation, geology and physiography using available air photos (1943, 1970 and 1991). A GIS database was assembled comprising rain forest extent for the three time periods, geology, elevation, slope, aspect, proximity to streams and roads. Using standard GIS procedures, a sample of 6996 10 × 10 m cells (0.5% of study area) was selected randomly and attributed for vegetation structure (rain forest and non‐rain forest), and landscape features. Associations of rain forest expansion with landscape features were examined with logistic regression using the subset of cells that had changed from other vegetation types to rain forest, and remained rain forest over the assessment period, and comparing them with cells that showed no change from their original, non‐rain forest condition. Results Rain forest in the air photo study area increased from 45 km2 in 1943 to 78.1 km2 by 1970, and to 82.6 km2 by 1991. Rainfall (and atmospheric CO2 concentration) was markedly lower in the first assessment period (1943–70). Modelled rates of rain forest invasion differed predominantly with respect to substrate type, occurring faster on substrates possessing better moisture retention properties, and across all elevation classes. Greatest expansion, at least in the first assessment period, occurred on the most inherently infertile substrates. Expansion was little constrained by slope, aspect and proximity to streams and roads. On schist substrates, probability of invasion remained high (> 60%) over distances up to 1500 m from mature rain forest margins; on less favourable substrates (diorite, granites), probability of expansion was negligible at sites more than 400 m from mature margins. Main conclusions (i) Rain forest expansion was associated primarily with release from burning pressure from c. the 1920s, following major disruption of customary Aboriginal lifestyles including hunting and burning practices. (ii) Decadal‐scale expansion of rain forest at Iron Range supports extensive observations from the palaeoecological literature concerning rapid rain forest invasion under conducive environmental conditions. (iii) The generality of these substrate‐mediated observations requires further testing, especially given that landscape‐scale rain forest invasion of sclerophyll‐dominated communities is reported from other regions of north‐eastern Australia. 相似文献
