Level or jump detectors generate the reconstructed time series from a noisy record of patch-clamp current. The reconstructed time series is used to create dwell-time histograms for the kinetic analysis of the Markov model of the investigated ion channel. It is shown here that some additional lines in the software of such a detector can provide a powerful new means of patch-clamp analysis. For each current level that can be recognized by the detector, an array is declared. The new software assigns every data point of the original time series to the array that belongs to the actual state of the detector. From the data sets in these arrays distributions-per-level are generated. Simulated and experimental time series analyzed by Hinkley detectors are used to demonstrate the benefits of these distributions-per-level. First, they can serve as a test of the reliability of jump and level detectors. Second, they can reveal beta distributions as resulting from fast gating that would usually be hidden in the overall amplitude histogram. Probably the most valuable feature is that the malfunctions of the Hinkley detectors turn out to depend on the Markov model of the ion channel. Thus, the errors revealed by the distributions-per-level can be used to distinguish between different putative Markov models of the measured time series. Chorismate synthase (CS) catalyses the conversion of 5- enol pyruvylshikimate 3-phosphate (EPSP) to form chorismate, which is the last common intermediate in the synthesis of the three aromatic amino acids phenylalanine, tyrosine and tryptophan. Despite the overall redox-neutral reaction, catalysis has an absolute requirement for reduced flavin. In the fungus Neurospora crassa , a flavin reductase (FR) activity able to generate reduced flavin mononucleotide in the presence of NADPH is an intrinsic feature of a bifunctional CS. In all bacterial and plant species investigated to date, purified CSs lack an FR activity and are correspondingly 8–10 kDa smaller than the N. crassa CS (on the basis of SDS–PAGE). The cloning of N. crassa CS and subsequent characterization of the purified heterologously expressed enzyme indicates that, surprisingly, the FR probably resides within a region conserved amongst both mono- and bifunctional CSs and is not related to non-homologous sequences which contribute to the larger molecular mass of the N. crassa CS. This information directed this work towards the smaller Saccharomyces cerevisiae CS, the sequence of which was known, although the protein has not been extensively characterized biochemically. Here the characterization of the S. cerevisiae CS is reported in more detail and it is shown that the protein is also bifunctional. With this knowledge, S. cerevisiae could be used as a genetic system for studying the physiological consequences of bifunctionality. The phylogenetic relationship amongst known CSs is discussed.  相似文献

Simulating the Long-term Response of Tropical Wet Forests to Fragmentation   总被引:1,自引:0,他引:1  
In the coming decades, a large fraction of the world's tropical forests will be fragmented into remnants surrounded by secondary vegetation, land-use areas, or roads. It is important to develop integrative tools to monitor the evolution of these fragmented ecosystems. We used the individual-oriented and process-based forest growth simulator FORMIND2.0 to investigate the spatial and temporal effects on standing biomass and functional diversity of various intensities and patterns of fragmentation within a forest landscape. The simulator was calibrated for an old-growth wet forest in French Guiana, South America. We found that the standing biomass of forest remnants was reduced significantly compared to a similar area of nonfragmented forest. When fewer but larger remnants were created rather than many small ones, the total loss in biomass and the increase in the abundance of early-successional species were significantly reduced, confirming that edge effects dominate the functioning of forest remnants. We also performed simulations of secondary succession after the landscape had been abandoned. The simulated recovery time in those secondary forests depends on both the size of cleared area and the spatial pattern of the remnant forests. After mechanical spraygun inoculation barley mild mosaic virus (BaMMV) was detected in barley cv. ‘Gerbel’ (control) as well as in rye cv. ‘Somro’, but not in wheat cv. ‘Kanzler’ and oat cv. ‘Alfred’. ELISA values of infected barley and rye were similar. Furthermore, infected rye plants developed symptoms typical for barley yellow mosaic virus infection.  相似文献

Aim This study was conducted to investigate the potential of predicting alpha diversity and turnover rates of a highly diverse herbivorous insect family (Geometridae) based on vascular plant species richness and vegetation structure. Location The study was carried out on the south‐western slopes of Mount Kilimanjaro within a wide range of habitats between 1200 and 3150 m elevation. Methods The floristic and structural composition of the vegetation was recorded at 48 plots of 400 m2. Geometrid moths were sampled manually at light sources located at the plot centres. Principal components analysis, redundancy analysis and multiple linear regression were used to explore how alpha diversity and species turnover of geometrid moths are related to vegetation structure and plant species richness. Results Alpha diversity of geometrid moths was significantly correlated with species diversity patterns in the most common vascular plant families (R2 = 0.49) and with plant structural parameters (R2 = 0.22), but not with overall floristic diversity. Species turnover of geometrid moths was strongly linked to diversity changes in a range of plant families (40% explained variance), less strongly to changes in vegetation physiognomy (25%), and only weakly to overall floristic diversity (5%). Changes in elevation were a better predictor of both alpha diversity and species turnover of geometrid moths than any principal component extracted from the vegetation data. Main conclusions Vegetation composition, diversity and structure all showed significant correlations with the diversity and species composition of geometrid moth assemblages. Nevertheless, in most cases relationships were indirect, via environmental parameters such as temperature and humidity, which influenced both vegetation and moth fauna. Possible direct links between geometrid diversity and potential food plants were much weaker. The lack of a significant correlation between overall plant species richness and geometrid diversity indicates that tropical geometrid moths may not be very selective in their food plant choice. Accordingly, a clear correlation between floral diversity and herbivore species richness must be regarded as overly simplistic, and the diversity of vascular plants cannot universally be used as a suitable biodiversity indicator for diverse insect taxa at higher trophic levels.  相似文献

Abstract The anaerobic degradation of microcrystalline cellulose by thermostable cellulolytic enzyme complexes from Clostridium thermocellum JW20 (ATCC 31449) was monitored. For quantitative investigations as enzyme-coupled spectrophotometric assay has been developed. The assay allows for the evaluation of the release of cellubiose-/glucose-units from native cellulose. Kinetic studies revealed that the anaerobic breakdown of crystalline cellulose (CC) at 60°C follows Michaelis-Menten kinetics K m CC values have been determined for different aggregation states of the cellulolytic complex. The presented assay seems well suited to screen for CC-degrading enzymes of various sources, and to further explore the mechanism of CC-breakdown.  相似文献

A 1.6 kb r ep etitive DNA s equence (RPS) from Petunia hybrida was identified that destabilizes expression of a GUS marker transgene. Following polyethylene glycol (PEG)-mediated tobacco and petunia protoplast transformations, GUS expression patterns analysed on callus and plant levels were clearly more variable when constructs contained the RPS sequence. The effect on transgens expression required chromosomal integration since the two different RPS constructs employed did not exhibit reduced levels of GUS activities in transient assays. DNA methylation analysis implies a hypermethylated default state of endogenous RPS copies present in the petunia genome. Analysis of the transgens DNA in different transgenic tobacco plants showed almost complete hypermethylation of a particular Hhal site of the RPS sequence. It is proposed that, due to the presence of specific signals within the RPS region or based on interaction of RPS with other endogenous homologous sequences, RPS functions as an initiation region for de novo methylation and induces expression variegation in adjacent sequences.  相似文献

Using dynamic vegetation models to simulate plant range shifts   总被引:3,自引:0,他引:3  
Dynamic vegetation models (DVMs) follow a process‐based approach to simulate plant population demography, and have been used to address questions about disturbances, plant succession, community composition, and provisioning of ecosystem services under climate change scenarios. Despite their potential, they have seldom been used for studying species range dynamics explicitly. In this perspective paper, we make the case that DVMs should be used to this end and can improve our understanding of the factors that influence species range expansions and contractions. We review the benefits of using process‐based, dynamic models, emphasizing how DVMs can be applied specifically to questions about species range dynamics. Subsequently, we provide a critical evaluation of some of the limitations and trade‐offs associated with DVMs, and we use those to guide our discussions about future model development. This includes a discussion on which processes are lacking, specifically a mechanistic representation of dispersal, inclusion of the seedling stage, trait variability, and a dynamic representation of reproduction. We also discuss upscaling techniques that offer promising solutions for being able to run these models efficiently over large spatial extents. Our aim is to provide directions for future research efforts and to illustrate the value of the DVM approach.  相似文献
