Due to the considerable role of N-cadherin in cancer metastasis, tumor growth, and progression, inhibition of this protein has been highly regarded in recent years. Although ADH-1 has been known as an appropriate inhibitor of N-cadherin in clinical trials, its chemical nature and binding mode with N-cadherin have not been precisely specified yet. Accordingly, in this study, quantum mechanics calculations were used to investigate the chemical nature of ADH-1. These calculations clarify the molecular properties of ADH-1 and determine its reactive sites. Based on the results, the oxygen atoms are suitable for electrophilic reactivity, while the hydrogen atoms that are connected to nitrogen atoms are the favorite sites for nucleophilic reactivity. The higher electronegativity of the oxygen atoms makes them the most reactive portions in this molecule. Molecular docking and molecular dynamics (MD) simulation have also been applied to specify the binding mode of ADH-1 with N-cadherin and determine the important residues of N-cadherin involving in the interaction with ADH-1. Moreover, the verified model by MD simulation has been studied to extract the free energy value and find driving forces. These calculations and molecular electrostatic potential map of ADH-1 indicated that hydrophobic and electrostatic interactions are almost equally involved in the implantation of ADH-1 in the N-cadherin binding site. The presented results not only enable a closer examination of N-cadherin in complex with ADH-1 molecule, but also are very beneficial in designing new inhibitors for N-cadherin and can help to save time and cost in this field. 相似文献

Fish is an important concentrator of mono-methyl mercury and the main route to human contamination. We compared fish Hg bioaccumulation
(within similar weight ranges) in two Amazonian river habitats during high-water seasons. The Rio Madeira has been greatly
impacted by agriculture, alluvial gold extraction, and a hydroelectric reservoir, whereas the Rio Negro is much less affected
by these human activities. The species at the top of the food web, Hoplias malabaricus (piscivorous; 80-668 ng Hg/g) and Cichla spp. (piscivorous; 42–747 ng Hg/g) showed the highest range of Hg concentrations. Nonpiscivorous species with comparable
weight range, such as Potamorhina latior (detritivorous; 20–157 ng Hg/g) and Myleus torquatus (herbivorous; 2–182 ng Hg/g), had lower Hg concentrations. Triportheus elongatus (omnivorous; 5–350 ng Hg/g), with the lowest weight range, also showed a low range of Hg concentrations. Despite the Rio

Labeling experiments using [1-13C]acetate or [1-13C]glucose were performed with opportunistic pathogenic bacteria, with innocuous bacteria related to pathogenic species or with phytopathogenic species. The labeling pattern was determined in the isoprenic moiety of ubiquinone or menaquinone derivatives. These experiments showed that Acinetobacter, Citrobacter, Erwinia, Pseudomonas, Burkholderia, Ralstonia and Mycobacterium synthesize their isoprenoids via the mevalonate-independent glyceraldehyde 3-phosphate/pyruvate route. Variation in annual flowering effort is described for 16 long datasets from 11 species of Chionochloa (Poaceae) in New Zealand. All populations exhibited extreme mast seeding. The most variable species was C. crassiuscula (coefficient of variation, CV=3.02) over 26 years at Takahe Valley, Fiordland, which is the highest published CV we know of worldwide. The other populations also had high CVs (lowest CV=1.42, mean CV=1.84) which were higher than for other well‐studied genera such as Picea, Pinus and Quercus. There were also frequent years of zero flowering (mean across all populations was 37.2% zero years; maximum 53% for C. rubra and C. crassiuscula over 19 years) whereas zero years are rare in other published masting datasets.Flowering was highly synchronous among species within a site (mean r=0.886), and also (though significantly less so) among sites. Among sites, synchrony was not significantly higher within‐species (mean r=0.711) than between‐species (r=0.690). Warm summer temperatures led to heavy flowering the following summer. Flowering synchrony increased with increasing synchrony in local deseasonalised summer temperatures, and decreased with increasing distance between sites.Mast seeding has been shown in Chionochloa to reduce losses to specialist flower or seed predators. Among‐species synchrony may be adaptive if species share a common seed predator. Developing seeds of at least 10 Chionochloa species are attacked by larvae of an undescribed cecidomyiid. In Takahe Valley, where masting is most pronounced, cecidomyiids attacked all six Chionochloa species in all four years studied. Mean annual losses were almost constant (10.0 to 13.4%) while flowering effort varied 100‐fold. The invariant losses are consistent with other evidence that the cecidomyiid may have extended diapause, which would make it harder to satiate by mast seeding. We hypothesise that one possible factor favouring such extremely high levels of mast seeding in Chionochloa is that its seed predator is very hard to satiate. 相似文献

The impact of leaf vein cavitation and embolism on stomatal response and leaf hydraulic conductance was studied in potted plants of sunflower subjected to water limitation. Plant dehydration was achieved either by cutting well‐watered plants near their base and leaving them dehydrating in air or by depriving intact plants of irrigation. The vein cavitation threshold (ΨCAV) was estimated in terms of ultrasound acoustic emissions (UAE) from the leaf blade versus leaf water potential (ΨL). This was found to be the same (ΨCAV ≈ ?0.6 MPa) for leaves of both cut and intact plants where stomata began to close in coincidence with starting vein cavitation. Vein embolism was detected by infiltrating leaves at different ΨL with 0.7 mM fluorescein and measuring the percentage fluorescent area as percentage of total leaf surface area. A distinct loss of vein functionality (up to 50%) was found to occur in leaves at progressively decreasing ΨL, starting when leaves reached ΨCAV. A linear positive relationship with high statistical significance was found to exist between gL and percentage leaf fluorescent area, thus indicating that stomata were sensitive to vein embolism. The hydraulic conductance (KL) of the leaf was affected by leaf dehydration less than expected (KL decreased by about 20% between near full turgor and ΨL = ?1.3 MPa). When the extravascular leaf compartment was excluded either by killing cells by immersing leaves in 70% ethanol or by cutting the main leaf venous system through to allow flow to bypass it, KL turned out to increase 5.5 times, thus suggesting that the high dominance of the hydraulic resistance of the extravascular leaf compartment over the total leaf resistance might buffer or mask possibly large local changes in KL inducing stomatal closure. 相似文献

The prevalence of antibiotic-resistant bacteria in the marine environment is a growing concern, but the degree to which marine mammals, seabirds and fish harbor these organisms is not well documented. This project sought to identify the occurrence and patterns of antibiotic resistance in bacteria isolated from vertebrates of coastal waters in the northeastern United States. Four hundred and seventy-two isolates of clinical interest were tested for resistance to a suite of 16 antibiotics. Fifty-eight percent were resistant to at least one antibiotic, while 43% were resistant to multiple antibiotics. A multiple antibiotic resistance index value ≥0.2 was observed in 38% of the resistant pathogens, suggesting exposure of the animals to bacteria from significantly contaminated sites. Groups of antibiotics were identified for which bacterial resistance commonly co-occurred. Antibiotic resistance was more widespread in bacteria isolated from seabirds than marine mammals, and was more widespread in stranded or bycaught marine mammals than live marine mammals. Structuring of resistance patterns based on sample type (live/stranded/bycaught) but not animal group (mammal/bird/fish) was observed. These data indicate that antibiotic resistance is widespread in marine vertebrates, and they may be important reservoirs of antibiotic-resistant bacteria in the marine environment. 相似文献

During mouse pregnancy, uterine stromal cells transform into morphologically distinct decidual cells under the influence of the implanting embryo and a proper hormonal environment. Mechanical stimulation of hormonally primed uterine stromal cells leads to the same morphologic alterations. The decidualization of stromal cells is characterized by the accumulation of pericellular basement-membrane material. Decidualized stromal cells of pregnancy differ in this respect from stromal cells obtained from nonpregnant uteri, which are not associated with any significant amounts of basement-membrane components. Mouse decidual cells isolated from 6- to 7-day pregnant uteri explanted in vitro continue to synthesize basement-membrane-like extracellular matrix. Using immunohistochemistry and metabolic labeling followed by immunoprecipitation, SDS-PAGE, and fluorography, it was shown that the decidual cells synthesize laminin, entactin, fibronectin, type-IV collagen, and heparan sulfate proteoglycan. Stromal cells isolated from nonpregnant uteri and explanted in vitro produced the same basement-membrane components. Apparently, these cells were stimulated by the procedure used during isolation and explanation to undergo pseudodecidualization. We thus showed that stromal cells from pregnant and nonpregnant mouse uteri synthesize significant amounts of basement-membrane components in vitro, and hence could serve as a good model for the study of normal basement-membrane components. 相似文献
